We report a case of pulmonary infarction in a 50-year-old man who was referred for an asymptomatic coin lesion in the periphery of the right lower lobe. Computed tomography of the chest demonstrated a subpleural solitary nodule with spicular radiation. Video-assisted thoracoscopic surgery was performed to obtain a histological diagnosis. Thoracoscopic examination showed that the tumor was dark purple and the central portion was gray, indicating blood retention in the lung and necrotic and fibrotic tissue in the central portion. This is the first case of pseudotumor due to pulmonary infarction that was diagnosed by video-assisted thoracoscopic resection.
We experienced 9 cases of percutaneous bladder neck stabilization (Vesica) for patients with urinary incontinence who were diagnosed with stress incontinence from December 1995 to June 1998. None of the patients were able to control continence with any drugs. Chain cystography was performed on all of the patients before and after surgery. The posterior urethro-vesical angle before surgery was over 130 degrees in all of the patients, and it improved after the operation. Moreover, the mean volume of the pad test greatly decreased from 47 g to 1 g. All of the patients were treated with epidural anesthesia. The operation time was less than 60 minutes. The subjective and objective findings were satisfactory. We conclude that percutaneous bladder neck stabilization is a very useful surgical treatment for stress incontinence.
An improved procedure for ABO blood typing by the absorption-elution test using commercially available monoclonal antibodies was established. The optimized elution temperature for anti-A monoclonal antibodies to be liberated from bloodstains was 54 degrees C and the temperature to deactivate the liberated antibodies was over 56 degrees C. The test condition was established using a monoclonal antibody manufactured by Biotest. For the anti-A monoclonal antibody the most effective elution time was 5 min and the long incubation time decreased the activity of the liberated antibodies. On the other hand, the conditions formerly used for absorption-elution tests were suitable for anti-B monoclonal antibody. A Type-A test and a Type-B test have to be performed separately for ABO blood grouping from forensic samples. The conditions established in this report were applied to the absorption-elution test to achieve an improved result for ABO blood grouping from hair samples.
Myocarditis is a complication of Schistosoma mansoni infection, although the literature does not provide much information regarding the frequency of myocarditis. In order to analyze the relationship between myocarditis and S. mansoni infection, different laboratory animals were infected with different dose of cercariae. At different weeks of post infection the hearts of infected animals were collected and processed for histopathological examination. Myocarditis was characterized by interstitial inflammatory cell infiltration with or without granuloma. ddY and ICR infected mice showed eosinophilic egg-granuloma in the heart where as neither eosinophil nor egg-granuloma were observed in the heart of infected gerbils. Higher number of eosinophils and greater size of the granuloma were found in the ddY mice than ICR mice. The number of eosinophils was significantly higher in severe myocarditis. Incidence of myocarditis was higher in ddY mice (69% with 100) than ICR mice (35%) and gerbils (23%). The results indicate that ddY mice were more susceptible to S. mansoni infection in the development of myocarditis and myocardial severity was associated with greater eosinophil infiltration. These findings suggest that eosinophils might be involved in the development of myocarditis, although the involvement of immunological reaction can not be ruled out.
Diesel exhaust particles (DEP) contain various carcinogens and mutagens, and chronic exposure to diesel exhaust (DE) induces pulmonary cancer in experimental animals. However, the oncogenes involved in pulmonary carcinogenesis have not been identified. After F344 rats were exposed to DE containing 6 mg/m3 DEP for 4 weeks, oncogenes and related genes expressed in their lungs were surveyed using a new technique, cDNA microarray, and the results were confirmed by northern blot analysis. Expression of A-raf and proliferating cell nuclear antigen (PCNA) mRNAs was induced in rat lung by exposure to DE. These results suggest that A-raf and PCNA might contribute to pulmonary carcinogenesis in rats.
