Cell therapy for ischemic heart disease.

Despite advances in pharmacological therapies, cardiovascular surgery, use of mechanical assist devices, and organ transplantation, more than 50% of the patients with clinically evident heart failure die within 5 years of the initial diagnosis. The use of cellular therapy offers a promising approach for both the prevention and treatment of heart failure. This review will discuss the current state of this emerging field and the prospects to introduce the method into clinical practice. Since functional restoration of the damaged heart presents a formidable challenge, developing strategies for the prevention of postinfarct heart failure remains of utmost priority. Recent research has provided evidence that several cell lines including adult or embryonic stem cells, skeletal myoblasts, fetal cardiomyocytes or fibroblasts may be useful in strategies that aim to both prevent and treat heart failure through establishment of new blood vessels supplying surviving heart muscle cells and replacement of damaged heart muscle cells themselves. It is therefore reasonable to anticipate that new strategies will be developed to optimize cell delivery, homing and survival in the failing myocardium improving myocardial recovery after acute or chronic deterioration.

In situ photoemission study of the room temperature ferromagnet ZnGeP2:Mn.

The chemical states of the ZnGeP(2):Mn interface which shows ferromagnetism above room temperature have been studied by photoemission spectroscopy. Mn deposition on the ZnGeP2 substrate heated to 400 degrees C induced Mn substitution for Zn and then the formation of metallic Mn-Ge-P compounds. Depth profile studies have shown that Mn 3d electrons changed their character from itinerant to localized along the depth, and in the deep region, dilute divalent Mn species (<5% Mn) was observed with a coexisting metallic Fermi edge of non-Mn 3d character. The possibility of hole doping through Mn substitution for Ge and/or Zn vacancy is discussed.

Nonlinear dynamics of a Bose-Einstein condensate in a magnetic waveguide.

We have studied the internal and external dynamics of a Bose-Einstein condensate in an anharmonic magnetic waveguide. An oscillating condensate experiences a strong coupling between the center of mass motion and the internal collective modes. Because of the anharmonicity of the magnetic potential, not only the center of mass motion shows harmonic frequency generation, but also the internal dynamics exhibit nonlinear frequency mixing. Thereby, the condensate shows shape oscillations with an extremely large change in the aspect ratio of up to a factor of 10. We describe the data with a theoretical model to high accuracy. For strong excitations we test the experimental data for indications of a chaotic behavior.

Performance of lactating sows in response to the dietary valine supply.

In order to determine the requirement of lactating sows for valine (Val) the effects of graded Val supplementations on feed intake, milk yield and changes of body weight of sows and piglets were examined using 54 multiparous sows over a total of 72 lactations (35 days). The sows were randomly assigned to six treatments (1-6). Each group received the same basic diet, covering the requirements of energy and nutrients except Val (4.5 g native Val, i.e. 3.5 g apparent ileal digestible Val). Adding crystalline l-Val dietary Val concentrations of 5.5, 6.5, 8.5, 10.5 and 14.5 g/kg, respectively, were realized. Feed intake of the sows was determined daily, body weight was determined on day 110 of gestation and day 1, 22 and 35 of lactation. The piglets were weighed on day 1, 21 and 35. The intake of piglet feed, which was offered from day 21, was determined at day 35 for each litter. Milk yield was determined using the weigh-suckle-weigh-method at day 13, 14, 20 and 21, fat and protein contents were analysed in hand-milked samples with infrared spectroscopy. Feed intake (kg/day) of the sows during lactation averaged 2.9, 4.3, 4.6, 4.6, 4.9 and 5.1 for treatments 1-6, respectively. For treatments 1 and 2 body weight losses of the sows during lactation were higher (1207 and 805 g/day, respectively) and milk production was lower (7076 and 8686 g/day, respectively), compared with the average of treatments 3-6 (625 g/day body weight loss; 9263 g milk/day). Daily weight gain of the piglets was also lower (146 and 171 g/day vs. 200 g/day) leading to lower weaning weights of 6.6 and 7.5 vs. 8.5 kg. As a consistent growth depression of the piglets in Val deficiency must be assumed, at least 6.5 g Val corresponding to 5.5 g apparent ileal digestible Val per kg lactation feed must be recommended.

Off pump coronary artery bypass grafting in EuroSCORE high and low risk patients.

