Overlap of IGF- and heparin-binding sites in rat IGF-binding protein-5.

Using site-directed mutagenesis, we have undertaken a study of a potential IGF-binding site in the C-terminal domain of rat IGFBP-5, lying close to or within a previously described heparin-binding domain (residues 201-218) in this protein. After analysis of binding activity using three different methods - ligand blotting, solution phase equilibrium binding and biosensor measurement of real-time on- and off-rates - we report that the mutation of two highly conserved residues within this region (glycine 203 and glutamine 209) reduces the affinity of the binding protein for both IGF-I and IGF-II, while having no effect on heparin binding. In addition, we confirm that mutation of basic residues within the heparin-binding domain (R201L, K202E, K206Q and R214A) results in a protein that has attenuated heparin binding but shows only a small reduction in affinity for IGF-I and -II. Previous findings have described the reduction in affinity of IGFBP-5 for IGFs that occurs after complexation of the binding protein with heparin or other components of the extracellular matrix (ECM) and have postulated that such an interaction may result in conformational changes in protein structure, affecting subsequent IGF interaction. Our data suggesting potential overlap of heparin- and IGF-binding domains argue for a more direct effect of ECM modulation of the affinity of IGFBP-5 for ligand by partial occlusion of the IGF-binding site after interaction with ECM.

The mutation of insulin receptor substrate-1 gene in Chinese patients with non-insulin-dependent diabetes mellitus.

OBJECTIVE: To identify the relationship between mutation in the insulin receptor substrate-1 (IRS-1) gene and the incidence of non-insulin-dependent diabetes mellitus (NIDDM) in the Chinese population. METHODS: Samples were obtained from 68 Chinese patients with NIDDM and 68 control subjects. The +1700-(+)4437 bp fragment of the IRS-1 gene was screened by polymerase chain reaction (PCR)-single strand conformation polymorphism (SSCP) analysis. All SSCP variations were submitted to DNA sequence analysis. RESULTS: Two amino acid variations [GGG-->AGG (G971 R) and CCT-->TCT (P1079 S)] and 3 silent mutations [GAT-->GAC(D422D), CCA-->CCC(P737 P) and GCA-->GCG (A804 A)] were identified, among which the CCA-->CCC(P737 P) and CCT-->TCT(P1079S) have not been previously reported. All five variations were found in Chinese patients with NIDDM, while GCA-->GCG(A804A) was the only one found in control subjects. The overall incidence of the five variations in Chinese patients with NIDDM were much higher than that in control subjects (38.2% vs 7.4%, chi 2 = 18.42, P < 0.01). The most common polymorphism in the Chinese population was GCA-->GCG (A804A), and its frequency was significantly higher in Chinese patients with NIDDM than in controls (26.5% vs 7.4%, chi 2 = 8.84, P < 0.01). The homozygotes of the variation in patients with NIDDM and control subjects were 8.8% and 1.5%, respectively (chi 2 = 2.41, P > 0.05). CONCLUSION: These results indicate that there may be a relation between these nucleotide variations of IRS-1 gene and Chinese patients with NIDDM.

[Apoptosis and uterine cervical carcinogenesis].

OBJECTIVE: To investigate the possible role of apoptosis in the development of uterine cervical carcinoma. METHODS: Formalin-fixed, paraffin-embedded samples from 190 patients [41 patients with severe dysplasia (SD); 37 with carcinoma in situ(CIS); 31 with microinvasive carcinoma (MIC), 40 with fran invasive large cell non-keratinizing epidermoid carcinoma (IC)], and 41 samples from normal cervical squamous epithelium (NE) were studied. The number of apoptotic cells was assessed in situ by the TDT-mediated dUTP-biotin nick end labeling (TUNEL) method. PCNA, p53 and bcl-2 were demonstrated immunohistochemically. RESULTS: (1) In NE, TUNEL-positive cells were found in the superficial layer and PCNA-positive cells were confined in the lamina profunda, while in cervical neoplasia these cells were irregulasly scattered throughout the cervical lesions. (2) The TUNEL staining index decreased while PCNA increased with progression of the neoplasm, showing a significant negative correlation between apoptosis and proliferation. (3) In patients with SD and CIS who had overexpression of p53 and bcl-2 proteins, the cells positively stained by TUNEL were significantly less in number than in these with negative p53 and bcl-2 expression. No such observation for PCNA expression. CONCLUSION: These results suggest that apoptosis is associated with the early process of cervical carcinogenesis and apoptosis is closely correlated with overexpression of p53 and bcl-2 proteins.