Autoxidation rates of the full-length neuronal nitric oxide synthase (nNOS) were analyzed and found to be composed of three phases, 60 s(-1) (28%), 5.5 s(-1) (11%) and 0.048 s(-1) (61%). Addition of L-Arg, N(G)-hydroxy-L-Arg (NHA), and N(G)-monomethyl-L-Arg markedly decreased the rate constants for the first and second phases down to 12-20 s(-1) and 0.32-2.6 s(-1), respectively. Addition of (6R)-5,6,7,8-tetrahydro-L-biopterin (H4B) increased the amplitude of the second phase up to 29% of the total. Addition of NHA decreased the rate of the first phase by 4.4-fold in the presence of H4B, whereas addition of L-Arg and other modulators did not significantly affect the rates under the same conditions. Thus, we deduce that (1) L-Arg stabilizes the O2-bound ferrous complex for efficient O-O bond cleavage to occur; (2) H4B influences the O2-bound ferrous complex in a fashion different from L-Arg; and (3) NHA induces a characteristic distal-site structure in the presence of H4B, reflecting a difference in the mechanism of activation of O2 in the first step (monooxygenation of L-Arg) and the second step (monooxygenation of NHA).
The relationship between l-cystine transport and intracellular glutathione (GSH) levels was investigated in cultured pancreatic AR42J acinar and betaTC3 islet cells exposed to diethylmaleate, an electrophilic agent known to activate cellular antioxidant responses. Cystine transport was mediated predominantly by the Na+-independent anionic amino acid transport system x-c, with influx inhibited potently by glutamate and homocysteate but unaffected by cationic or neutral amino acids. Saturable cystine transport was 10-fold higher in AR42J (531 pmol (mg protein)-1 min-1) than in betaTC3 (49 pmol (mg protein)-1 min-1) cells, and GSH levels were higher in AR42J cells. Treatment with 2-mercaptoethanol increased GSH levels in betaTC3 cells from 7.5 to 36 nmol (mg protein)-1, whilst the GSH content in AR42J cells (64 nmol (mg protein)-1) was not altered significantly. Incubation of AR42J or betaTC3 cells with homocysteate (2.5 mM, 0-48 h), a competitive inhibitor of cystine transport via system x-c, reduced intracellular GSH levels and resulted in a time-dependent (6-24 h) induction of system x-c transport activity. Treatment of AR42J cells with diethylmaleate (100 microM, 0-48 h) resulted in a time- (5-10 h) and protein synthesis-dependent induction of cystine transport, with intracellular GSH levels initially decreasing and then increasing 2-fold above control levels after 24 h. Diethylmaleate also depressed GSH levels in betaTC3 cells, but cystine transport was not elevated significantly. In both AR42J and betaTC3 cells, inhibition of gamma-glutamyl cysteine synthetase by buthionine sulphoximine (100 microM, 24 h) reduced GSH levels but had no effect on cystine transport. The present findings establish that induction of system x-c leads to changes in GSH levels in pancreatic AR42J acinar and betaTC3 islet cells, with changes in the intracellular redox state stimulating transporter expression. Induction of activity of system x-c, together with adaptive increases in GSH synthesis in response to oxidative stress, may contribute to cellular antioxidant defences in pancreatic disease.

Matrix metalloproteinases (MMPs) are believed to be involved in morphogenesis. Association of MMPs in a model of kidney tubulogenesis was studied using Madin-Darby canine kidney (MDCK) epithelial cells in an in vitro morphogenetic system. MDCK cells form branching tubules in three-dimensional collagen gel matrix in the presence of hepatocyte growth factor (HGF). The addition of specific MMP inhibitor BB-94 and tissue inhibitor MMP (TIMP)-2 but not TIMP-1 to such collagen gel cultures reduced the formation of branching tubules induced by HGF. The induction of membrane-type 1-matrix metalloproteinase (MT1-MMP) mRNA expression was observed in MDCK cells cultured in the collagen gel. Stable expression of MT1-MMP antisense RNA interfered with the tubule formation of MDCK cells induced by HGF-collagen gel culture. These observations implicate MT1-MMP in kidney tubulogenesis and TIMP-2-specific inhibition suggests a direct role of MT1-MMP rather than a gelatinase A-mediated effect.
