Immunobiological activities of synthetic lipid A analogs with low endotoxicity.

Synthetic lipid A analogs, beta(1-6)glucosamine disaccharide 1,4'-bisphosphates, which possesses four tetradecanoyl groups at the 2- and 2'-amino, and 3- and 3'-hydroxyl groups (LA-17-PP), and each two of the (R)-3-hydroxytetradecanoyl and tetradecanoyl groups at the 2- and 2'-amino and 3- and 3'-hydroxyl groups, respectively (LA-18-PP), were far less endotoxic than synthetic (506, LA-15-PP) and bacterial Escherichia coli type lipid A's; neither compound showed any detectable lethal toxicity in chicken embryos or preparatory activity for the local Shwartzman reaction in rabbits. Also both compounds were only weakly pyrogenic and comparably less lethally toxic in galactosamine-loaded mice than the reference synthetic and bacterial lipid A's and a synthetic counterpart to biosynthetic lipid A precursor Ia (406, LA-14-PP). Nevertheless, LA-17-PP and LA-18-PP exhibited definite in vivo immunoadjuvant activity in mice, and the ability to induce a possible tumor necrosis factor and alpha/beta interferon in Mycobacterium bovis BCG and Propionibacterium acnes-primed mice, respectively, although these activities were weaker than those of the reference lipid A's. 4'-Monophosphate analogs of the above two test compounds exhibited neither endotoxic nor beneficial activities, but they showed remarkable in vitro bioactivities comparable to those of the corresponding bisphosphate compounds; the ability to activate the human complement system and the clotting enzyme cascade of horseshoe crab amoebocyte lysate, stimulatory effects on guinea pig and murine peritoneal macrophages, and murine splenocytes.

Werner's syndrome associated with cholangiocarcinoma.

A 38-year-old Japanese man had cholangiocarcinoma in association with typical features of Werner's syndrome. The replicative capacity of fibroblasts in culture was characteristically reduced, and the in vitro natural killer cell activity was deficient. He died of massive G-I tract bleeding 8 months after admission.

Heterogeneity of epithelial cells and reactive components in thymomas: an ultrastructural and immunohistochemical study.

Fourteen thymomas were studied by electron microscopy and immunohistochemistry. Based on the ultrastructure of the neoplastic epithelial cells in comparison with normal thymic epithelium, four cortical-, three mixed-, five medullary-, and two corpuscular-type tumors were categorized. Histologically the tumors of cortical type showed prominent lymphocytic infiltration, but scant interdigitating reticulum cells (IDCs) were demonstrated by immunoperoxidase method on paraffin sections with anti-S-100 protein antiserum. Fewer lymphocytes but more IDCs were present in the tumors of medullary and corpuscular types, although variable in those of mixed type. This corticomedullary difference among thymomas was confirmed in some of them by the immunoperoxidase method on frozen sections with monoclonal antibodies. The cortical-type tumors were HLA-DR positive in tumor cells and infiltrated predominantly with cortical thymocytes (OKT-6+, OKT-3-, both Leu 3a/3b+ and OKT-8+), whereas the medullary- and corpuscular-type tumors were HLA-DR positive primarily in IDCs but not in tumor cells and were infiltrated more with medullary thymocytes (OKT-6-, OKT-3+, either Leu 3a/3b+ or OKT-8+). The classification of thymomas based on neoplastic epithelial cells will serve to refine the traditional classification based on reactive lymphocytes.

Small cell undifferentiated carcinoma of the uterine cervix: histology, ultrastructure, and immunohistochemistry of two cases.

Two clinical stage IB small cell undifferentiated carcinomas (SCUC) of the cervix were studied by light and electron microscopy and immunohistochemistry. Both cases occurred in women aged less than 31 years. Despite radical hysterectomy and external pelvic radiotherapy, both patients died of recurrent disease within 14 months after initial therapy. The tumors consisted of sheets of closely packed, uniform small, round to oval cells with hyperchromatic nuclei and scant indistinct cytoplasm. One case was associated with cervical squamous cell carcinoma. The neoplastic cells had few organelles and desmosome-like junctions and lacked mucinous or neurosecretory granules or tonofilaments. Immunohistochemistry failed to reveal S-100, CEA, neuropeptides or neuron-specific enolase. SCUC probably arises either from basal cells of the cervical squamous epithelium, or gland cells of the endocervical epithelium, or still from subcolumnar endocervical reserve cells. Based on ultrastructure and immunohistochemistry, SCUC seems to represent the undifferentiated variant of small cell neuroendocrine tumors of the cervix.

[Analysis of serum CA125, CEA, AFP, LDH levels and LDH isoenzymes in patients with ovarian tumors--correlation between tumor markers and histological types of ovarian tumors].

