Estrogen receptor and alpha-fetoprotein in human breast cancer: brief communication.

Concentrations of estrogen receptor (ER) and alpha-fetoprotein were determined by dextran-coated charcoal assay and analyzed with Scatchard plots and radioimmunoassay, respectively, in cytosols of 72 human breast cancers. The values for ER ranged from 0 to 340 fmoles/mg cytosol protein. The concentrations of alpha-fetoprotein, which were low in all tumor cytosols examined, ranged from less than 0.1 to 1.1 ng/mg cytosol protein. No positive relationship was found between ER and alpha-fetoprotein concentrations. These results show that ER, but not alpha-fetoprotein, usually accounts for most of the high estrogen-binding capacity in cytosols of human breast cancers.

Analytical aspects of symptoms of nasopharyngeal malignancies.

The various symptoms of 766 patients with nasopharyngeal malignancies (641 carcinomas and 125 malignant lymphomas, confirmed histologically) were analysed statistically from the viewpoint of early diagnosis. These can be divided into aural, nasal, pharyngeal, ophthalmic, cervical lymph node and cranial nerve symptoms. The frequencies of these symptoms were compared in patients with tumours of different histopathological classifications: the nasopharyngeal malignancies were divided into carcinoma and malignant lymphoma, and the carcinomas were subclassified according to their degree of differentiation. On the whole, lymphoma patients more often had nasal and pharyngeal symptoms, and carcinoma patients ophthalmic and cranial nerve symptoms. It is considered that this difference is due to the different ways in which the two kinds of tumour develop. The frequencies of the symptoms naturally increased with the clinical course of the disease; the frequencies of the symptom both as an initial sign and as observed during the clinical course of the disease were examined. Nasopharyngeal malignancies are not indicated by localized symptoms but become evident when adjoining organs, such as the ear or nose, give some sign of abnormality. In not a few cases, a neck swelling is the initial sign; and in some patients, cranial nerve involvement is the first symptom. Since a patient may not understand the significance of his symptom and may consult a general surgeon, physician, ophthalmologist and/or paediatrician, these specialists should be alerted to the recognition of this disease.

Histopathological types of nasopharyngeal carcinoma in a low-risk area: Japan.

NPC in Japan was studied histopathologically by examining 816 NPCs from among 3 338 biopsies from patients with tumours of the nasopharynx and other parts of the upper respiratory system. In addition, a comparative study on NPC among Taiwanese and an immunoserological study were carried out. When comparing histological types of malignant tumours of the upper respiratory tract, poorly-differentiated squamous-cell carcinoma predominated only in those of the nasopharynx. Malignant nasopharyngeal tumours in Japanese patients were characterized by two histological features: predominance of squamous-cell carcinoma, especially of the poorly-differentiated type, and a relatively high frequenty of malignant lymphoma. An analysis of 731 cases of NPC showed that a vast majority (86.7%) were poorly-differentiated and a minority (13.3%) well-differentiated squamous-cell carcinomas. The former included 45.8% spindle-polygonal-cell carcinomas, 26.6% transitional-cell carcinomas ana 14.3% lymphoepitheliomas. Although the transitional-cell carcinomas and lymphoepitheliomas showed a peculiar morphology, it was confirmed that they are of a squamous nature. A comparative study of the histology of NPCs in a high-risk area, Taiwan, and in a low-risk area, Japan, revealed considerable differences between the two groups. Well-differentiated carcinomas were infrequent in both groups but were more frequent in Japanese than in Taiwanese, while the frequency of poorly-differentiated carcinomas, especially transitional-cell carcinomas and lymphoepitheliomas, was much higher among Taiwanese. A seroepidemiological study on the relation of anti-VCA antibody titres to histological type of tumour in 84 Japanese NPC patients revealed that the rate of positivity and the geometric mean of the titres were considerably higher in patients with lymphoepitheliomas. Althougropathological study along this line would appear to be important, since the results obtained in Japanese NPC cases may suggest that EBV genomes in NPC cells vary with the grade of differentiation of NPC.