When the two-dimensional crystal of bacteriorhodopsin (bR), purple membrane, is incubated at high temperature (32 degreesC) with a small amount of the neutral detergent octylthioglucoside in the presence of the precipitant ammonium sulfate, a large fraction of the membrane fragments is converted into spherical vesicles with a diameter of 50 nm, which are able to assemble into optically isotropic hexagonal crystals when the precipitant concentration is increased. The vesicularization of purple membrane takes place under such a condition that the miscibility of the detergent to the aqueous phase becomes very low, and we suggest that it is initiated by insertion of the detergent molecules into the membrane. At low temperature, the transformation into the vesicular structure is inhibited and no large crystal is produced directly from membrane/detergent/precipitant mixtures. When a suspension of the spherical vesicles produced at the high temperature is cooled and concentrated below 15 degreesC, however, a birefringent hexagonal crystal is produced that diffracts X-rays beyond 2.5 A resolution. This new crystal belongs to the space group P622 with unit cell dimensions of a=b=104.7 A and c=114.1 A, and it is shown to be made up of stacked planar membranes, in each of which the bR trimers are arranged on a honeycomb lattice and the space among the proteins is filled with the detergent molecules and native lipids. These stacked membranes are suggested to be produced by successive fusion of the spherical vesicles. This implies that the crystallization is achieved without any step for complete solubilization of the protein. The present result offers a unique crystallization method that may be applicable to such membrane proteins that are liable to denature in the presence of an excess amount of detergent.
To study the molecular epidemiology of HIV-1 spread in Myanmar and the interplay with the epidemic in surrounding Southeast Asian countries, we determined the HIV-1 subtypes prevailing in Myanmar. Thirty HIV-positive blood specimens were sampled in the capital city, Yangon, and an additional 459 sera were collected nationwide in 1995. Genetic subtyping based on the env C2/V3 sequence and serologic data, using a V3 peptide enzyme immunoassay (PEIA), revealed three patterns of HIV spread in different geographic regions in Myanmar: (1) in the capital city, Yangon, HIV-1 subtype B' ("Thai-B" cluster within subtype B) predominated both in IDUs and heterosexuals; (2) in the cities near the border with Thailand, including Tachelaik and Kawthaung, where heterosexual transmission is a major pathway of HIV-1 spread, HIV-1 subtype E was predominantly distributed among the commercial sex workers and heterosexuals; (3) in central and northeast Myanmar, both HIV-1 subtypes B' and E occurred in a mixed distribution, without showing any significant segregation by risk group. In addition, the PEIA data implied the occurrence of other subtype(s) in these areas. The interperson nucleotide sequence variations in env C2/V3 regions of B' and E, prevailing in Yangon, were 6.7 +/- 2.1 and 7.1 +/- 0.7%, respectively. They were similar to those levels observed in Thailand. These findings are consistent with the view that HIV spread in Myanmar might have taken place at about the same time as that in Thailand, and that multiple entries and exchanges of HIV-1 with neighboring countries are important factors contributing to the current distribution of subtypes in Myanmar.
The Hakata antigen is a novel, thermolabile beta2-macroglycoprotein that reacts with sera from patients suffering from systemic lupus erythematosus. In this study we present the structure and the function of the Hakata antigen. We have identified cDNA clones encoding the Hakata antigen and analyzed its function. The cDNA included a possible open reading frame of 897 nucleotides, encoding 299 amino acids. The Hakata antigen consisted of a collagen-like domain in the middle section and a fibrinogen-like domain in the COOH terminus, both of which are homologous to human ficolin-1 and opsonin P35, indicating that these three molecules form a distinct family. The molecular mass of the Hakata antigen expressed in transfected cells was 35 kDa under reduced conditions, and it formed ladder bands under nonreducing conditions compatible with the previous result that the Hakata antigen exists in serum as homopolymers. Purified Hakata antigen sustained lectin activity, showing affinity with GalNAc, GlcNAc, D-fucose as mono/oligosaccharide, and lipopolysaccharides from Salmonella typhimurium and Salmonella minnesota. These results suggest that the Hakata antigen, a new member of the ficolin/opsonin P35 family, plays a role in the serum exerting lectin activity under physiological conditions.
