[Antibody to human herpesvirus type-8 in the general populations of Xinjiang Autonomous Region(A.R.)].

OBJECTIVE: Kaposi 's sarcoma (KS) does not rarely occur in Xinjiang A. R. Much of the evidence show that human herpes virus-8 (HHV-8) is associated with KS, and the different incidence of classic KS among different countries reflects the different prevalence in general populations. The aim of this study was to know whether the prevalence of HHV-8 is higher in general populations of Xinjiang than the other places of the country. METHODS: We used BCBL-1 cell line as antigen and collected 1071 serum sample of different nationalities (Uighur, Khalkhas, Kazak and Han nationality) from south and north regions of Xinjiang A. R. for detection of HHV-8 IgG antibody by IFA. RESULTS: The results showed that HHV-8 IgG antibody positive rate in Khalkhas was 48% (96/200), Uighur 30.4%(62/204), Kazak 12.5% (25/200) and Han 16.9% (79/467). While sera were diluted in 1:20, the positive rate in Khalkhas still was as high as 30.5% (61/200). In addition, the positive rate of HHV-8 IgG antibody in minorities in the same native area all were higher than Han except Kazak nationality. CONCLUSIONS: These data illustrated that the prevalence of HHV-8 IgG antibody in general populations of different nationality was different, Khalkhas and Uighur were higher than Kazak and Han. Most of the KS cases were reported from Uighur nationality, but no case was reported from Khalkhas thought a highest HHV-8 IgG antibody detection rate (48%) there was demonstrated. The reason for it would be further studied.

[Precise determine the position of the opposite upper lid after correcting the congenital unilateral ptosis and their relationship].

OBJECTIVE: The good results should be obtained after correction of the congenital unilateral ptosis for bilateral eyes. METHODS: A formula was made and determined the position of the opposite upper lid according to Hering's law. RESULTS: Thirty-two patients with congenital unilateral ptosis were operated. The correct rate is 94.12% according to the formula and the control is 66.67%. CONCLUSION: The position of the opposite upper lid should be determined after the ptosis eye corrected before operation in order to obtain a good results.

Hepatocellular adenoma and focal nodular hyperplasia: a series of 24 patients with clinicopathological and radiological correlation.

OBJECTIVE: To investigate two rare benign lesions, hepatocellular adenoma (HCA) and focal nodular hyperplasia (FNH), and evaluate differential diagnosis. METHODS: Twenty-four consecutive patients with presumed HCA and FNH were studied at the Liver Cancer Institute from January 1996 to May 1999. Preoperative assessment included clinical evaluation, symptoms and laboratory tests. New imaging techniques were prospectively appraised in addition to usual techniques. All had hepatic resections and follow-up. Histologic examination of surgical specimens was obtained in all cases. RESULTS: In every instance, FNH was an incidental finding. FNH consists of nodular aggregates of cytologically normal hepatocytes with foci of intranocular bile duct proliferation. In this series, patients with HCA had larger tumors and more often were symptomatic but the occurrence was unrelated to oral contraceptive steroids (OCS) usage. Intralesional hemorrhage or necrosis is common, and was seen in 75% of cases. The best imaging procedure in the diagnosis of FNH was MRI. Color Doppler US was a useful adjunct, but CT lacked specificity, making histological diagnosis mandatory. All patients underwent tumor resected were tumor--free during the follow-up. CONCLUSIONS: FNH is a distinct histopathologic entity, and is distinguishable from HCA. FNH is a hyperplastic response by the liver parenchyma to a pre-existing arterial malformation. HCA is a liver neoplasia and has the potential of malignant transformation to HCC. Based on these findings, we believe that if the clinical suspicion of HCA or FNH is strong, resection is usually the best approach if technically feasible and histologic diagnosis is mandatory.

Molecular epidemiologic study of Mycoplasma genitalium infection in high risk populations of sexually transmitted diseases in China.

