From October, 1963 to December, 1992, 1,213 cases with cervical cancer were treated surgically in our hospital, among whom 922 cases, including 305 in stage 0, 123 in stage IA, 212 in stage IB, 265 in stage IIA, 17 in stage IIB, were operated before February, 1990. Among the 617 evaluable cases, excluding those in stage ), the five-year survival rates were 95.1% in stage IA, 91.0% in stage IB, 83.1% in stage IIA and 59.0% in stage IIB, respectively. The results showed that cervical tumour greater than 4 cm in diameter, invasion in muscular layers, lower degree of differentiation and pelvic lymph nodes metastasis would lead to worse therapeutic effects. The method of pelvic lymphadenectomy, pathological types, and ages of the patients, however, did not at all correlate with the survival rates of the patients. For those who have above-mentioned risk factors active adjuvant treatments are indicated.
Four compounds were isolated from Coleus carnosifolius. They were identified as betulic acid, betulin, oleanic acid and beta-sitosterol by spectral analysis and comparison with authentic samples.

OBJECTIVE: To investigate the role of tamoxifen in the treatment of pre- and post-menopausal patients with endometrial carcinoma, its mechanism and effects. METHODS: Forty patients of endometrial cancer were divided into pre- and post-menopausal groups. Each group was treated with oral tamoxifen 20mg three times per day for 10 days. The changes of serum levels of sex hormones, levels of estrogen and progesterone receptor (ER, PR) in carcinoma tissue as well as histopathologic changes following tamoxifen treatments were observed and compared. RESULTS: Histological changes showed that the cell nucleus to cell plasma ratio decreased, the nuclear mitoses reduced and the nuclear shape varied from stick to circular, indicating a tendency of cancer cells towards better differentiation. For pre-menopausal patients, follicle-stimulating hormone (FSH) increased, the concentration of ER either rose or went down while PR increased. For post-menopausal patients, FSH decreased, the concentrations of ER and PR were mainly on the increase. CONCLUSION: Tamoxifen was effective in treating patients with endometrial cancer. For pre-menopausal patients, it acted as an antioestrogen. For post-menopausal patients, it acted as a weak oestrogen.
The growth thermograms of strain 6, 2b, 3b, 5b, 1a, x, and y of Shigella flexneri with the inhibitory action of a synthetic medicine were determined by using the 2277 thermal activity monitor. Using a microorganism growth model with inhibitory conditions, the specific growth rate (mu) at different concentrations (c) of a synthetic medicine were calculated and the mu approximately c equations were established. From these mu approximately c equations, the concentration of the synthetic medicine with which the specific growth rate was minimum could be calculated. Therefore, we can analyze the bacteriostatic action of the synthetic medicine on these bacteria.
The protein encoded by the multidrug resistance-associated protein (MRP) gene was examined after infection of SF21 insect cells with recombinant baculovirus containing a full-length MRP cDNA. The time course of appearance of the protein as determined by western blot analysis revealed that maximum levels occurred 2 days postinfection. The amount of MRP made in this system was somewhat variable, but levels that were about 4-fold greater than that found in HL60/ADR cells could be achieved. The protein appeared to be full-length but was present in a highly deglycosylated form. The P170 (MRP) was phosphorylated and located exclusively in membranes of infected cells. P170 (MRP) synthesized in this system was capable of carrying out the ATP-dependent transport of leukotriene C4 into isolated membrane vesicles. The results thus indicate that MRP synthesized in insect cells is functional and has properties similar to the authentic protein found overexpressed in certain multidrug-resistant isolates.
OBJECTIVE: To investigate the effect of carboplatin based combination chemotherapeutic regimen given intraperitoneally and intravenously on ovarian epithelial cancer. MATERIALS AND METHODS: Carboplatin based combination chemotherapy was given intraperitoneally and intravenously to 48 patients with epithelial ovarian cancer. Of them, 10 cases with ascites had received no treatment before, and 38 patients had undergone cytoreductive surgery prior to the study. RESULTS: The results of this study showed that carboplatin based combination chemotherapy could not only reduce the tumor mass but also decrease the amount of ascites (P < 0.05) in patients with ascites. The response rate was 80% (clinical complete response rate was 10%) in untreated patients with ascites. The 3-year survival rate of patients with ovarian epithelial cancer was 92.3% in stage I, and 30% in stage III patients after the cytoreductive surgery. Tumor recurrence occurred during the course of chemotherapy in 15% of stage I patients and 53.3% of stage III patients. CONCLUSION: Carboplatin based combination chemotherapy given intraperitoneally and intravenously has potent effect on untreated ovarian cancer with ascites. But carboplatin based chemotherapy could not overcome the problem of drug resistance. To improve the prognosis of ovarian cancer, much more researches on the mechanisms of drug resistance need to be carried out.
