Endothelin-1 promotes corneal epithelial wound healing in rabbits.

This study is an attempt to determine the effect of endothelin-1 (ET-1)-containing eyedrops on the rate of corneal epithelial wound closure in rabbits. After corneal epithelial debridement of about 50-60 mm2 with n-heptanol, eyedrops containing either 10(-8) M ET-1 or 10(-6) M ET-1 were applied at five different times for the next three days. The wound was serially photographed 12, 24, 36, 48, 60 and 72 hours later to measure the extent of healing with a computerized planimeter. The respective wound healing rate at 10(-6) and 10(-8) M ET-1 was 1.293 +/- 0.040 mm2/hour (n = 11) and 1.262 +/- 0.087 mm2/hour (n = 6), respectively. These rates were 29% (p < 0.01) and 26% (p < 0.05) above the control value, 1.005 +/- 0.063 mm2/hour (n = 13). Reepithelialization was not associated with any epithelial hyperplasia, neovascularization or conjunctival hyperemia. These results suggest that ET-1-containing eyedrops could be of therapeutic value in the remedy of corneal epithelial defects.

[Efficacy and safety of electroshock therapy without convulsions under general anesthesia in senile depression].

We examined the efficacy and safety of electroshock therapy (EST) under general anesthesia in 26 depressed patients (9 males, 17 females, age range 55-79). The scores of the Hamilton Psychiatric Rating Scale for depression were significantly decreased after EST and clinical symptoms such as depressive mood. psychomotor retardation, anxiety, agitation, suicidal tendencies, hypochondria and sleeplessness improved in all of the patients. Complications included amnesia (16/26), delirium (3/26) and transient arrhythmia (1/26) after EST, but neither lethal nor lasting complications were observed. In summary, EST was an effective and convenient method of treatment for senile depression.

Inhibition by prolactin of membrane-associated phosphatidylinositol kinase of human endometrial fibroblast.

Certain malignant tumors synthesize and secrete a putative peptide mitogen, which elicits a potent proliferative response in their supporting stromal cells. We recently demonstrated that prolactin (PRL) binds to human endometrial fibroblasts and inhibits mitogenicity of an endometrial carcinoma extract (Imai A, et al. Proc Soc Exp Biol Med 203:117-122, 1993). In this report, we have studied inhibitory regulation by PRL of phosphatidylinositol (PtdIns) kinase activity associated with plasma membranes isolated from human endometrial fibroblasts. Incubation of the isolated plasma membrane with [gamma-32P]ATP and exogenous PtdIns caused [32P]phosphate incorporation into PtdIns phosphate (PtdInsP); 95% of the 32P-labeled PtnInsP was accounted for by PtdIns 4-P. The PtdIns phosphorylation by membrane preparations was selectively stimulated in a dose-dependent manner by vanadate, in parallel with an elevated autophosphorylation of endogenous membrane proteins. Concomitant exposure of the membrane preparations to PRL led to a remarkable inhibition of the vanadate-responsive PtdIns phosphorylation and protein autophosphorylation. This inhibition was dependent on PRL dose, and half-maximal effect occurred at a concentration 1-10 nM of PRL. Degradation of the produced PtdInsP in the plasma membranes was not affected by PRL. Similar inhibition of PtdIns kinase activities were observed in membranes prepared from cells that had been pretreated in vivo with PRL prior to assay in vitro. These findings demonstrate that PtdIns kinase activity associated with protein autophosphorylation is suppressed by PRL in plasma membrane isolated from endometrial fibroblasts. The inhibition of vanadate-responsive PtdIns kinase by PRL suggests an involvement of this enzyme in the antimitogenic action of the hormone on human endometrial fibroblasts.

Insulin-like growth factor-related genes, receptors, and binding proteins in cultured human retinal pigment epithelial cells.

PURPOSE: It was determined in cultured human retinal pigment epithelial (HRPE) cells whether there is gene expression for insulin, insulin-like growth factors (IGFs), insulin and IGF receptors, and IGF-binding proteins (IGFBPs); if these peptides are secreted by the HRPE cells, and whether they have mitogenic effects on these cells; and if IGF-related peptides bind to HRPE cells. METHODS: Reverse transcriptase-polymerase chain reaction (RT-PCR), nucleotide sequencing, and Southern blot analyses were done to identify gene expression. The presence of IGFs in the tissue culture medium was detected by radioimmunoassay. For determination of mitogenic effects and receptor binding characterization, [3H]-thymidine incorporation and radioreceptor binding measurements were performed. RESULTS: The genes for IGF-I and IGF-II, IGFBP-2, insulin receptor, and type I and II IGF receptors were detected. In the tissue culture supernatant, there was immunoreactivity for IGF-I and IGF-II. Insulin, IGF-I, and IGF-II stimulated DNA synthesis with EC50s of 3, 10, and 30 to 100 nM, respectively. Scatchard analyses of [125I]-IGF-I binding and [125I]-insulin binding to the cells showed that the cells have relatively abundant insulin and IGF-I binding sites. CONCLUSIONS: HRPE cells express genes for IGF-I and IGF-II, and conditioned medium from these cells is immunoreactive to their protein products. The cells express genes for insulin receptor, type I and II IGF receptors, and IGFBP-2. IGF-I, IGF-II, and insulin are all mitogenic, possibly as a result of their interactions with either the insulin or type I IGF receptor. The cells bind insulin and IGF-I with high affinity. These results suggest that IGF-I and IGF-II production by HRPE cells may be essential for autocrine/paracrine-mediated regulation of proliferation. The presence of insulin receptor suggests that insulin has a role in the regulation of HRPE function.