Ferritin is an intracellular iron storage protein whose synthesis is regulated post-transcriptionally by a mechanism that involves binding of cytoplasmic iron regulatory protein (IRP) to iron-responsive element (IRE) in the 5' untranslated region of ferritin mRNA. In this study, we have shown that in mouse peritoneal macrophages, the synthesis of ferritin was enhanced and the IRE binding activity of IRP-1 was diminished when the oxygen tension was decreased. Iron is known to induce ferritin synthesis and even in the presence of a low concentration of iron, synthesis of ferritin was enhanced and the activity of IRP-1 was decreased under hypoxia. The enhanced synthesis of ferritin under hypoxia was abolished by the addition of O2(-)-generating agents but not H2O2. The decreased activity of IRP-1 under hypoxia was reversed by adding O2(-)-generating agents. These data suggest that O2- generated in the cell is involved in alterations of ferritin synthesis and the activity of IRP-1 by oxygen.
Oxidation of low-density lipoprotein (LDL) can be mediated in vitro by cultured cells, including macrophages. This cellular oxidation is dependent on the production of free thiols by the cells in the presence of transition metal ions. We now report that the production of thiols by macrophages is greatly enhanced when cells are cultured with lipopolysaccharide (LPS) or tumor necrosis factor-alpha (TNF-alpha). Oxidation of LDL by macrophages is markedly augmented by pre-treatment of the cells with LPS or TNF-alpha. The results suggest that activation by endotoxins or TNF-alpha is a necessary step for macrophages to mediate oxidation of LDL.

Urokinase plasminogen activator (uPA) has been associated with invasion and metastasis in breast cancer. The expression of uPA and 92 kDa type IV collagenase (gelatinase B/MMP-9) is regulated by growth factors, receptor-type tyrosine kinases and cytoplasmic oncoproteins. Here, we have identified transcriptional requirements for the induction of uPA and 92 kDa type IV collagenase by epidermal growth factor (EGF). EGF stimulates the motile and invasive activities specifically in the ErbB-2-overexpressing SK-BR-3 cells. Expression of extracellular matrix-degrading proteases including type I collagenase/MMP-1, 92 kDa type IV collagenase/MMP-9, uPA and uPA receptor were induced. EGF also transiently stimulated expression of the transcription factors Ets-1 and Ets-2. Reporter transfection assays revealed the activation of uPA and MMP-9 collagenase promoters by EGF and the requirement of each of the composite Ets and AP-1 transcription factor binding sites for an EGF response. Most notably, transfections with the Ets-1 and Ets-2 expression vectors potentiated uPA and MMP-9 promoter activation in response to EGF. Mutation of the threonine 75 residue of chicken Ets-2 conserved in the Pointed group of the Ets family proteins abrogated the ability of Ets-2 to collaborate with EGF. Ets-1 and Ets-2 were highly expressed in invasive breast tumor cell lines. Our results suggest that Ets-1 and Ets-2 provide the link connecting EGF stimuli with activation of uPA and 92 kDa type IV collagenase promoters and may contribute to invasion phenotypes.
The dissociation constant (Kd) for CO from neuronal nitric oxide synthase heme in the absence of the substrate and cofactor was less than 10(-3) microM. In the presence of L-Arg, it dramatically increased up to 1 microM. In the presence of inhibitors such as N(G)-nitro-L-arginine methyl ester and 7-nitroindazole (NI), the Kd value further increased up to more than 100 microM. Addition of the cofactor, 5,6,7,8-tetrahydrobiopterin (H4B), increased the Kd value by 10-fold in the presence of L-Arg, whereas it decreased the value to less than one 250th in the presence of NI. Addition of H4B increased the recombination rate constant (k(on)) for CO by more than two-fold in the presence of L-Arg or N6-(1-iminoethyl)-L-lysine, whereas it decreased the k(on) value by three-fold in the presence of L-thiocitrulline. Thus, the binding fashion of some of inhibitors, such as NI, may be different from that of L-Arg with respect to the H4B effect.