OBJECTIVE: Off pump coronary artery bypass grafting (OPCAB) is claimed to reduce the operative morbidity and mortality in high risk patients. It was the aim of the study to compare the outcome of OPCAB patients classified as high- and low risk according to the EuroSCORE. METHODS: Medical records of patients undergoing off pump coronary artery bypass grafting (n=126) at our institution between 1998 and 2001 were retrospectively reviewed. We classified them into two subgroups: low risk (EuroSCORE < or = 5, n=72, male 58 (81%), female 14 (19%), age 61 (37-78) years) and high risk (EuroSCORE >5, n=54, male 32 (59%), female 22 (41%), age 73 (42-83) years). RESULTS: EuroSCORE high risk patients showed significantly higher rates of blood transfusion (70 vs 31%; P<0.0001), intraaortic balloon pump insertion (16 vs 3%; P=0.013), atrial fibrillation (43 vs 22%; P=0.014), and renal failure (13 vs 3%; P=0.028). ICU length of stay was significantly longer in the high risk group (25 vs 22 h; P=0.002). There was also a higher perioperative mortality in the high risk group (9 vs 0%; P=0.008). CONCLUSION: From these data we conclude that using off pump coronary artery bypass grafting results as predicted by the EuroSCORE can be achieved. OPCAB is safe for low risk patients. Major complications seem to occur preferentially in the high risk group.

Comparative study of a block copolymer morphology by transmission electron microscopy and scanning force microscopy.

Using transmission electron microscopy (TEM) and scanning force microscopy (SFM) together, it was possible to verify important structural features of a nanostructured bulk material such as the kp-morphology in an ABC triblock copolymer. By applying suitable imaging techniques during the SFM measurements it was possible to determine the morphology without additional manipulation steps in between. In comparison, TEM investigations on this type of material usually require selective staining procedures prior to the measurement. Also electron beam damage is often encountered during TEM measurements especially if components such as poly(methacrylates) are present. In contrast, SFM measurements can be assumed not to significantly change the phase dimensions of the components.

Bose-Einstein condensation in a surface microtrap.

Bose-Einstein condensation has been achieved in a magnetic surface microtrap with 4 x 10(5) (87)Rb atoms. The strongly anisotropic trapping potential is generated by a microstructure which consists of microfabricated linear copper conductor of widths ranging from 3 to 30 microm. After loading a high number of atoms from a pulsed thermal source directly into a magneto-optical trap the magnetically stored atoms are transferred into the microtrap by adiabatic transformation of the trapping potential. In the microtrap the atoms are cooled to condensation using forced rf-evaporation. The complete in vacuo trap design is compatible with ultrahigh vacuum below 2 x 10(-11) mbar.

Function of Neurolin (DM-GRASP/SC-1) in guidance of motor axons during zebrafish development.

Neurolin (zf DM-GRASP), a transmembrane protein with five extracellular immunoglobulin domains, is expressed by secondary but not primary motoneurons during zebrafish development. The spatiotemporally restricted expression pattern suggests that Neurolin plays a role in motor axon growth and guidance. To test this hypothesis, we injected zebrafish embryos with function-blocking Neurolin antibodies. In injected embryos, secondary motor axons form a broadened bundle along the common path and ectopic branches leave the common path at right angles. Moreover, the formation of the ventral and the rostral projection of secondary motor axons is inhibited during the second day of development. Pathfinding errors, resulting in secondary motor axons growing through ectopic regions of the somites, occur along the common path and in the dorsal and rostral projection. Our data are compatible with the view that Neurolin is involved in the recognition of guidance cues and acts as a receptor on secondary motor axons. Consistent with this idea is the binding pattern of a soluble Neurolin-Fc construct showing that putative ligands are distributed along the common path, the ventral projection, and in the area where the rostral projection develops.

Functional heterogeneity of ROMK mutations linked to hyperprostaglandin E syndrome.

BACKGROUND: The renal K(+) channel ROMK (Kir1.1) controls salt reabsorption in the kidney. Loss-of-function mutations in this channel cause hyperprostaglandin E syndrome/antenatal Bartter syndrome (HPS/aBS), which is characterized by severe renal salt and fluid wasting. METHODS: We investigated 10 HPS/aBS patients for mutations in the ROMK gene by single-strand conformation polymorphism analysis (SSCA) and direct sequencing. To assess the functional consequences, Ba(2+)-sensitive K(+) currents were measured in five mutants of the core region as well as one mutant with truncated C-terminus, using the two-electrode voltage-clamp technique after an injection of mutant cRNA into Xenopus oocytes. RESULTS: Three novel ROMK mutations were identified together with six mutations described previously. The mutations were categorized into three groups: (1) amino acid exchanges in the core region (M1-H5-M2), (2) truncation at the cytosolic C-terminus, and (3) deletions of putative promoter elements. While the core mutations W99C, N124K, and I142T led to significantly reduced macroscopic K(+) currents (1 to 8% of wild-type currents), the A103V and P110L variants retained substantial K(+) conductivity (23 and 35% of wild-type currents, respectively). Coexpression of A103V and P110L, resembling the compound heterozygous state of the affected individual, further reduced macroscopic currents to 9% of the wild-type currents. All mutants in the core region exerted a dominant-negative effect on wild-type ROMK1. The C-terminal frameshift (fs) mutation (H354fs) did not change current amplitudes compared with ROMK1 wild type, suggesting that a mechanism other than alteration of the electrophysiological properties may responsible for loss of channel activity. CONCLUSIONS: Analysis of ROMK mutants linked to HPS/aBS revealed a spectrum of mechanisms accounting for loss of channel function. Further characterization of the molecular defects might be helpful for the development of new therapeutic approaches.