[New full length cDNA cloned from normal pituitary and pituitary tumors].

OBJECTIVE: To clone new full length cDNA from normal pituitary and pituitary tumors. METHODS: cDNA libraries of human normal pituitary and pituitary tumors were constructed, and large-scale sequencing and bioinformatics were used. Silico cloning and rapid amplification of cDNA ends (RACE) were also used. RESULTS: 67 new full length cDNAs were cloned: 41 were cloned from normal pituitary, and 26 from pituitary tumors. Of the 67 cDNAs, 16 were related with endocrine function; at least 2 with a classical signal peptide were secretory protein, and 3 with classical transmembrane domains were transmembrane protein. Among the 5 cloned new full length cDNAs involved in signal transduction pathway of hormone, at least 2 participated in signal transduction pathway of receptors with tyrosine kinase activition. CONCLUSION: During short time, 67 new full length cDNA were cloned from normal pituitary and pituitary tumors by large-scale producing expressed sequence tags. Some of them may play an important role in the regulation of physiological and pathological processes of pituitary.

[The study of nitric oxide synthase in ovary tumor].

OBJECTIVE: To study the activity of nitric oxide synthase (NOS) in ovarian tumors. METHODS: Spectrophotometry was used to examine NOS activity and NADPH-diaphorase histochemistry to identify its localization. RESULTS: The activity of NOS was significantly higher in malignant than in normal ovarian tissue and benign ovarian tumors (P < 0.01). There was negative correlation between NOS activity and tumor cell differentiation. NOS reaction product was mainly located in cancer cells, and was negative in the interstitial tissue. Furthermore, NOS was mostly located in membrane and cytoplasm of the cancer cell. CONCLUSION: Increase in nitric oxide synthase activity might be related to the growth and malignant behavior of ovarian cancer.

[Investigation of urinary interleukin-6 level in chronic renal failure patients and the influence of Rheum palmatum in treating it].

OBJECTIVE: To assess the clinical significance of urinary interleukin-6 (IL-6) level in chronic renal failure (CRF) patients and the effect of Rheum palmatum (RP) in treating it. METHODS: RP and captopril were given to the study group, while captopril was given to the control group alone, and level of urinary IL-6 was determined by sandwich-ELISA. RESULTS: Urinary IL-6 level in CRF patients was obviously higher, than that in healthy control (P < 0.01). After treatment, the urinary IL-6 and serum creatinine reduced significantly in the study group (P < 0.05), and there was significant difference compared with that in the control group (P < 0.05). CONCLUSIONS: Determination of urinary IL-6 level is useful in studying the severity of immune inflammation of CRF. RP improves renal function by inhibiting the production of IL-6 and lowering immune inflammation.

[The construction of a recombined E. coli strain with stable and high production of poly(3-hydroxybutyrate-co-3-hydroxyvalerate)].

Plasmid pJMC2 was constructed by cloning the parDE fragment of RK2 into pTZ18U-PHB which harbored phaCAB from Alcaligenes eutrophus and was transferred into E. coli HMS174 and E. coli JM107 separately. It is very stable in its hosts cultured in medium without ampicillin. E. coli HMS 174(pTZ18U-PHB) and E. coli JM107(pTZ18U-PHB) produced P(3HB-co-3HB) in a low phosphate concentration medium(18 mmol/L). The proportion of 3-hydroxyvalerate(3HV) in the polymer was 5%-8%. A fed-batch culture of E. coli HMS174(pJMC2) was conduct in a 5 L automatically controlled fermentor, the final dry cell weight, P(3HB-co-3HV) content, and th 3HV proportion were 42.5 g/L, 70% and 4.9% respectively.