OBJECTIVE: To detect Mycoplasma genitalium (Mg) infection in high risk populations of sexually transmitted diseases (STDs) from several cities in China and to clarify the possible role of Mg in the pathogenesis of non-gonococcal urethritis. METHODS: Polymerase chain reaction (PCR) based on 2 pairs of primers, one for Mg-Pa (adhesion protein) and the other for 16S-rRNA (Mycoplasma genera) was used to detect Mg infection. Urogenital specimens of different high risk populations and urethritis patients, were collected from the Guangdong, Kunming, Shanghai, Nanjing, and Changzhou areas. RESULTS: The positive detection rate of Mg-DNA in high risk populations of STDs was significantly higher than that of the control group (chi 2 = 7.82, P < 0.01). The positive detection rates in Guangdong STD clinics and promiscuous persons from Kunming were higher than those from the Shanghai, Nanjing, and Changzhou areas (chi 2 = 8.54, P < 0.01 and chi 2 = 5.89, P < 0.05). Mg DNA could be detected in those patients without other relevant pathogens. Some patients were simultaneously infected with Mg and other microbes, such as Chlamydia trachomatis and/or Ureaplasma urelyticum. The positive Mg-DNA detection rate in patients with urethritis symptoms was higher than in patients without the symptoms (chi 2 = 11.68, P < 0.01). CONCLUSION: Mg infection exists in high risk populations of STDs in China, and the Mg infection rate is different among different high risk groups.

[The study of T helper cell subsets and relationship to relative cytokines and ventilatory function in asthmatic patients].

OBJECTIVE: To investigate the changes of T helper cell(TH) subsets and relationship to the relative cytokines, serum total IgE, eosinophil count and ventilatory function in asthmatic patients. METHODS: 20 asthmatic patients and 10 healthy subjects were selected. The counts of the TH subsets, the level of IL-4 and IFN-gamma in the supernatant of peripheral blood mononuclear cell(PBMC), serum total IgE concentration and eosinophil count in peripheral blood were determined in acute attack and stable patients with asthma and healthy subjects. RESULTS: The TH2/TH1 ratio in the acute attack and the stable patients with asthma were increased significantly, compared with that in normal group (P < 0.01). The relation analysis showed that the value of TH1 had significantly positive relation with that of IFN-gamma, the value of TH2 had significantly positive relation with that of IL-4. The total serum IgE had significantly positive relation with the level of IL-4. FEV1 had significantly positive relation with that of TH1, negative relation with that of TH2, and TH2/TH1 significantly. The total serum IgE and the number of eosinophils had significantly positive relation with that of TH2. CONCLUSION: The balance of TH in asthmatic patients deviates significantly. The deviation of balance of TH has relation with severity of asthma.

[Effect of IGF1 receptor gene antisense oligodeoxynucleotide on T24 urinary bladder cancer cells].

OBJECTIVE: To evaluate the effects of autocrine blockage on T24 urinary bladder cancer cells. METHODS: Semiquantitative RT-PCR, MTT determination, 3H thymidine incorporation, flow cytometry and electron microscopy were used to study the effects of antisense oligodexynucleotide (ODN) targeted against insulin-like growth factor 1 receptor (IGF1R) gene on IGF1R gene expression, drug sensitivity, proliferation and apoptosis of T24 cells. RESULTS: After being treated with antisense ODN specific for IGF1R gene for 48 hours, the intrinsic IGF1R mRNA expression of T24 cells reduced approximately by 72.9%, which caused decrease in survival and increased sensitivity to mitomycin-induced apoptosis(P < 0.05). CONCLUSION: To block the autocrine loop with IGF1R antisense oligonucleotide may serve as a potential therapeutic approach to bladder cancer.

[Nitric oxide synthase and vascular endothelial growth factor expression in hepatocellular carcinoma and their relation to angiogenesis].