Sixty-nine patients (121 sides) received endoscopic sinus surgery between May, 1993 and October, 1994. All of the patients exhibited following clinical signs: bilateral or unilateral nasal polyps and paranasal sinusitis. There were 43 men and 26 women, ranging in age from 14 to 68 years. Among the patients, 14 (25 sides) were treated with functional ethmoidectomy; 33 (59 sides) with total ethmoidectomy; and 21 (36 sides) with sphenoethmoidectomy. 23 patients (36 sides) were treated simultaneously with radical maxillary sinus surgery. According to the data of postoperative follow-up, nasal obstruction disappeared completely in 57 cases (57/59); headache remitted in 51 (51/56); and 35 (35/54) were free of purulent nasal discharge. No severe complications were observed. The incidence of residual and recurrent polyp were 12.39% (15/121 sides). The curative effect of endoscopic sinus surgery was analysed. The indications of operation, the preoperative CT scan of paranasal sinuses, and the endoscopic techniques were discussed.
OBJECTIVE: It is known that the development of systemic lupus erythematosus (SLE) is associated with the highly activated B cells. Interleukin-1 (IL-1) can obviously promote the proliferation and secretive activity of B cells. Moreover, interleukin-1 receptor antagonist (IL-1ra) can block the effect of IL-1 by specifically combining with IL-1 receptor, we, therefore studied the immunoregulative action of IL-1ra in lupus-like NzB/w F1 mice to detect any possible way to prevent lupus nephritis. METHODS: 12 femal NZB/W F1 mice of 13 weeks (Weight: 30-35 g) were divided randomly into 2 groups. Each mouse in the treated group was intraperitoneally injected with IL-1ra once per 2 weeks for 3 times by the dosage of 100 micrograms (0.1 ml) per time, while in the control group injected with 0.1 ml normal saline. All the mice were killed at the age of 9 months and the immunologic function was detected. RESULTS: This dosage could not prevent absolutely the development of lupus nephritis, but the renal damage was alleviated and the urine protein decreased. Moreover, it could improve the immunofunction by significantly reducing the levels of serum IL-1 alpha and obviously increasing the activities of NK cells and IL-2 induced by ConA in mononuclear cells of spleen. There was no significant difference on the levels of serum IL-6 and TNF-alpha between the treated group and control group. CONCLUSION: IL-1ra has certain regulatory effect on the immunologic function of lupus-like NZB/W F1 mice.
Monoclonal antibodies (McAb) 19A211, M344 against transitional cell carcinoma (TCC) was used to assay the tumor associated antigens on paraffin-embedded tissue sections of 51 different grades bladder cancer patients with immunohistochemical stain method. The positive rates of 19A211, M344 antigens expression were 82.4% and 88.2% in 17 grade I patients 77.7% and 83.3% in 18 grade II patients, 37.5% and 37.5% in 16 grade III patients. 19A211 and M344 antigens expression in statistical analysis was as follows: 19A211, grade I/grade III, P < 0.01, grade II/grade II, P < 0.05, M344, grade I/grade III, P < 0.01, grade II/grade II, P < 0.05, showed significant differences. Combined use of McAb 19A211 and M344 assay bladder TCC tumor associated antigens, showed that the positive rates of antigen expression in grade I was 100% (17/17), in grade II was 88.8% (16/18), in grade III was 68.8% (11/16). The results indicate that McAb 19A211, M344 were fit to diagnose grade I, II bladder TCC. Using both of two McAb can increase the positive rate in finding superficial bladder TTC.
To explore the origin of olfactory sulcus polyp (OSP) and observe the recovery of olfaction after functional endoscopic sinus surgery, fifty-nine olfactory sulcus polyps in 31 patients were reviewed. The origin of OSP was carefully observed. In these patients 69.5% (41 sides) originated from the mucosa of posterior ethmoid sinus, including the superior meatus (23 sides) and ethmoid cells near the ostia (18 sides). OSP originated from middle turbinate (8 sides) and superior turbinate (4 sides). Six OSP originated from the mucosa of both ethmoid and sphenoid sinus. Olfactory changes were dynamically observed after endoscopic sinus surgery in 26 patients. The smell improved significantly in 26 sides (52.0%) within three months. The olfaction improved slowly in those patients with recurrent polyps and polysinusitis associated with polyps. We suggest that minimal endoscopic sinus surgery is the treatment of choice for OSP.