[Cardiac allograft arteriosclerosis and myocardial necrosis].

OBJECTIVE: To study the development of myocardial necrosis, fibrosis, and arteriosclerosis in cardiac allograft in order to provide a theoretical basis for the prevention and treatment of anti-rejection. METHODS: The recipient was pretreated with inoculation of donor splenocytes (SPC) followed by cyclophosphamide (CP). Heterotopic cervical heart transplantation was performed. The allografts stained with Hematoxylin and eosin (HE), Masson and Van Gieson were studied about inflammatory cell infiltration, myocardial necrosis, fibrosis and arteriosclerosis. RESULTS: Preconditioning with SPC followed by CP prolonged the survival time of cardiac allograft. Inflammatory cell infiltration, myocardial necrosis, fibrosis, and arteriosclerosis in cardiac allograft were significantly decreased. CONCLUSIONS: The development of acute cardiac allograft rejection originated from the epicardial area to the intimal area of the heart in rat. The infiltration of collagenous fiber in the coronary arteries is one of the major causes of concentric stenosis. The pretreatment of SPC followed by CP can prevent and relieve the rejection.

The role of hysterectomy in the therapy of gestational trophoblastic tumor.

OBJECTIVE: To evaluate the role of hysterectomy for patients with gestational trophoblastic tumor. METHODS: We retrospectively analyzed 68 cases of gestational trophoblastic neoplasia treated by hysterectomy from 1985 to approximately 1997 at PUMC hospital. Thirty-eight cases were diagnosed of choriocarcinoma and 30 were invasive mole. RESULTS: Twenty-three elder patients who didn't desire to preserve fertility were selected for hysterectomy after shorter courses of chemotherapy, 22 of them had a complete remission (95.6%), the total average courses of chemotherapy was 4.2. Of twenty-seven chemorefractory cases who were suspected of a refractory isolated lesion in the uterus, delayed hysterectomy as an adjunct to chemotherapy was performed, 20 of them got a complete remission (71.1%), the total average courses of chemotherapy were 9.4. Emergency hysterectomy is indicated in 18 patients with uterine perforation or life-threatening hemorrhage, 17 cases had a complete remission (94.4%), the total average courses of chemotherapy were 7.6. CONCLUSION: Although the development of effective chemotherapy has resulted in improved survival of patients with gestational trophoblastic tumor, hysterectomy remains an important adjuncts in the treatment of a selected subset of patients; in order to operate more completely and prevent recurrence, it's better to perform extended hysterectomy for the indicated patients.

[Expression of epidermal growth factor receptor in gestational trophoblastic tumours].

OBJECTIVE: To investigate the expression of epidermal growth factor receptor (EGFR) in gestational trophoblastic tumours and its clinical significance. METHODS: Placentas of 10 normal early pregnancies, 20 molar pregnancies, 10 samples from invasive mole and 8 samples of choriocarcinoma were used for the study. EGFR expression was evaluated by immunohistochemistry using monoclonal antibody. RESULTS: EGFR was expressed both in normal chorionic villi and tissues of trophoblastic tumours. EGFR was expressed more strongly in normal chorionic villi and non-malignant transformed hydatidiform mole than in malignant transformed molar pregnancies and malignant trophoblastic tumours (P < 0.05). Univariate and multivariate analysis indicated that EGFR immunostaining intensity was significantly related to the clinical stage in malignant trophoblastic tumours (P = 0.026, OR = 9.874). CONCLUSION: EGFR expression could be used as an index for prediction of malignant transformation of hydatidiform mole and staging of malignant trophoblastic tumours.

[Effect of high-lipid diet on glomerular mesangial matrix in adriamycin-induced nephrotic rats].

