Cloning and nucleotide sequence analysis of immunodominant heat-shock protein of Yersinia enterocolitica.
Yersinia enterocolitica, a highly antigenic 60-kDa protein, designated cross-reacting protein antigen (CRPA), is a member of the chaperonin-60 family of molecular chaperones. The gene encoding CRPA was cloned, expressed and sequenced. A partial library from Y. enterocolitica 0:3 genomic DNA was constructed in vector pUC19 and was screened by the immunoreactivity to monoclonal antibody 1A4, which has specificity for a species-specific epitope on the CRPA molecule. The crpA gene region consists of a putative two-cistron operon encoding proteins of 549 and 97 amino acids. The operon structure was led by a consensus heat-shock promoter sequence. Homology searches using the derived amino acid sequence have revealed that CRPA is 88.2% identical to GroEL of Escherichia coli. CRPB, another protein encoded by the operon, shows extensive sequence homology, 91.8% identical to GroES of E. coli which is a member of chaperonin-10.
Perceptions of schizophrenic patients and their therapists: application of the semantic differential technique to evaluate the treatment relationship.
This study is an attempt to evaluate the treatment relationship with schizophrenic patients by examining the patients' and their therapists' perceptions of themselves and each other, which are hypothesized to reflect features of the relationship. One hundred fifty-eight schizophrenic patients and 11 psychiatrists who maintained a supportive relationship with the patients as a therapist estimated their perceptions using the semantic differential (SD) technique with 17 adjective pairs. Eight composite scales with sufficient internal consistency were constructed from the estimations. The interrelationship among the perceptual elements, which was represented by correlation analysis of the composite scale scores, seemed consistent with our clinical experience. A factor-analytic study of the scales yielded 3 orthogonal factors that could be assumed to characterize the treatment relationship. The patient-therapist cooperation factor indicated the degree of trust between the two participants, supposedly the affective or relational aspect of the therapeutic alliance. The therapist passivity factor reflects the therapist's passive role-taking and the clinical stability of the patient. The patient strength factor was related to the condition-related and characterological strength of the patient. It is demonstrated that the estimations performed by patients and therapists are valid and useful for evaluation of the treatment relationship in the current status.

Double-blind controlled study on the effects of dietary diacylglycerol on postprandial serum and chylomicron triacylglycerol responses in healthy humans.
OBJECTIVE: The effects of dietary diacylglycerol (DG) on postprandial lipemia in healthy humans were investigated. METHODS: Forty normolipidemic male volunteers ingested fat emulsions containing either DG oil or triacylglycerol (TG) oil, at different doses: 10 g (n = 13), 20 g (n = 10) and 44 g (n = 17). Two test emulsions were given at seven-days intervals in random order. Fatty acid compositions of the test oils had been adjusted to be equal. Fasting and postprandial serum lipid concentrations in each group and plasma lipoprotein lipids in the 20 g-fat ingestion group were measured during the postprandial intervals. RESULTS: When DG emulsion was ingested, serum TG concentrations were significantly lower (p < 0.05) in the late postprandial phase, i.e., 4 hours, 6 hours as compared to the TG emulsion. The magnitude of postprandial lipemia (the area bounded by the curve above the fasting concentration) after ingestion of 44 g-DG emulsion was significantly less than that of 44 g-TG emulsion (6.54 +/- 5.12 and 8.45 +/- 7.54 mmol x h/L, mean +/- SD, respectively). Chylomicron TG, cholesterol, and phospholipid concentrations at 4 hours after ingestion of DG emulsion were significantly lower (p < 0.05) than those after the ingestion of TG emulsion at the same time point. No marked differences were observed for VLDL, LDL and HDL lipids between the test emulsions. CONCLUSION: In the usual range of fat intake (10-44 g), postprandial response after ingestion of DG emulsion was significantly less than that after ingestion of TG emulsion in healthy human subjects.
Pulmonary adenocarcinoma with micropapillary component: an immunohistochemical study. Case report.