Serum CA125, CEA, AFP, LDH levels and LDH isoenzymes were analyzed in ovarian tumor patients, who were treated at Kyoto University Hospital. CA125 was positive in 10/16 (62.5%) cases of common epithelial carcinoma, especially 100% positive in serous carcinoma, but was negative in mucinous tumors. CA125 was also negative in patients with germ cell and sex cord stromal tumors. CEA was positive in 13/32 (40.6%) cases of epithelial carcinoma, most frequently elevated in patients with mucinous carcinoma, pseudomyxoma peritonei, and Krukenberg tumor. AFP was positive only in those with endodermal sinus tumors. LDH was elevated in 16/39 (41%) cases of epithelial carcinoma, but was not specific for histological types. In contrast, all 8 cases of dysgerminoma, 1 of immature teratoma and 2 of endodermal sinus tumor showed extremely elevated LDH levels. Moreover, the normal pattern or deviation to H subunit of LDH isoenzymes was seen in such cases of germ cell tumor, while deviation to M subunit was noted in epithelial and metastatic tumor patients. These data indicate that each parameter is useful as a tumor marker for the specific histological type of ovarian tumor; CA125 for non-mucinous epithelial carcinoma, CEA for mucinous tumor and Krukenberg tumor, AFP for yolk sac tumor, LDH and LDH isoenzymes for dysgerminoma and other solid germ cell tumors. In addition, preoperative diagnosis of histological types of ovarian tumors may be possible by combining these tumor markers.

Ovarian fibroma with Leydig cell hyperplasia of the adjacent stroma: a light and electron microscopic study.

A virilizing left ovarian tumor removed from a 58-year-old woman was studied by light and electron microscopy. Histologically, the tumor was an ovarian fibroma around which Leydig cells, but no Sertoli cells, proliferated at a distance from the hilus. Although the fibroma itself did not contain Leydig cells, several Leydig cells were observed intermingled with ovarian stromal cells in the cortical tissue compressed by the fibroma. Ultrastructurally, in addition to mature Leydig cells with typical steroid-producing--cell features, immature cells with less-developed smooth endoplasmic reticulum and elementary tubular inclusions were identified. These light and electron microscopic findings suggest that the Leydig cells may have differentiated from ovarian stromal cells surrounding the fibroma. If so, this case should be distinguished from neoplastic disorders such as hilar cell tumors or stromal-Leydig cell tumors and be classified in the category of ovarian tumors with functioning stroma containing Leydig cells.

[Effect of rotenone on rabbit ovulation and histochemical activities of cytochrome oxidase and 3 beta-hydroxysteroid dehydrogenase of the follicle].

Various doses of rotenone, an inhibitor of respiratory chain, were administered to mature female rabbits concomitantly with an ovulatory dose of hCG. The effect of rotenone on ovulation was studied by counting the ovulated stigma under a dissecting microscope. Histochemical studies on the activities of 3 beta-hydroxysteroid dehydrogenase (3 beta-HSD) and cytochrome oxidase (CYO) in follicles at various intervals after hCG-rotenone injections were also performed to investigate the role of mitochondrial oxidation in the ovulatory process. Rotenone inhibited the hCG-induced ovulation in a dose respondent manner and reduced the sudden hCG-induced increase in the histochemical activities of 3 beta-HSD and CYO of granulosa cells. It is suggested that the activation of mitochondrial oxidation in the ovulating follicle is mandatory for ovulation and the induction of steroidogenic enzymes in granulosa cells.

[Effect of PGF2 alpha on ovulation and collagenolytic activities in the rabbit treated by indomethacin].

To clarify a regulatory role of prostaglandins in collagenolytic enzyme activities of the follicular wall at ovulation, we studied whether the ovulation-blocking effect of indomethacin (IM) on hCG-stimulated rabbits could be reversed by an intravenous infusion of PGF2 alpha. The activities of BANA hydrolase and DNP peptidase were also measured by using synthetic substrates N-benzoyl-DL-arginine-2-naphthylamide HCl (BANA) and DNP-Pro-Gln-Gly-Ile-Ala-Gly-Gln-D-Arg-OH (DNP peptide), respectively, every hour during the specific preovulatory period. The ovulation-blocking effect of IM (4mg/kg) was reversed by an intravenous infusion of PGF2 alpha (1.5 mg/kg) starting at 3 hr after the hCG injection. In the control rabbits treated with hCG(100 iu) alone, these enzymatic activities were increased toward ovulation with the peak level 7-9 hr after a hCG injection and decreased significantly at 10 hr. The preovulatory increase in these enzymatic activities disappeared following the ovulation-blocking doses of indomethacin. By the addition of the PGF2 alpha infusion, the IM-induced changes in BANA hydrolase and DNP peptidase activities were returned close to the control. It is suggested that the preovulatory increases in BANA hydrolase and DNP peptidase are mandatory for ovulation and PGF2 alpha facilitates ovulation through its regulatory action on these enzymes.

