Influence of Mobiletten on the effect of alcohol drinking in man. First communication: Influence on the pharmacokinetics of ethanol.

The interaction of the drug Mobiletten with the pharmacokinetics of ethanol was studied in 15 probands. In these experiments, the maximal ethanol concentrations in blood were found to be significantly decreased and delayed. The ethanol elimination remained unchanged. The uptake of Mobiletten resulted in a remarkable decrease of the urine volume. The quantitative determination of the ethanol content in the expired air by alcotest technique was not significantly affected.

The Munich Coma Scle (MCS).

The level of coma can be measured by the assessment of changes of behaviour after different intensities of stimulation. Two additive scales (Guttman scales) for susceptibility to stimulation and reactivity were developed, constituting the Munich Coma Scale (MCS). The findings of previous investigations were repeated in a new sample of 67 comatose patients. Implications of the additivity of reactivity are discussed and related to the concept of 'behavioural arousal' advanced in the recent literature.

[Studies on the stability of human ultradian rhythms (author's transl)].

It was investigated whether the REM-NREM (rapid eye movement-non-REM) sleep rhythm has a stable period during long-term observations. Sequences of 17 to 31 consecutive sleep records were analyzed for 6 test subjects and 1 patient. Period stability was confirmed for three experimental conditions: a) undisturbed night sleep, b) inversion of the sleep-waking cycle, c) absence of external timing mechanisms. The period of the ultradian REM sleep rhythm is no integral submultiple of 24 h, so that the remainder causes a daily drift in the REM sleep rhythm. It is assumed that the ultradian process is controlled endogenously. In contrast to the circadian rhythm the ultradian rhythm appears to be free-running under normal conditions. The stability of the ultradian period has been shown in long-term observations.

[A rapid technique for isolation of Escherichia coli type I in food (author's transl)].

A rapid isolation method for E. coli Type I is being presented: A 4-6 hours resuscitation period at 24 +/- 3 degrees C is immediately followed by the 44 +/- 0.1 degrees C incubation period. Using this method the result can be read off after 24 hours, in exceptional cases after 48 hours. This technique enables the demonstration of an hygienic parameter important for food microbiology within a short time. Examinations will now be started in the Federal Health Office with the aim for defining E. coli-standards for various foods. It is planed to recommend these standards to the federal health authorities for including them in appropriate food regulations.

[Measurement of attention deficit in the course of intoxications (author's transl)].

The level and course of attention was measured hourly in 9 drug intoxicated patients after a suicide attempt over periods which varied between 12 and 72 hrs. Attention was measured by the use of two additive 5 step scales for susceptibility to stimulation and reactivity, which were developed by the authors in earlier investigations and proven to be very reliable. Although, the original data set of attention measures was different among the patients, some common features could be elaborated: 1. The level of attention varies very little within 1 hr. Differences greater than one step on the scales were rarely observed between two measurements. 2. The mean course of recovery from attention deficit is linear throughout the scales while the variance is substantial at each step of the scales. For quantification of attention deficit a measure was defined which gives the relation between the actual deficit and full attention. Since the correlation between both scales is high over the whole observation period, it was concluded that the intoxication alters the level but not the structure of attention.

Phase shift in the REM sleep rhythm.

The periodic alternation between REM and NREM sleep was analyzed. Usually, sleep records of consecutive nights of a subject are regarded to be independent events. However, it may be that consecutive nights are realizations of a continuously ongoing rhythm. This was tested in the present study. The temporal patterns of REM and NREM sleep in sequences of about 30 consecutive nights for 3 subjects were analyzed. The results show that only the onset of the first REM sleep phase during any one night may be predicted from the sleep onset time, whereas a systematic phase shift between consecutive nights was observed in the later REM sleep phases. Thus, the onset of later REM sleep phases is better predicted by assuming a rhythm with stable period length which controls the appearance of REM sleep phases in successive nights. Under the experimental conditions the phase shift was between 5 and 10 min per 24 hrs for the 3 subjects. The result is accordance with Kleitman's basic rest activity cycle (BRAC) hypothesis.