Isolation and characterization of membrane proteins responsible for attachment of polyribosomes to rough microsomal fraction of rat liver.

A protein fraction which has a high affinity for polyribosomes was isolated from rough microsomal membranes of rat liver. The mode of polyribosome binding to this fraction (R-fraction) was studied by using CsCl equilibrium centrifugation and compared with that for stripped rough microsomal membranes. The following were found. (1) The polyribosome-binding cpacity of the R-fraction was heat-labile and sensitive to trypsin, and was suppressed by increasing KCl concentration and addition of 0.1 mM-aurintricarboxylic acid. (2) Of the four subfractions obtained by gel filtration of the R-fraction on a Sephadex G-200, only the R1-fraction, eluted at the void volume, showed a high affinity for polyribosomes. The polyribosome-binding capacity of the R1-fraction decreased with time on storage at 4 degrees C. (3) The R1-fraction contained three major proteins with mol. wts. 108,000, 99,000 and 65,000.

Circumscribed carcinoma of the uterine cervix, with marked lymphocytic infiltration.

The present study was carried out to investigate a distinctive type of carcinoma of the uterine cervix categorized under the designation as circumscribed carcinoma with lymphocytic infiltration. Grossly this carcinoma is characterized by defined circumscription with a superficial ulceration. A microscropic feature characteristic of this tumor is the presence of a loose fibrillary stroma infiltrated densely and uniformly by lymphocytes throughout the tumor mass. The tumor is arranged in solid cords separated by a lymphoid stroma with evidence of minimal squamous differentiation. The authors picked up 709 patients with carcinoma of the uterine cervix who underwent radical hysterectomy and pelvic lymphadenectomy at the Cancer Institute Hospital, Tokyo, between 1956-1967, without preoperative radio- or chemotherapy. Among them, 39 or 5.5% were identified as this type of carcinoma. Patients with this types of carcinoma had a significantly better prognosis than those with other types of cervical carcinoma of the same stage (p less than 0.05). This favorable prognosis is probably due to the less regional node metastases found in the group. A proposal was made to segregate this particular type of carcinoma from other types of cervical carcinoma on the basis of its morphologic and prognostic distinctiveness.

Fine body movement during autogenic training. Longitudinal and short-term changes.

A sensitive electronic device (static sensograph) was used to record fine body movement in two similar groups of normal high school girls at intervals over a few months. One group received weekly instructions in the technique of autogenic training (AT) and was urged to practice it daily, whereas the other (control group) received no such instrutions. In the AT group mean body movement (eyes closed, sitting position) decreased with every session. At 3 and 4.5 months after training was begun (session 3 and 4, respectively) mean body movement of this group was significantly lower than in the first session, before training was begun. In the fourth (but not earlier) sessions the AT group also showed a significant decrease in mean body movement during AT compared with the periods immediately preceding and following it. In contrast, the control gorup showed no significant differences in means between any sessions or periods within a session. The results suggest that fine body movement as measured by a static sensograph is a useful parameter for studying AT and probably other relaxation procedures.

Estrogen receptor and endocrine responsiveness in Japanese versus American breast cancer patients.

We have compared the incidence of estrogen receptor (ER) in breast tumors and its clinical correlation with responses to endocrine therapies in Japanese and American patients. There was no correlation between tumor histopathology and the presence of ER, and the ER values in primary and metastatic lesions from the same patients were similar in most Papanese cases. Japanese patients with low and high plasma estradiol levels had identical incidences of ER-positive tumors. The correlation between tumor ER and response to endocrine therapy is similar between Japanese and American patients. The incidence of ER-positive tumors is higher in postmenopausal American patients in both primary and metastatic lesions. It is possible that the reported increase in tumor lymphocyte infiltration in Japanese patients may explain this difference. The reported 5-year survival advantage of Japanese breast cancer patients cannot be explained by differences between the two populations in the response to endocrine therapy for advanced disease.