OBJECTIVE: To analyse inducible nitric oxide synthase (iNOS), endothelial nitric oxide synthase (eNOS), vascular endothelial growth factor (VEGF) protein expression in hepatocellular carcinoma (HCC) and their relation to angiogenesis. METHODS: Tissue sections from 71 HCC patients were examined immunohistochemically for protein expression of iNOS, eNOS, and VEGF. Microvessal density (MVD) was counted by endothelial cells immunostained by anti-CD34 antibody. RESULTS: Positive iNOS and eNOS immunostaining was detected in 83.1% and 85.9% of HCC respectively. iNOS and eNOS were not detected in normal hepatic tissue. MVD was 34.3 +/- 1.5/HP and 38.6 +/- 1.6/HP in HCC positive for iNOS and VEGF while it was 31.2 +/- 2.8/HP, and 22.4 +/- 2.0/HP in HCC negative for iNOS and VEGF respectively (P < 0.01). A correlation between NOS expression and VEGF in HCC was not observed. CONCLUSION: iNOS and eNOS may play a role in malignant transformation of post-hepatic cirrhosis. The expression of iNOS and VEGF favors angiogenesis of HCC.

[Clinical studies on the use of hyperbaric oxygen in replantation of young permanent teeth].

OBJECTIVE: To investigate the treatment effects of hyperbaric oxygen(HBO) in replantation of young permanent teeth. METHODS: A total of 138 luxation injuries to young permanent teeth undergoing treatment. The teeth were replanted and then divided into 2 groups. The HBO group was exposed to 0.25 MPa pure oxygen 40 x 2 minutes each day for 10 days continuously while the control group were compared. RESULTS: The total effective rates of the HBO group and the control group were 97.26% and 70.77% respectively, and showed significant difference (P < 0.05); respective frequencies of missing tooth after 1 year were 1.37% and 18.46%, and provided a statistical significance (P < 0.001). CONCLUSION: HBO used in replantation of luxated young permanent teeth could promote the recovery of periodontal membrane, pulp, and prevent resorption of root.

[Protective effect of nitric oxide on pancreas and its relation to sulfhydryl compounds and oxygen free radicals].

OBJECTIVE: To investigate the effect of endogenous nitric oxide (NO) on acute necrosis pancreatitis in rats and its relation to sulfhydryl compounds and lipid peroxidation. METHODS: Acute necrosis pancreatitis in rats was induced by retrograde sodium taurocholate (5%) infusion into the pancreatobiliary duct (1 ml/kg body weight), and N(G)-nitro-L-arginine (L-NNA) was used as the inhibitor of endogenous NO. The effect of endogenous NO on pancreatic injury, serum amylase level, the pancreatic tissue levels of sulfhydryl compounds, and malonaldehyde (MDA, the end product of lipid peroxidation) was evaluated, respectively. RESULTS: Sodium taurocholate administration induced evident pancreatic tissue edema and acinar necrosis, and intrapancreatic hemorrhage occurred in 2/7 rats. Both serum amylase and tissue MDA [(1.25 +/- 0.28) nmol/mg x pr vs. (0.5 +/- 0.03) nmol/mg x pr, P < 0.05] were significantly increased, but tissue sulfhydryl compounds were decreased markedly. Pretreatment with the NO inhibitor, L-NNA (12.5 mg/kg body weight), significantly intensified acinar necrosis and increased the intrapancreatic hemorrhage (10/12). L-NNA also resulted in a further increase of serum amylase and tissue MDA [(3.0 +/- 0.40) vs. (1.25 +/- 0.28) nmol/mg x pr, P < 0.05], but it had no effect on the tissue sulfhydryl compounds. CONCLUSION: Endogenous NO has the effect of pancreatic protection, and its antioxidation may be responsible, at least in part, for the protective mechanisms. Sulfhydryl compounds may not be involved in NO's pancreatic protection mechanisms.

[An experimental study on F-heparin surface modified IOLs implanted into the Rhesus monkeys' eyes].

OBJECTIVE: To evaluate the biocompatibility of F-heparin surface modified IOLs. METHODS: 13 monkeys were divided into 3 groups, and implanted with different surface modified and non-modified IOLs into their eyes. All of the eyes were examined by slitlamp microscope and Schiötz tonometer at postoperative 15, 30, 60, 90 and 180 days. Postoperatively, at different periods the aqueous was aspirated from the anterior chamber to calculate cells. RESULTS: F-heparin surface modified IOLs induced milder inflammatory reaction and less dense posterior capsula opacification (PCO) than non-modified IOLs. CONCLUSION: F-heparin surface modified IOL has a better biocompatibility.

