Liver cell heterogeneity. The distribution of fructose-bisphosphatase in fed and fasted rats and in man.

Periportal and perivenous hepatocytes were isolated by microdissection from lyophilized liver slices (16 micrometer) from fed and fasted rats and from a human patient. NADP/NADPH cycling was used to determine fructose-1,6-bisphosphatase activity in the isolated hepatocytes (10 ng dry weight). The periportal hepatocytes contain 3 times as much fructose-1,6-bisphosphatase activity as the perivenous hepatocytes. A 24 h fast led to two-fold increase in the activity in the periportal hepatocytes and a four-fold increase in the perivenous hepatocytes. Fructose-1,6-bisphosphatase parallels closely with the key enzyme phosphoenolpyruvate carboxykinase, and therefore can be considered a suitable marker for gluconeogenic capacity.

[Stress-induced blood protein and blood lipid changes and their dependence on learning and conditioning].

Rats were subjected to two kinds of stress and consequent changes in blood protein and lipids were investigated. Furthermore, an attempt was made to ascertain whether or not increases or decreases in these blood parameters were reproducible by conditioning. In the first experiment animals were subjected to stress in a modified running wheel (Noble Collip drum). A series of 10 conditional stimuli (light-sound combined) was delivered together with the stress-producing stimulus. After the first exposure to stress a relative alpha1-globulin increase was observed. After 10 stress exposures the hitherto neutral stimulus alone produced a conditioned increase in the alpha1-globulin fraction. The gamma-globulin values were decreased after the first exposure to stress, but increased after 10 stress exposures. This increase could also be conditioned. In the second experiment "white noise" stress was used (110 dB). The conditional stimulus was light. After short exposure to the stress-inducing stimulus, the gamma-globulins showed a slight drop and remained thus during the whole experiment. The analysis of blood lipids showed increased stress values for alpha-lipoprotiens and reduced values for beta-fractions. Cholesterol and triglycerides reacted like the beta-lipoproteins. The total blood lipid content was, however, increased. The conditioned increase in alpha-lipoproteins and total lipids was statisticallysignigicant. It is noteworthy that the conditioned alpha-lipoprotein levels shoeed significant differences between experimental and control groups. The conditioned stress increased the levels of cholesterol and triglycerides, whereas under real stress conditions decreases were found. Research on "conditioned stress" could beof improtance in regard to clinical aspects of coronary heart disease.

[Synthesis and biological activity of the decapeptide Gly-Pro-Cys-Lys-Ala-Arg-Phe-Gly-Gly-Cys as a model for the active center of the basic trypsin inhibitor from bovine organs (Kunitz inhibitor) (author's transl)].

The synthesis of the above mentioned decapeptide as a model for the active center of the inhibitor molecule is described. The Ki value of this peptide was found to be 4.7 X 10(-4)M. This Ki value differs from the Ki value of the Kunitz Inhibitor by a factor of 10(10).

[Clinical and experimental results with acupuncture-analgesia (author's transl)].

Operations under acupuncture analgesia have been performed in Vienna since March 1972. A total of 102 procedures has been performed with "classical" Chinese acupuncture analgesia. Additionally given analgesics, psychodynamic factors and neurophysiological components are of joint importance. Our success rate in these cases was 64 p.c. Since October 1972 we call "two-phase-acupuncture-analgesia". Up to June 1975 72 operations were performed under some modifications of this method. In a neurophysiological study 17 volunteers and in 2 patients undergoing stereothalamotomies depression or even disappearance of evoked potentials was be observed during acupuncture with alectrical stimulation. Thus an interaction between pain stimuli and acupuncture stimuli in the thalamus could for the first time be demonstrated in man.

Sodium- and potassium-activated ATPase. A possible target of aldosterone.

Na-K-ATPase activity was measured with an ultramicromethod in single portions of the proximal and distal convolution and of the thick ascending limb of Henle from adrenalectomized rats and after treatment with 5 mug aldosterone per 100 g body wt. The activity in all tubular structures returned to normal within 1 h after injection. This rapid activation of Na-K-ATPase induced by hormone was completely prevented by actinomycin D and cycloheximide. It appears that this aldosterone effect on Na-K-ATPase requires an intact protein synthetic process.

[Synthesis and biological activity of some cystine peptides from the active center of the basic trypsin inhibitor from bovine organs (Kunitz inhibitor) (author's transl)].

The synthesis of the following 3 model peptides from the active center of the inhibitor molecule is described: (see article). The 3 peptides are split by trypsin with velocities increasing from (I) to (III). The following Ki values were determined: (I) Ki = 2.3 x 10(-2)M; (II) Ki = 4.9 x 10(-3)M; (III) Ki = 2.1 x 10(-3)M. These Ki values differ from the Ki values of the Kunitz inhibitor by a factor 10(11) to 10(12).