On the structure-function relationship of acyl carrier protein of Escherichia coli.

The conformations of Escherichia coli acyl carrier protein (ACP) and acetylated ACP have been studied as a function of pH and salt concentration by circular dichroism measurements. The results show that the amino groups of ACP in their protonated form are important for maintaining the native conformation of the protein at physiological pH. However, externally added cations (divalent more effectively than monovalent ones) can substitute for the ammonium groups in maintaining the ordered structure pf ACP. It is suggested that both the ammonium groups of ACP and externally added cations reduce the repulsion between carboxylate groups of ACP and thereby prevent the unfolding of the protein. A reduction of the number of negatively charged carboxylate groups by either protonation or chemical modification abolished the requirement for either ammonium groups or other cations. A qualitative agreement between the effect of salt on the conformation and on the biological activity of acetylated ACP has been observed. The single arginine residue of acetylated ACP has been modified by treatment with a trimer of 2,3-butanedione with the resulting derivative of ACP retaining most of its biological activity.

Excretion of urinary testosterone in Klinefelter's syndrome.

The urinary testosterone excretion in 55 patients with Klinefelter's syndrome was determined by gas-chromatographic methods. The mean testosterone level in the group of Klinefelter patients was significantly decreased (15.7 mug/24 hrs) compared with the excretion by 30 normal men (44.1 mug/24 hrs). An age dependency of excretion levles was not obvious in chromatin-positive men when compared to healthy subjects. A correlation between testosterone excretion and configuration types seemed to be probable by reason of observations and statistical values. Average testosterone excretion increased with increasing normalization of body proportions. Gynecomastia was noticed in 56% of those examined. With increasing severity of gynecomastia the mean testosterone levels decreased. A correlation between beard and body hair, which was decreased in almost all subjects, and testosterone excreiton was not visible. With diminishing mean testosterone excretion an increase of osteoporosis seemed to be probable. A statistically significant difference was found when comparing patients who never had ejaculations with those who had regular libido and ejaculations. Klinefelter patients who had not succeeded in completing school or an occupational education showed a significantly decreased testosterone level as compared with others who had a profession.

[Scaling of disturbances of attention in neurological patients (author's transl)].

In a previous report two additive scales were developed for measuring susceptibility to stimulation (with 4 experimental stimuli) and reactivity (with 4 types of reactions). The degree of loss of attention in neurological patients as determined by the position of a patient correlates with the frequencies of his reactions. Thus the measurement of frequencies does not add any further information. There is no patterning of reactions due to the different etiology of the disorder of attention. These results support the suggested one-factor theory of attention.

[Development of an apparatus for measuring attention in patients with cerebral damage and functional disorders].

58 patients with various underlying neurological diseases, who had an impairment of attention, were examined. 12 patients without clinically evident disorders of attention were examined as a control group. The aim of the study was the development of a standardized procedure for the assessment of impaired attention. An additive, 4 step scale of the "susceptibility to stimulation" (Guttman scale) was constructed out of several, heterogeneous stimuli. In addition, an additive 4 step scale of "reactivity" was constructed out of a catalogue of defined reactions. The two scales permitted the assessment of reactivity and susceptibility to stimulation, the degree of which could be demonstrated as being proportional to one another. The transformation of susceptibility to stimulation into reactivity is thought to be performed by an internal system which is attributed to attention.

Thiolases of Escherichia coli: purification and chain length specificities.

The presence of only one thiolase (EC 2.3.1.9) in wild-type Escherichia coli induced for enzymes of beta oxidation was demonstrated. A different thiolase was shown to be present in a mutant constitutive for the enzymes of butyrate degradation. The two thiolases were purified to near homogeneity by a simple two-step procedure and were found to be associated with different proteins as shown by gel electrophoresis. The thiolase isolated from induced wild-type Escherichia coli cell was active on beta-ketoacyl-coenzyme A derivatives containing 4 to 16 carbons, but exhibited optimal activity with medium-chain substrates. In contrast, the thiolase isolated from the constitutive mutant was shown to be specific for acetoacetyl-coenzyme A.