[Antisense oligonucleotides resistance to coxsackievirus B3 infection in HeLa cells].

OBJECTIVE: Antisense oligodeoxynucleotides (ODNs) hold great promise as therapeutic agents for the treatment of many human diseases. Oligonucleotides specifically hybrid with a selected sequence of RNA or DNA and inhibit the flow of genetic information DNA (or RNA) to proteins. In accordance with the present study, several oligonucleotide sequences that targeted CVB(3) RNA coding Vp(4), Vp(2), Vp(1), cleavage enzyme (P2A) or untranslational region were designed for observation. METHODS: Antiviral activities of these oligonucleotides were evaluated by plaque reduction assay and cytopathic effect (CPE). Cells were also treated with random oligonucleotides as a specificity control. RESULTS: Original ODNs, displaying sequence-specific inhibition ranging from 50% to 90% were identified. Three ODNs (at 5 microM concentration), which targeting at the pyrimidine - rich tract (S(CB1)), structural protein VP(1) (S(CB4)) and initiation of translation (S(CB2)) were shown to exhibit inhibition of viral infection 75% to 90% at 1 MOI. There was only low antiviral activity in S(CB5) and S(CB3), and random oligonucleotide were not found having antiviral activity. Oligonucleotides in this study were not toxic at 5 microM concentration. CONCLUSION: A better antiviral antisense oligonucleotides as S(CB1) and S(CB4), S(CB2), could provide candidate antisense oligonucleotides for the treatment of CVB(3) infection.

[The effect of nordihydroguaiaretic acid on endothelial cell migration induced by glioma cells].

OBJECTIVE: To investigate the effect of nordihydroguaiaretic acid (NDGA) on the migration of endothelial cells (ECs) induced by glioma cells (GCs) in vitro. METHODS: Cells of human umbilical vein endothelial cell line ECV-304 and human malignant glioma cell line SHG-44 were co-cultured in the Falcon Cell Culture Insert system, and the effect of NDGA on the migration of ECs induced by GCs was investigated. The expression of vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) was examined with immunohistochemistry. RESULTS: The expressions of VEGF and bFGF in SHG-44 cells were reduced after treatment with 100 micromol/L NDGA for 1 to 3 days. 100 micromol/L NDGA significantly inhibited not only the chemotactic migration of ECs induced by either glioma cells or glioma-conditioned media, but also the random motility of ECs (P < 0.01). NDGA at a concentration between 50 micromol/L and 200 micromol/L inhibited the chemotactic migration of ECs in a dose-dependent manner (the inhibition ratios were 12.0%, 37.4%, 86.7% and 82.2%). CONCLUSION: The results confirm the inhibitory effects of NDGA on the migration and angiogenic factor expression of ECs, which suggests that NDGA may suppress angiogenesis of glioma.

[Study on method of sampling and analysis of acephate in the air].

Acephate in the air was collected with polyurethane foamed plastics, desorbed with methanl, separated with a column 2% DEGS and determined by GC-FPD. The detectable limit was 0.052 mg/m3. When the concentration of standard solution was 3-15 micrograms/ml, the relative standard deviation(RSD) was 4.5%-2.3%. The desorption efficiency was above 85%. The sampling efficiency was 100%. The samples were stable for 24 h. It was proved that the method was accurate, fast and simple. It was suitable for the determination of acephate in the air.

[Numerical taxonomy of medicinal plants from Areae in Zhejiang Province].

Numerical taxonomic studies were done on 13 species from 3 genera of tribe Areae in Zhejiang, used as 13 operational taxonomic units (OTUS). 40 morphological characters were used for analysis. Euclid distance coefficients used to show quantitative index of similarity among OTUS were computed by standardized data. The dendrograms from 8 systematic cluster methods including single linkage method, complete linkage method, median method (beta = 0 WPGMA) and (beta = -0.25 WPGMA), centroid method, group average method, variable group average method (beta = -0.25) and variable method (beta = 0.25) were constructed respectively. WPGMA was slected as the optimal one by computing the comparative coefficients of every cluster result and the boundary of taxa in its dendrogram determined by a method to treated IBM computer with the program by BASIC language. The Results showed that 13 OTUs were classified as 3 clusters by broken line L2, namely, cluster I: Typhonium; cluster II: Arisaema; cluster III: Pinellia, and cluster II further as 4 groups by L1, which belong to Section Pistillata, Arisaema, Sinarisaema and Tortuosa respectively. Being consistent with those from classical taxonomy, the results in this study may be helpful to the classification and identification of the medicinal plants from tribe Areae in Zhejiang Privince.