Micropapillary carcinoma has been described in various organs, including the breast, urinary bladder, ovary and lung. We here present a case of pulmonary micropapillary carcinoma in a 72-year-old Japanese man who died of respiratory failure and septic shock, following which autopsy was performed. A mass measuring 2.5 x 2.5 x 2.5 cm was observed in the left lower lobe of the lung. The tumor showed moderately differentiated papillary adenocarcinoma with a focal micropapillary component. Carcinomatous lymphangiosis was also observed in the left lung and metastatic lesions were observed in the bilateral lung, liver, vertebra, muscle layer of the urinary bladder, right adrenal gland, spleen and lymph nodes. The micropapillary component was predominant at some metastatic sites. Immunohistochemically, both the adenocarcinoma and micropapillary components were positive for cytokeratin (CK) 7, CK19, TTF (thyroid transcription factor)-1, carcinoembryonic antigen (CEA) and surfactant apoprotein A (SP-A), and negative for CK20, estrogen receptor, progesterone receptor, uroplakin III, and CA125. The invasive area of the conventional adenocarcinoma component contained a large number of myofibroblasts, whereas the stroma of the micropapillary component contained a small number of myofibroblasts. However, no myofibroblasts were observed in the stroma of the central core of the non-invasive micropapillary carcinoma. Several lymphatic invasions by neoplastic cells were identified in the peripheral area of the micropapillary component using D2-40 antibody. The immunohistochemical profile may be helpful in determining the primary location of the neoplasm containing micropapillary features. Myofibroblasts are present in the stroma of the invasive neoplastic nests in the micropapillary component as well as the conventional adenocarcinoma component, and D2-40 monoclonal antibody may be useful for evaluating the lymphatic invasion of pulmonary micropapillary carcinoma.
Lymphoblastoid cell lines with integrated human herpesvirus type 6.
OBJECTIVE: Attempts were made to establish stable in vitro cell lines latently infected with human herpesvirus type 6 (HHV-6). STUDY DESIGN/METHODS: We previously studied a patient with B-cell acute lymphoblastic leukemia infected with HHV-6. The peripheral blood mononuclear cells (PBMCs) from this patient were immortalized by infection with Epstein-Barr virus (EBV) and herpesvirus saimiri (HVS). RESULTS: Infection of the PBMCs with EBV and HVS gave rise to B- and T-lymphoblastoid cell lines, respectively. Both cell lines were positive for HHV-6 DNA, as confirmed by polymerase chain reaction (PCR) and Southern blot hybridization. Fluorescence in situ hybridization (FISH) demonstrated integration of HHV-6 in these cell lines. Only one integrated site of viral DNA was detected in metaphase chromosome spreads, and it was preferentially located at the long arm of chromosome 1 (1q44). HHV-6 appeared latent in the infected cells, since neither the HHV-6 immediate-early gene transcript nor virion-associated protein was detected. CONCLUSIONS: The HHV-6-positive lymphoblastoid cell lines would be useful for study of the mechanism of HHV-6 integration.
Growth inhibitory effect of N-(phosphonacetyl)-L-aspartic acid on human myeloid leukemia-derived cell lines and modulation by dipyridamole.
Dipyridamole (DIP) inhibited the uptake of tritiated thymidine (TdR), deoxyuridine (UdR) and uridine (UR) in myeloid leukemia-derived cell lines, PL-21 and KCL-22. DIP also inhibited the growth of PL-21 (50% inhibitory concentrations, IC50, 15 microM) and KCL-22 (IC50, 3.0 microM). PALA inhibited the growth of PL-21 (IC50, 68 microM) and KCL-22 (IC50, 62 microM). The growth inhibition by PALA was significantly enhanced by the addition of 1 microM (PL-21) and 0.1 microM (KCL-22) of DIP. The inhibitory effect of PALA or PALA with DIP was completely abolished by the addition of 50 microM of UR in both cell cultures. Clinical evaluation of PALA in combination with DIP is warranted for the treatment of myeloid leukemia.
