[Cases of numerous diffuse plaques in the neocortex but without severe senile changes in the hippocampus].

Using modified Bielschowsky method, we studied neuropathologically 159 aged subjects autopsied during the period from 1976 to 1988, of which we found 19 cases (average age at death: 82.6 ys) with numerous diffuse plaques in the frontal and temporal neocortex and no severe senile changes in H1-H3 of Ammon's horn (dp group). Amyloid angiopathy had been excluded and one case was excluded because of considerable cerebrovascular lesions. The dp group was divided into 8 demented (average age at death: 86.0 ys) and 10 nondemented patients (average age at death: 79.7 ys). We compared the number, type, and ratio of types of senile plaques in the frontal cortex, temporal cortex, and putamen of the demented and nondemented groups, and obtained the following results: (1) Eight (14%) of the 59 nondemented and 8 (40%) of the 20 demented cases in which no severe senile changes in the neocortex and hippocampus had been detected by Bodian stain showed numerous diffuse plaques in the neocortex when the modified Bielschowsky method was used. (2) The ratio of classic and primitive plaques to diffuse plaques in the frontal cortex was the same in both groups, but the nondemented group had exclusively diffuse plaques in the temporal cortex. (3) In the putamen 2 nondemented cases (20%) and 6 demented cases (75%) had exclusively diffuse plaques. We considered that classic and primitive plaques are more closely related to dementia than are diffuse plaques in the temporal lobe in cases without severe senile changes in the hippocampus.

Rapid isolation with Sep-Pak C18 cartridges and wide-bore capillary gas chromatography of some butyrophenones.

A simple and rapid method for isolation of five butyrophenones with Sep-Pak C18 cartridges from human samples, and their wide-bore capillary gas chromatography (GC), are presented. The GC was made by both flame ionization and electron capture detections. The drugs contained in alkaline samples were directly applied to the cartridges and eluted with chloroform/isopropanol (9:1). The recoveries with use of the cartridges were excellent for most drugs in both urine and plasma samples. We can recommend the Sep-Pak C18 cartridges for isolation of butyrophenones because of simplicity and rapidity, and also wide-bore capillary GC because of high sensitivity and low decomposition of drugs during passage through the column.

Semicarbazide-sensitive amine oxidase activity in catfish tissues.

1. Semicarbazide-sensitive amine oxidase activity was found in five tissues of the siluroid catfish, Parasilurus asotus, using benzylamine as substrate. It was highest in the intestine, followed by the liver and skin. 2. The apparent Km value and optimal pH for benzylamine in the intestine were 49.8 microns and 8.8, respectively. 3. Substrate specificity of the enzyme was tested in the intestine and the highest activity was found with beta-phenylethylamine, followed by tryptamine and benzylamine.

Rapid isolation with Sep-Pak C18 cartridges and wide-bore capillary gas chromatography of some barbiturates.

A simple and rapid method for isolation of nine barbiturates with Sep-Pak C18 cartridges from human urine, plasma and whole blood, is presented; the detection of drugs was made by wide-bore capillary gas chromatography (GC) with flame ionization. The drug-containing samples, after mixing with dilute acid solution, were directly applied to the cartridges and eluted with either chloroform/methanol (9:1) or acetonitrile. Separation of the nine drugs was satisfactory with use of an intermediately polar HP-17 capillary column. Recoveries of most compounds were excellent for both chloroform/methanol and acetonitrile as elution solvents. However, backgrounds were cleaner and evaporation time was much shorter for the chloroform/methanol system. The present isolation method with use of Sep-Pak C18 cartridges and wide-bore capillary GC seem very useful in the fields of forensic chemistry, clinical toxicology and clinical pharmacology.

Effects of a weak diffuse-brain-stimulation on insomniacs with vibration syndrome.

To verify the efficacy of a weak diffuse-brain-stimulation (DBS) method that was specially devised by this research team, a weak DBS was administered to seven male inpatients, who had vibration syndrome due to a prolonged use of a vibratory tool and who were simultaneously complaining of insomnia. Sleep data obtained from the polygraphic recordings on the DBS-treated night were compared with those on the pseudo DBS-treated. As the result, this DBS method proved to be effective in improving patients' sleep initiation and/or maintenance, and also their subjective ratings on the quality of actual sleep. In conclusion, the present DBS was determined to produce a decrease in the sympathetic tone which is needed at the onset of sleep.