OBJECTIVE: To determined the effect of hypercholesterolemia induced by high-lipid diet on glomerulosclerosis. METHODS: Urinary protein excretion and serum cholesterol were assayed, Image analysis and techniques of pathology, immunohistochemistry, molecular biology were used to determine morphological changes in glomeruli and the production of glomerular mesangial matrixes in adriamycin-induced nephrotic syndrome (NS) and non-NS rats fed with standard chow and high-lipid chow. RESULTS: The serum total cholesterol level was significantly higher in rats with high-lipid chow in both non-NS[(2.2 +/- 0.3) g/L vs (0.9 +/- 0.1) g/L, P < 0.01] and NS[(9.5 +/- 0.2) g/L vs (2.3 +/- 0.3) g/L, P < 0.01]. The urinary protein excretion was significantly higher in the high-lipid diet rats than that in standard chow rats [(76.2 +/- 24.2) mg/24 h vs (44.8 +/- 13.6) mg/24 h, P < 0.05] in NS rats. Significant increases in the mesangial matrix and mesangial cells were observed in rats with high-lipid diet in both NS and non-NS group. Especially, more obvious pathological changes were found in NS group, such as lipid deposits and foam cell formation in mesangial areas, and progressing to focal and segmental glomerulosclerosis in some glomeruli. The immunohistochemical assay showed that the production of fibronectin was increased in the rats with high-lipid diet in non-NS group, whereas, the production of three major components (collagen IV, fibronectin, and laminin) was increased in NS group, especially in the rats with high-lipid chow. The increased expression of laminin mRNA was also detected with slot blotting in both NS and non-NS rats with high-lipid chow, and it was more obvious in the rats with NS. CONCLUSION: Our findings indicated that diet-induced hyperlipidemia can lead to over-production of mesangial matrix components, and further aggravate glomerulosclerosis in adriamycin-induced nephrosis.

[Diagnosis and management of gestational trophoblastic tumor with liver metastases].

OBJECTIVE: To evaluate management options and clinical prognosis for women presenting gestational trophoblastic tumor (GTT) with liver metastases. METHODS: We retrospectively analyzed sixteen GTT patients with liver metastases treated in the Peking Union Medical College (PUMC) hospital from 1985 to 1998. All were treated with 5-FU combined chemotherapy or EMA/CO regimen. Eight of them received hepatic artery infusion chemotherapy. Treatment outcome was measured by beta subunit human chorionic gonadotropin assay (beta-hCG) and by imaging studies which included ultrasound and computerized tomography. RESULTS: Of 16 cases, 4 achieved complete remission; biochemical remission was obtained in 2 patients who were alive with residual tumor; 10 patients died of the disease. The overall survival rate was 37.5% (6/16). CONCLUSIONS: The outcome for women presenting with liver metastases from GTT is poor. Early diagnosis and prompt initiation with vigorous multi-agent chemotherapy given through multi-routes are emphasized for improving the treatment outcome.

[Characteristic immunodeficiency syndrome of rapid fatal type of simian immunodeficiency virus infected monkeys].

OBJECTIVE: To observe the characteristic immunodeficiency syndrome of the rapid fatal type of simian immunodeficiency virus (SIV) infected monkeys. METHODS: Eighty rhesus monkeys and 4 cynomolgus monkeys were intravenously inoculated with SIVmac or SIVmac251. The virus isolation and viral titer, estimation by indirect immunofluroresence and viral antibody were determined periodically from monkeys' plasma; lymph node biopsies were performed for pathohistological examination. RESULTS: Twelve out of 84 macaque (14.2%) died of rapid progressive type after inoculation of SIVmac and SIVmac251 in the duration 3 to 4 months. Dying monkeys showed persistent high viremia and low level titre antibody. Eight of 10 pathohistological changes showed severe depletion of lymphoid tissue in spleen and lymph nodes, there were remarkable immunodeficiency with opportunity infection. The other two monkeys appeared moderate lymphoid tissue deletion and hyperplasia without opportunity infections. The survived monkeys' (72/84) lymph nodes biopsies revealed hypoplasia of lymphoid tissue. CONCLUSIONS: The characteristic immunodeficiency syndrome of rapid fatal type of simian immunodeficiency virus infected monkeys could be made with persistent high viremia, low level antibody, severe lymphoid tissue deletion in lymph nodes and spleen, as well as complicated opportunity infections.