Clinical significance of abnormal prothrombin (DCP) in relation to postoperative survival and prognosis in patients with hepatocellular carcinoma.

OBJECTIVES: In this study we correlate plasma des-gamma-carboxy prothrombin (DCP) levels with prognosis in patients with hepatocellular carcinoma (HCC) in combination with serum alpha-fetoprotein levels (AFP). METHODS: Levels of DCP were measured in 165 patients with HCC by an enzyme immunoassay (EIA, E-1023) with an anti-DCP monoclonal antibody. RESULTS: There was no correlation between the plasma DCP levels and the serum AFP levels. The positive rate obtained from the combination assay was 72.1%. The survival rates of the patients with elevated levels of both DCP and AFP were significantly lower than those of the groups with normal DCP and AFP levels (p < 0.05). The disease-free survival rates of patients with elevated DCP and AFP levels were also significantly lower than those of patients with normal DCP and AFP levels (p < 0.05). The recurrence rate within a postoperative period of 2 yr was higher in the patients with elevated DCP and AFP levels than in those with normal levels. CONCLUSIONS: The determination of plasma DCP levels combined with AFP levels appears to be useful for the diagnosis and prognosis of HCC, and is useful also in postoperative monitoring for recurrence.

[Expression and prognostic indicators of type 1 and type 2 Lewis blood group antigens in colorectal cancers].

The immunohistochemical analysis using the monoclonal antibodies to the blood group antigens was performed on the tissues of 387 colorectal cancers and 307 normal colonic tissues, and the relation between expression of these antigens and prognosis of the patients with stage 2 was also investigated. La, Le(b), and CA 19-9 were used for type 1 antigens, and Le(x), Le(y), and Sialyl Le(x) for type 2, respectively. Expression of Ca 19-9 and Silyl Le(x) remarkably increased in cancers throughout the colorectum. Le(b) and Le(y) were expressed in only 0-17% of the normal distal colon, but in almost colonic tumors. Therefore CA 19-9 and Sialyl Le(x) are tumor-associated antigens in the whole colorectum, and Leb and Le(y) in the distal colon. Expression of CA 19-9 in primary tumor provided more prognostic information and risk of liver metastasis than other Lewis blood group antigens.

Quantitative analysis of c-erbB-2 protein in breast cancer tissue by enzyme immunoassay.

The introduction of c-erbB-2 protein to clinical medicine as a prognostic indicator and tumor marker is desired. The authors determined the concentration of c-erbB-2 protein in 81 breast cancer tissues by enzyme immunoassay. The concentration of c-erbB-2 protein in breast cancer tissues showed a broad spectrum. A significant correlation was observed between tissue c-erbB-2 protein concentration and estrogen receptor status or immunohistochemical determination. In patients with distant metastases, there was a close association between c-erbB-2 protein concentration in the primary tumor and serum c-erbB-2 protein level. Cases with a high tumor concentration of c-erbB-2 protein (> 1000 U/mg total protein; 18.5% of cases) had a poor prognosis. From the above, we concluded the measurement of tissue c-erbB-2 protein by enzyme immunoassay to be clinically useful in breast cancer.

Interleukin-6 as a marker of hepatic metabolism following hepatectomy.

This study evaluated the clinical significance of interleukin-6 levels after hepatectomy. Thirty-one patients who underwent hepatectomies were examined. Perioperative data involving serum laboratory tests, serum concentrations of interleukin-6, and liver energy status as indicated by arterial ketone body ratio (AKBR) were analyzed. In each patient, the maximum concentration of interleukin-6 in the postoperative course (IL-6-M) was observed by the 2nd postoperative day. IL-6-M correlated significantly with the maximum concentration of total bilirubin and with the postoperative decrease in AKBR. IL-6-M also correlated with the duration of surgery and blood loss, but no relationship with aminotransferase was found. We conclude that after hepatectomy IL-6-M may be a marker for the metabolic status of the liver as well as operative stress, and is useful because it can be evaluated in the early postoperative period.