In vitro inhibition of human platelet monoamine oxidase by phenothiazine derivatives.

Nineteen phenothiazines were tested for in vitro inhibition of human platelet type B monoamine oxidase (MAO). The inhibition potency was highly dependent on structures of their side chains. The inhibition was most potent for drugs with (hydroxyethyl-piperazinyl)propyl chains followed in decreasing order by those with (N-methylpiperazinyl)propyl, (2-dimethylamino-2-methyl)ethyl and 3-dimethylaminopropyl chains. Kinetic analyses were carried out for promazine, promethazine, perazine and perphenazine as representatives of each group; the four drugs showed competitive inhibition, and Ki values of 124, 31.4, 19.2 and 22.6 microM, respectively.

Determination of beta-phenylethylamine in catfish (Parasilurus asotus) tissues by gas chromatography/mass spectrometry.

1. beta-Phenylethylamine (PEA) was detected and quantitated in tissues of the catfish, Parasilurus asotus, by very specific and sensitive gas chromatography/mass spectrometry. 2. The selected ion monitoring was made with a strong quasi-molecular ion of the pentafluoropropionic derivative of PEA in the positive chemical ionization mode. 3. PEA was found in all tissues tested ranging from 2.8 to 38.2 ng/g wet wt tissue. It was highest in the spinal cord, followed by the skin, brain and intestine.

Rapid isolation of benzodiazepines with Sep-Pak C18 cartridges.

A rapid method for isolation of various benzodiazepines from human samples with Sep-Pak C18 cartridges before wide-bore capillary gas chromatography is described. The drugs dissolved in alkaline samples were directly applied to the cartridges and eluted with hexane/isopropanol (9:1). The recoveries were excellent for all drugs in urine samples, but were somewhat lower for some drugs in plasma samples. The latter problem could be easily circumvented by using a deproteinization process before their application to the cartridge. We can recommend the Sep-Pak C18 cartridges for isolation of benzodiazepines because of their simplicity and rapidity.

[Heart rates in male Wistar rats reared under different conditions of the weaning period and social housing (author's transl)].

The electrocardiogram (ECG) was recorded from unrestrained rats reared under the 6 conditions: weaned on the 14th, 18th or 28th postnatal day and thereafter housed in isolation (14I, 18I and 28I) or grouped with littermates (14G, 18G and 28G). The heart rate (HR) was measured on a 30 second sample of the ECG recorded from the 5th to the 20th week of age. For all rearing conditions, HR was confirmed to follow the normal distribution in respective weeks, and changes in HR through development were investigated. (a) HRs of 14G and 18G were significantly higher at the 5th week, and were later to reach the stabilized HR than 28G and isolated rats. (b) The variability between individuals (Vbi) reduced markedly at the 7th week in 18G and 28G, this may be attributed to the reduction in the variance within litter (Vwl) due to the interactions among littermates. In isolated rats, it was a relative low value in Vbi perhaps because of a low value in the variance between litters (Vbl). Contrary to isolated rats, in 14G Vbi was high, Vbl was high as well, but Vwl was low owing to the early separation from the mother and long periods of group housing.

[Heart rates in male Wistar rats reared under different conditions of the weaning period and social housing].

A prior study reported the developmental changes in heart rate (HR) in male rats. In this paper, we carried out more laborious analyses to check and compare the differences among effects of the rearing conditions. In group-housed rats, the earlier the weaning periods, the lower the variance within litter, but the higher the variance between litters. In contrast to this, high values within litter were maintained in the earlier weaned isolates On the occasion, 18 G was close to 28 G in changes in the variance, and 18 I showed the same trends as 14 I, thus indicating the different significance of the periods of 14-18 days after birth and 18-28 days concerning the interaction of mother and offspring. The rats that interacted with both littermates and mother as 18 G and 28 G, showed the marked reduction in the variance between individuals along with age as opposed to 14 I, 18 I and 14 G in which the infantile form of higher variances were maintained after the 10 th week. A dynamic model of HR was led, which showed two distinct phases according to the changes in the variance. These phases corresponded to the development and aging stages. In addition, the implications of "intrinsic process", "immaturity" and "enriched environments" were discussed.