[Uptake of drug-containing nanoparticles in dog catotid and femoral arteries].

OBJECTIVE: To study the uptake of drug-containing nanoparticles in artery for local therapy of restenosis. METHODS: Polylactic polyglycolic acid copolymer (PLGA) nanoparticles containing an antiproliferative agent 2-amino-chromone were formulated with an oil-in-water sonication emulsion/solvent evaporation technique. Arterial uptake of nanoparticles was assessed both in vitro and in vivo models of dog and rat carotid and femoral arteries. Epoxide, cyanoacrylate, fibrinogen, fibronectin, didodecyldimethylammonium bromide (DMAB), L-alpha-phosphatidylethanolaine, and lipofectin were selected to modify the surface of nanoparticles to enhance arterial uptake of nanoparticles. RESULTS: The nanoparticle size ranged from 100 nm to 200 nm, the drug loading in nanoparticles was about 15%, the nanoparticle morphology was observed by scanning electron microscopy and showed spherical shape with smooth surface. Arterial uptake of nanoparticles was enhanced greatly by surface modified nanoparticle with positively charged active agent DMAB. Once nanoparticles located in artery, a high drug level could be maintained at local site for 2 days in vivo model. CONCLUSION: The primary results of animal experiments suggested that nanoparticle could be used as drug carrier of local drug delivery for treating cardiovascular diseases.

[The research development of single nucleotide polymorphism].

Single nucleotide polymorphism(SNP) is a novel set of polymorphism based on the single nucleotide variations at the genomic level, which distributes widely across the human genome and has high polymorphism information content (PIC). Now, tens of thousands SNPs have been discovered among the human genome and are introduced by many biological medicine networks. With the development of identification and analysis technique for SNP, especially the combination with DNA chips, they, as the third generation of genetic markers, are gradually fitable for fine-scale mapping of diseases, especially the complex diseases susceptibility genes and will eventually substitute for the most commonly used microsatellite markers presently.

The bi-directional regulation of filamin on the ATPase activity of smooth muscle myosin.

OBJECTIVE: The aim of this study is to investigate the functional relationship between filamin, a known actin binding protein, and myosin and the effects of filamin on the interaction between myosin and actin. METHODS: Ultra-centrifugation method was used to investigate the binding of filamin to both phosphorylated and unphosphorylated myosins. Mg-ATPase activities of both phosphorylated and unphosphorylated myosins in the presence and absence of actin were measured to observe the effects resulted from filamin-actin and filamin-myosin interactions. RESULTS: It was found that filamin is also a myosin binding protein. Filamin inhibited the actin activated Mg-ATPase activity of phosphorylated myosin and stimulated Mg-ATPase of phosphorylated myosin in the absence of actin; in addition, filamin stimulated Mg-ATPase activity of unphosphorylated myosin in both the presence or absence of actin. CONCLUSION: The result suggest that the effects of filamin on the myosin Mg-ATPase activities are bi-directional, i.e., stimulatory via binding to myosin and inhibitory via binding to actin.

[Indirect determination of iodide by flame atomic absorption spectrometry].

A new method was developed for the indirect determination of iodide by flame atomic absorption spectrometry (FAAS). Precipitation of CuI, obtained with the reaction of Cu+ with I, was dissolved in Na2S2O3. Copper content in the solution was determined by FAAS and the iodide content was calculated using the content of copper. The linear range of the proposed method is 0-800 micrograms. mL-1 of I-, and the relative standard derivation at 0.4 mL of I level is 0.7%. The proposed method was applied to determination of iodine in iodized tablets salt, cydiodine tablets and laver with a recovery range of 99.9%-100.3%.
