Purification and properties of human serum esterase.

Human serum esterase was purified by affinity column chromatography on a column of covalently linked p-trimethylammoniumanilinium dichloride to Sepharose 4B. The purified preparation hydrolysed both benzoylcholine and tributyrin. p-Trimethylammoniumanilinium dichloride inhibited non-competitively the hydrolysis of tributyrin and inhibited competitively the splitting of benzoylcholine. The Km value was 0.62 . 10(-3) M for tributyrin and 0.4 . 10(-3) M for benzoylcholine. Antiserum to this purified esterase was prepared in rabbit and it was found that the antiserum did not inhibit esterase activities of human liver, muscle and adipose tissue, although it could inhibit completely the esterase activities of human serum.

Blood vessel transplantation to bone.

In dogs an artery, a vein, and a vascular bundle were transplanted into intact bone, isolated bone segments, necrotized bone, and homografts of bone. Active proliferation of new blood vessels and formation of new bone occurred in all instances where the vascular bundle was used, when the vein was implanted into the intact bone and isolated bone, and only when the artery was implanted into intact bone. As a result of these experiments, vascular bundle transplantation has been performed in patients with Kienboeck's disease, and avascular necrosis of the scaphoid, the femoral head, the talus, and other conditions.

Age related gross changes of the liver and right diaphragm, with special reference to partial eventration.

A total of 38937 6 X 6 cm chest films, made in three series of check-up programmes for local residents above the age for nursery school, were examined for partial eventration of the right dome of the diaphragm in relation to age. It was found that after nursery school age, eventration and irregularity of the right dome increased with age, particularly among women over the age of 60. In a series observed in a rural region, marked eventration was found in more than 1% of the women above 60 years. In another study on 1 332 liver scans in patients without intrinsic liver disease, a prominent, dome-like elevation of the right lobe of the liver was also very frequent among older women. It is concluded that the majority of right-sided eventrations are acquired and may be attributable to dietary and dressing habits.

Effects of pantetheine on cholesteryl ester synthesis in the arterial wall of rats on high cholesterol diet.

Increase of acyl-CoA synthesis was observed when extracts of rat arterial wall were incubated with pantetheine [D-bis-(N-pantothenyl-beta-aminoethyl)-disulfide]. Cholesteryl ester synthesis from palmitate in the arterial wall extract in vitro was higher with arteries from rats on high cholesterol diet than with those from rats on normal diet, but the synthesis was reduced in the arteries of rats on high cholesterol diet with pantetheine. Triglyceride synthesis was higher with arterial wall extracts of rats on high cholesterol diet than with preparations from rats on normal diet and was not reduced with those of rats on high cholesterol diet plus pantetheine. The value of the effects of pantetheine on lipid metabolism in the prevention of atherosclerosis is pointed out.

Role of phospholipid in adrenaline-induced lipolysis and cyclic AMP production.

Lipid micelles consisting of a glyceride mixture (triolein, diolein, and monoolein) and lecithin bound adrenaline-14C more strongly than did micelles consisting of the glyceride mixture only. Lipid micelles consisting of the glyceride mixture and phosphatidic acid also bound adrenaline-14C effectivily, whereas lipid micelles consisting of the glyceride mixture and diglyceride, obtained from lecithin, did not bind the hormone strongly. Both phenoxybenzamine (an alpha- blocker) and propranolol (a beta-blocker) strongly inhibited the association between adrenaline-14C and lipid micelles consisting of the glyceride mixture and lecithin. Propranolol, inhibited adrenaline-induced lipolysis in both fat cells and fat globules, whereas, phenoxybenzamine, did not affect adrenaline-induced lipolysis. Both agents reduced adrenaline-induced adenyl-cyclase activation in fat cell ghosts. Phospholipid was also found to be related with adrenaline-mediated adenylcyclase activation.

Studies on reduction of lipolysis in adipose tissue on freezing and thawing.

Adrenaline-induced lipolysis in fat cells was remarkably reduced when the cells were preincubated in a dry ice-aceton bath, but their adenylcyclase and lipase activities were not reduced. In the reconstructed lipid micelles which consisted of lipase-depleted lipid micelles and lipase-containing adipose tissue extract, adrenaline, theophylline and DBcAMP-induced lipolysis was not found when lipase-depleted lipid micelles were preincubated in a dry ice-aceton bath but was found when lipase was preincubated.

Studies on the mechanism of action of ACTH on triglyceride synthesis in fat cells.

It was found that ACTH greatly reduced lipogenesis in fat cells in the presence of calcium ion, but not in the absence of calcium ion. Of the enzymes involved in triglyceride synthesis from fatty acid in lipid micelle membranes, only acyl-CoA synthetase was inhibited by calcium ion, the apparent Ki value of calcium ion being 4.2 X 10(-4) M. The Km values of the enzyme for palmitate and ATP were 2.0 X 10(-4) M and 2.5 X 10(-4) M, respectively and calcium ion caused non-competitive inhibition with both palmitate and ATP. The acyl-CoA synthetase activity of lipid micelle membranes was inhibited by treatment with phospholipase A or C, but not by treatment with phospholipase D. The mechanism of inhibition of triglyceride synthesis by ACTH is discussed on the basis of these results.