From the roots of Ligusticum brachylobum, which is used as a substitute for traditional Chinese medicine Radix Peucedani in the southwest of China, 5 compounds were isolated and identified as umbelliferone, selinidin, anomalin, nodakenin and ferulic acid.
From the EtOH extract of Aralia fargesii, 8 compounds were isolated and identified as umbelliferone, esculetin, scopoletin, scoparone, oleanic acid, falcarindiol, mellissic acid and beta-sitosterol.
Previously, this laboratory has demonstrated that epidermal growth factor (EGF) increases adenylyl cyclase activity in cardiac membranes and elevates cAMP accumulation in hearts and cardiac myocytes. Since EGF does not increase cAMP accumulation in all tissues, we investigated the possibility that the expression of a specific isoform of adenylyl cyclase (AC) was necessary to observe EGF-elicited stimulation of cAMP accumulation. HEK 293 cells were transfected with different isoforms of AC, and the ability of EGF to increase AC activity as well as elevate cAMP accumulation was determined. In cells transfected with AC I, II, V, and VI cDNAs, neither the expression nor the amount of the two isoforms of Gs alpha (45 and 52 kDa) were altered. Similarly, EGF-elicited phosphorylation of cellular proteins on tyrosine residues in various transfectants was unaltered. However, EGF increased AC activity and elevated cAMP accumulation only in cells expressing the rat and canine ACV. EGF did not alter either AC activity or cAMP accumulation in cells overexpressing types I, II, and VI isozymes. As assessed by the ability of an anti-Gs alpha antibody to obliterate the effect, stimulation of AC activity in AC V transfectants involved the participation of Gs alpha, a finding consistent with previous data concerning EGF effects on cardiac AC (Nair, B. G., Parikh, B., Milligan, G., and Patel, T. B. (1990) J. Biol. Chem. 265, 21317-21322). Thus we conclude that the expression of AC V isoform confers specificity to the ability of EGF to stimulate AC activity.
MAP kinase (mitogen activated protein kinase) represents a ubiquitously expressed family of kinases whose long term activation via phosphorylation is essential for the mitogenic response in fibroblasts. Two family members, p42 and p44 MAP kinase are cytosolic proteins in quiescent cells, but become nuclear following mitogenic stimulation. Inactivation of MAP kinases occurs via a specific phosphatase, MKP-1. Hence, we examined the localisation of this phosphatase, to determine the cellular site of MAP kinase inactivation. Transient transfection of CCL39 fibroblasts with epitope-tagged MKP-1 showed the protein to be entirely nuclear in both quiescent and mitogen stimulated cells, whereas a catalytically inactive mutant in which the essential cysteine was mutated to serine (MKP-1CS) was predominately cytoplasmic and again serum stimulation failed to alter the protein's localisation. Expression of either wild type or inactive MKP-1 did not alter the cytosolic localisation of p44 MAP kinase in quiescent cells nor the ability of MAP kinase to translocate to the nucleus following mitogen stimulation. Expression of wild type MKP-1 inhibited serum stimulated early (c-fos promoter) and late (dhfr promoter) transcriptional events as well as entry into S-phase. This inhibition was reversed by the co-expression of an active MAP kinase. We conclude that in the continual expression of MKP-1, the cellular localisation of MAP kinase is unaffected and that inactivation of MAP kinase by MKP-1 is a nuclear process leading to the inhibition of cell division.
Epidermal growth factor (EGF) stimulates adenylyl cyclase in the heart via activation of the stimulatory GTP-binding protein Gs. Therefore, employing peptides corresponding to regions in the cytosolic domain of the EGF receptor, we have investigated the ability of sequences within the EGF receptor to activate Gs. A 13-aa peptide (EGFR-13) corresponding to the juxtamembrane region in the cytosolic domain of the EGF receptor stimulated GTP binding and GTPase activity of Gs. This peptide did not stimulate GTP binding to Gi but increased the GTPase activity of this protein. Additionally, phosphorylation of the protein kinase C site (threonine residue) within EGFR-13 decreased the ability of the peptide to stimulate Gs and increase GTPase activity of Gi. Further, in functional assays of Gs employing S49 cyc- cell membranes, EGFR-13 increased the ability of Gs to stimulate adenylyl cyclase; phospho-EGFR-13 and a 14-aa peptide corresponding to a sequence in the cytosolic domain of the EGF receptor did not alter the functional activity of Gs. Hence, the juxtamembrane region of the EGF receptor can activate Gs and, by stimulating GTPase activity of Gi, inactivates this latter G protein. Phosphorylation of the threonine residue within this region attenuates the activity of the peptide as a modulator of G-protein function.
