Mechanism of action and selective toxicity of ascamycin, a nucleoside antibiotic.

An unidentified Streptomyces sp. produces two nucleoside antibiotics, ascamycin and its dealanyl derivative. In contrast to the broad antibacterial activity of dealanylascamycin against various gram-negative and gram-positive bacteria, ascamycin showed selective toxicity against Xanthomonas citri and X. oryzae. Both ascamycin and dealanylascamycin inhibited the protein synthesis of X. citri, but only dealanylascamycin inhibited that of Escherichia coli. In cell-free systems from E. coli and X. citri, both antibiotics, at ca. 0.04 micrograms/ml, inhibited the polyuridylate-directed synthesis of polyphenylalanine by ca. 50%. These data suggest that ascamycin cannot permeate the bacterial membrane. The dealanylating activity toward ascamycin was found only on the cell surface of bacteria susceptible to ascamycin. Dealanylascamycin must then have been transported into cytoplasm, where it inhibited protein synthesis.

Vectorcardiogram with McFee-Parungao lead system in spontaneously hypertensive rats.

Vectorcardiograms (VCG) were recorded from five spontaneously hypertensive rats (SHR) and five age-matched normal wistar rats (WR). Arterial systolic blood pressure was 115 +/- 10.8 mmHg in WR and 176 +/- 7.7 mmHg in SHR. The basic procedures for recording VCG in the rat were investigated. After rats were anesthetized with urethane-chloralose, VCG were taken by using the McFee-Parungao lead system in restrained ventral position. VCG in SHR were characterized by superior, posterior and leftward shift of QRS loops, a more elliptical form of QRS loops in the horizontal plane, and an increase in QRS-T angles in all planes, when compared with WR. These vectorcardiographic findings in SHR were quite similar to those produced by left ventricular hypertrophy in humans.

Translation of human macrophage activating factor (for glucose consumption) mRNA in Xenopus laevis oocytes.

Total messenger RNA was extracted from a human T cell hybridoma, clone H-E4-9, which strongly produced macrophage activating factor for glucose consumption (MAF-G). This messenger RNA gave rise to functional MAF-G when translated in Xenopus laevis oocytes. Isoelectric focusing of culture supernatants of the mRNA-microinjected oocytes and the H-E4-9 cells revealed that the former contained MAF-Gs with isoelectric points of pH 5.0 and 3.0 while the latter contained MAF-Gs with isoelectric points of pH 5.0 and pH 3.3. Sucrose density gradient centrifugation analysis showed that MAF-G mRNA prepared from H-E4-9 cells sedimented at about 11.5 S.

Synthesis and biological activity of nucleoside antibiotics, ascamycin and its amino acid analogs.

Synthesis of an alanylsulfamoyl nucleoside antibiotic, ascamycin was achieved by the condensation of N6-t-butyloxycarbonyl-2-chloro-9-(2',3'-O-isopropylidene-5'-O-sulfamoyl- beta-D- ribofuranosyl)adenine(3) with t-butyloxycarbonyl-L-alanylimidazole in the presence of NaH in DMF. Deprotection with 90% trifluoroacetic acid gave ascamycin in 61% overall yield. This procedure may be applicable for preparation of a number of amino acid analogs of ascamycin.

Changes in the serum LDH isoenzymes in monkey during chronic exposure to simulated high altitude.

When the monkey was exposed to a simulated high altitude of 18,000 ft (5,486 m) for 30 d, the serum LDH activity was increased to its maximum in 1 week. After the monkey returned to sea level, the enzyme activity showed a rapid recovery. On the basis of electrophoretic analysis of LDH isoenzymes, the percentages of LDH-1 and LDH-2 were decreased during the first 2-3 weeks of hypoxic exposure whereas those of LDH-3, LDH-4 and LDH-5 were increased. After 2 or 3 weeks of hypoxic exposure, LDH-3, LDH-4, and LDH-5 become predominant, indicating that the isoenzyme pattern shifted to an anaerobic form from an aerobic form. These results clearly showed that relative proportions of the five isoenzymes were significantly altered by chronic hypoxia of high altitude and that the elevation of total serum LDH activity induced by hypoxia came predominantly from rapid accumulation of M type LDH isoenzymes in serum.

The structure of health care service--so-called the worried well patients and psychosomatic medicine.

The desire of people to receive the best health care services without difficulty is an instinctive one. However, defects in public health education and the irresponsible or sometimes sensational medical information released by the mass media are causing an excessive increase in the population's demand for health care services. As a result, so-called "worried well" patients are increasing and health care institutions are crowded with such patients, who play a major role in increasing national medical spending. On the other hand, it is difficult to say that health care institutions are providing satisfactorily continuous, consecutive and comprehensive health care services. Needless to say, in dealing with worried well patients, the health care institutions should make efforts towards early detection of disease, if any, but at the same time, a psychosomatic approach should be made, considering patients as holistic people with both a mind and body. To this end, interdepartmental and interhospital liaison becomes very important. From the viewpoint of comprehensive health care services, the author presents in this paper such cases where the health care services provided are regarded as defective in terms of continuity and points out the importance of systematized health care information and systematized health care services at all health care levels from primary to tertiary.

Rotational dynamics of immunoglobulins with fluorescent haptens on a membrane surface.

The rotational dynamics of rabbit immunoglobulin G with fluorescent lipid haptens on a membrane surface has been studied by nanosecond fluorescence emission anisotropic spectroscopy. It has been found that the rotational angles of the antibody are very restricted on the membrane, but that the rotation rate itself is not appreciably lower than that in solution, and is independent of the membrane fluidity.

Increased novelty-induced grooming in aged rats: a preliminary observation.

Age-related changes in grooming activity in female Fischer-344 rats were observed in a home cage and in a novel testing chamber for 50 min. Compared to the home cage condition with brief handling treatment, excessive grooming was found in the novel situation. The amount of novelty-induced grooming of aged rats (26-28 months) was about two times that of young animals (6-8 months). This increase in novelty-induced grooming of aged animals was attributable to an increase in the number of grooming bouts, prolongation of each grooming bout, and a slight increase in the duration of licking elements. However, there were no age differences in the percentage of face washing and the duration of face-washing elements. These results are discussed in terms of behavioral aging and age-related changes in peptidergic pathways in the brain.

Structure and biological activity of ascamycin, a new nucleoside antibiotic.

A newly isolated strain of Streptomyces sp. produces a new nucleoside antibiotic, ascamycin and the corresponding dealanyl derivative. The structure of ascamycin was determined to be 2-chloro-9-beta-[5-O-(N-L-alanyl)sulfamoyl-D-ribofuranosyl]-adenine. Remarkable selective toxicity of ascamycin compared to the dealanyl derivative was accounted for on the basis of a dealanylating enzyme present in the envelope of sensitive bacteria. After dealanylation, it becomes permeable to cell membrane.

[Planning a health care delivery system and psychosomatic medicine].

This paper describes why planning a health care delivery system which aims at appropriate distribution of health and medical resources, as well as making an effort toward its urgent realization, is necessary. The relationship between the practice of comprehensive medical care and the significance of primary care is also explained. In giving comprehensive medical health care for the whole person, the author considered the important role that psychosomatic medicine must play in dealing with previously reported cases, including the author's cases. Furthermore, concerning primary care and the universal practice of psychosomatic medicine, their real value is as a basis for planning a health care delivery system. Therefore, the primary purpose of this paper is to show that the systematic development of a health care delivery system is of the utmost importance.