[Study on neodymium complexes by broad wavelength range photoacoustic spectroscopy].

Neodymium complexes were studied by photoacoustic(PA) spectroscopy from UV-Vis to mid-IR. According to the PA spectra, low excited energy levels, relaxation processes and fluorescence properties were discussed. Then the influence of interaction between Nd(III) and ligands in solid complexes was investigated on the basis of band shifts and intensity change.

[Study on the photoacoustic phase of rare earth complexes].

The theory of the photoacoustic (PA) phase has been studied. The PA phase spectra of rare earth complexes with dibenzoylmethide have been explained basing on the theory. The phase shifts of Tb complexes at the ligand absorption peak have been calculated. The phase shifts and their intramolecular relaxation processes have been studied also.

Functional characterization of TEL/AML1 fusion protein in the regulation of human CR1 gene promoter.

The TEL/AML1 fusion gene occurs in childhood B-cell acute lymphoblastic leukemia (ALL) as a result of the translocation of human chromosome 12;21. Using reporter gene assays, we have functionally characterized TEL, AML1 and TEL/AML1 fusion proteins in the regulation of the human CR1 gene. Analysis of transcription activities showed that AML1 increased the CR1 promoter activity and that TEL repressed the basal activity of the promoter. Increased activities of the CR1 promoter by AML1 protein were reduced by the TEL protein in a concentration-dependent manner. When TEL/AML1 and AML1 proteins are present in cells at the same time, the TEL/AML1 protein inhibits the transactivation activities of AML1 protein on the human CR1 promoter even though TEL/AML1 retains the transactivation domain of AML1. A mutation analysis of the human CR1 promoter revealed that the binding sites for TEL and AML1 are necessary for the action of TEL and TEL/AML1, respectively. Thus, production of the TEL/AML1 protein by translocation of human chromosome 12;21 may contribute to leukemogenesis by the specific inhibition of AML1-dependent activation of myeloid promoters.

Plexins are a large family of receptors for transmembrane, secreted, and GPI-anchored semaphorins in vertebrates.

In Drosophila, plexin A is a functional receptor for semaphorin-1a. Here we show that the human plexin gene family comprises at least nine members in four subfamilies. Plexin-B1 is a receptor for the transmembrane semaphorin Sema4D (CD100), and plexin-C1 is a receptor for the GPI-anchored semaphorin Sema7A (Sema-K1). Secreted (class 3) semaphorins do not bind directly to plexins, but rather plexins associate with neuropilins, coreceptors for these semaphorins. Plexins are widely expressed: in neurons, the expression of a truncated plexin-A1 protein blocks axon repulsion by Sema3A. The cytoplasmic domain of plexins associates with a tyrosine kinase activity. Plexins may also act as ligands mediating repulsion in epithelial cells in vitro. We conclude that plexins are receptors for multiple (and perhaps all) classes of semaphorins, either alone or in combination with neuropilins, and trigger a novel signal transduction pathway controlling cell repulsion.

Voltammetric response and determination of DNA with a silver electrode.

A current from DNA was obtained using a silver electrode with low overpotentials for the first time. Experimental results revealed that the voltammetric response of DNA was attributed to the redox reactions of purine bases. It was also shown that such a method provided a convenient and practical way to determine DNA. A linear dependence of the peak currents on ssDNA concentrations was observed in the range 0.5-2.5 microg/mL. The relative standard deviation was 3.5% for six successive determinations at 0.5 microg/mL. The detection limit was 50 ng/mL. Influence of the structure and the length of the nucleic acids on their electrochemical behavior was discussed. In view of the merits of the silver electrode, this technique might provide new possibilities for further electrochemical research and determination of nucleic acids.