[Rheumatology expertise].

Requests for expert appraisals in cases of osteoarthritic diseases increases continually, especially those addressed by the "Office de I'Assurance Invalidité (OAI)" or private insurances (PRIV). In this series of 413 cases, the mean age of the applicants was significantly lower than that of 10 years ago. There were more men than women, foreigners than Swiss natives and couples than people living alone but not significantly. 85% of complaints were for vertebral pain; fibromyalgia and depression were noted in 8% and 27% of cases respectively. Only 42% of those demanding time off work had heavy work; 66% did not continue their schooling beyond the minimum requirement. Less than 1/3 of work interruption for OAI and 1/6 for PRIV were confirmed; more than 1/2 of the patients for PRIV could not justify a work disability higher than 20%. 19% of our appraisals were disputed; 3% succeeded and 3% are still in progress. Chronic rheumatological complaints do not necessarily result in insurance compensations. Practitioners can contribute to chronification of rheumatological complaints by perpetuating work interruption for non medical reasons.

Neurolin, the goldfish homolog of DM-GRASP, is involved in retinal axon pathfinding to the optic disk.

Young axons of new retinal ganglion cells (RGCs) in the continuously growing goldfish retina fasciculate with one another and their immediate forerunners on their path toward the optic disk and along the optic nerve. They express the immunoglobulin superfamily cell adhesion molecules (CAMs) neurolin (DM-GRASP) and the L1-like E587 antigen. Repeated injections of Fab fragments from polyclonal antisera against neurolin (neurolin Fabs) into the eye of 3. 4-cm-long and rapidly growing goldfish caused highly aberrant pathways of young RGC axon subfascicles in the dorsal retina. Many axons grew in circles and failed to reach the optic disk. In contrast, E587 Fabs, used in parallel experiments, disrupted the fascicles but did not interfere with the disk-directed growth. Neurolin Fabs also disturbed axonal fasciculation in vivo as well as in vitro but less severely than E587 Fabs. Coinjections of both Fabs increased defasciculation of the dorsal axons in both aberrant and disk-directed routes. They also disrupted the order of young RGC axons in the optic nerve more severely than E587 Fabs alone. This demonstrates that the development of tight and orderly fascicles in the dorsal retina and in the optic nerve requires both E587 antigen and neurolin. More importantly, our results suggest an involvement of neurolin in RGC axonal guidance from the retinal periphery to the optic disk. Because disrupted fascicles and errant axon routes were found only in the dorsal retinal half, a cooperation with so-called positional markers may be conceived.

Expression of an L1-related cell adhesion molecule on developing CNS fiber tracts in zebrafish and its functional contribution to axon fasciculation.

E587 antigen, an L1-related cell adhesion molecule, is expressed by growing axons and has previously been shown to enhance axon growth and to mediate fasciculation of axons from newborn retinal ganglion cells in goldfish. In zebrafish, the monoclonal antibody E17 against E587 antigen stains all axons in the primary tracts and commissures from 17 h postfertilization (pf) onward and axons which are added subsequently to this scaffold. Moreover, Fab fragments of an E587 antiserum (E587 Fabs) injected into the ventricle of 30-h pf zebrafish embryos caused a marked defasciculation of distinct axon bundles in the posterior commissure, in hindbrain commissures, and in longitudinal tracts of the hindbrain, where they also caused increased crossings between fascicles. The regulated expression of E587 antigen by all developing axons and the effects caused by E587 Fabs show that E587 antigen contributes to the formation of tight and orderly fascicles in the developing CNS.

Clinical utility of contrast-enhanced MR angiography.

MR angiography (MRA) is a technique under ongoing discussion. Its non-invasiveness and sensitivity to flow irregularities make it an investigational technique which is easy to apply but which does not always lead to comprehensive results. It requires special skill to perform and also experience for correct interpretation of the results. The lengthiness of the procedure combined with certain physical properties tends to limit its use to mostly neurovascular applications. With the introduction of ultrafast MRA in conjunction with peripheral bolus-injection of extracellular contrast media, a new approach to the imaging of such regions as the thoracic and abdominal vasculature not to mention other vascular territories has become possible. In this paper, considerations of bolus and measurement optimization and timing protocols for dedicated indications are presented together with an overview on the experience acquired to date for CE-MRA of the carotid artery, mediastinal and pulmonary vessels, abdominal vasculature, and peripheral vessels. The main advantage of ultrafast CE-MRA is that patients are subjected to much more tolerable breath-hold investigations with the result that physiological motion such as peristalsis or major pulsation is minimised.
