Membranes and the genetics of ethanol response.

A combination of fluorescence polarization (FPZ) and nuclear magnetic resonance (NMR) techniques have revealed that ethanol has diverse and domain dependent effects on membrane order. Under some conditions, in the more superficial membrane domains, ethanol actually orders rather disorders membrane structure. Using 1H-NMR we have examined in synaptic membranes from LS and SS mice the effects of ethanol-d6 on membrane order. The lines differ most significantly in terms of the ethanol effects on the choline methyl resonances. Ethanol was significantly more potent in increasing choline methyl resonance intensity in LS synaptic membranes than in SS synaptic membranes; these data are interpreted to show a significantly greater disordering of the superficial domains in the LS membranes. The maximum ethanol effect was observed between 0.3% and 0.5% for the concentration range studied (0.1 to 1.0%). The methylene resonance data in general paralleled the choline methyl resonance data but with a somewhat attenuated response. Ethanol had only small effects on the terminal methyl resonance in both lines. Overall, we conclude that the LS and SS mice differ in the ethanol-induced perturbation of membrane structure, primarily at more superficial membrane domains.

Protection against duodenal ulceration by somatostatins.

The purpose of our investigation was to study the effect of two somatostatin analogs (SMS 201-995 and 008) on duodenal ulcer disease induced by cysteamine in rats. Male Wistar rats were given cysteamine (28 mg/100 g body wt.) by rubber stomach tube three times in a single day. All animals of the test group were injected subcutaneously either with SMS 201-995 in different doses or with 008. On the 3rd day stomach, duodenum, and adrenal glands were macroscopically examined for lesions (duodenal ulceration, gastrointestinal bleeding, adrenal hemorrhage). Our results showed a dose-dependent effect of SMS 201-995 on the mortality, incidence, and intensity of cysteamine-induced duodenal lesions in rats. The incidence and the intensity of gastrointestinal bleeding and adrenal hemorrhage were also dose-dependently reduced by SMS 201-995. There was no definite effect of 008 on the mortality, incidence, or intensity of cysteamine-induced duodenal ulceration.

New technique of injection of estradiol into preoptic-anterior hypothalamic area of newborn rats: technique limiting diffusion and duration, with preliminary results on postpuberal block of ovulation.

Previous studies have shown that sc administration of androgen or estrogen into newborn female rats will disturb postpuberal ovulatory function, as evidenced by a higher incidence of vaginal estrus, few ovarian corpora lutea, and the abolition of estrogen-induced luteinizing hormone surges after ovariectomy and chronic estrogen maintenance. Studies of the uptake of labeled estrogen and intracerebral lesion studies have indicated the preoptic area and anterior hypothalamic nuclei (POA-AH) as possible sites of steroid uptake and control of certain sexual functions in the rat. Attempts to localize the site of steroid effects to the POA-AH of newborn rats by chronic implants have been hampered by lack of definition of extent of localization in space and time. The present study introduces a microinjection technique for the delivery of subpicogram quantities of estradiol to these areas of the central nervous system in newborn rats, and statistically defines the extent of steroid localization in three dimensions as well as the duration of exposure. Delivery of estradiol by this method in the POA-AH region yielded an increased incidence of postpuberal vaginal estrus; a decrease in the number of corpora lutea; and in many but not all, led to abolition of estrogen-progesterone induced LH surges after castration and estrogen replacement. Injection 0.5 mm more caudad did not result in blockade of the estrous cycle.