Development of interstitial cells and ovigerous cords in the human fetal ovary: an ultrastructural study.

Differentiation and subsequent regression of interstitial cells in the human fetal ovary was studied by light and electron microscopy in specimens obtained from twelve fetuses between 12 and 40 weeks of gestation. Interstitial cells possessing the ultrastructural features associated with steroid production first differentiate from fibroblast-like cells by 15 weeks. This occurs within the medullary fibrous tissue penetrating the cortex. The number of such cells is maximal at 18 weeks around the ovigerous cords in the inner half of the cortex. In addition, gap junction formation is seen in the pregranulosa cells forming ovigerous cords. From 21 to 31 weeks primordial follicles develop from ovigerous cords, which themselves develop from undifferentiated cortex, in a sequence that is most advanced in the medullary portion of the gonad and least advanced at its surface. During this time the interstitial cells decrease in number. They are not observed around the follicles, but are occasionally seen in the outer layer of the cortex which is still at the stage of ovigerous cords. At 40 weeks, growing follicles associated with several layers of enveloping theca cells are observed in the innermost region of the cortex. Interstitial cells are rare in all parts of the ovary. The development of interstitial cells independent of follicles in the human fetal ovary implies that such cells are distinct from theca cells, and that they correspond to the primary interstitial cells of other mammalian species. In addition anatomical and temporal relationships are observed between the interstitial cells, the ovigerous cords, developing primordial follicles and the formation of gap junctions in pregranulosa cells.

Distribution of human leumorphin-like immunoreactivity in the monkey spinal cord revealed by immunocytochemistry.

The regional distribution of human leumorphin (HL)-like immunoreactivity (HL-LI) in monkey (Macaca fuscata) spinal cord and dorsal root ganglia was investigated by peroxidase-anti-peroxidase immunocytochemistry using specific antiserum. HL-LI-positive fibers and terminals were distributed densely in laminae (Rexed) I and II, and sparsely in laminae III-VII and X, but no immunoreactive elements were observed in the ventral horn, the white matter or the dorsal root ganglia. Many immunoreactive neuronal perikarya were found in laminae I and II. Intrathecal injection of colchicine also revealed the presence of immunoreactive neuronal perikarya in laminae III-VII and X. These results suggest the presence of HL-LI, which represents HL and/or its C-terminal fragment, in the neuronal elements of the monkey spinal cord.

The influence of serotonergic inputs on peptide neurons in the rat suprachiasmatic nucleus: an immunocytochemical study.

The influence of serotonin (5-hydroxytryptamine; 5-HT) innervation on peptide-containing neurons in the rat suprachiasmatic nucleus (SCN) was investigated by peroxidase-anti-peroxidase (PAP) immunocytochemistry. The 5-HT neuronal system was chemically severed by 5,6-dihydroxytryptamine (5,6-DHT) injection into the medial forebrain bundle bilaterally. After this treatment, a marked decrease of vasoactive intestinal peptide (VIP)-like immunoreactivity in neuronal perikarya occurred in the SCN corresponding to a decrease in number of 5-HT immunoreactive fibers and terminals. However, no alteration of arginine-vasopressin-like immunoreactivity was detected between 5,6-DHT-treated animals and the controls. It is speculated that VIP-like immunoreactive neurons play an important role in the SCN under the influence of strong 5-HT innervation.

Activation of urinary inactive kallikrein by an extract from the rat kidney cortex.

Activation of purified urinary inactive kallikrein by an extract from the rat kidney cortex was investigated. The extract produced a dose-dependent activation of the inactive kallikrein and the optimum pH for this activation was 5.0. Marked depression of the activation was observed when the extract was pre-incubated with E-64, p-CMB and iodoacetate, but not with DFP, PMSF or pepstatin A. The molecular weight of the inactive kallikrein (Mr 44,000) was reduced to 38,000 by treatment with the extract, this molecular weight value being identical with that of urinary active kallikrein. These results indicate that the rat kidney cortex contains a protease catalyzing conversion of urinary inactive kallikrein into its active form, and that the protease has properties compatible with those of a thiol protease, but not of trypsin which has been used as a tool for the activation of urinary inactive kallikrein. The thiol protease is probably one of regulators of the kallikrein-kinin system in the kidney.

Light and electron microscopic immunocytochemistry of beta-endorphin/beta-LPH-like immunoreactive neurons in the arcuate nucleus and surrounding areas of the rat hypothalamus.

beta-Endorphin/beta-LPH-like immunoreactive neurons in the hypothalamic arcuate nucleus and its surrounding areas were visualized by light and electron microscopic immunocytochemistry. Immunoreactive processes were found in the vicinity of the pia mater, in the lateral part of the external layer of the median eminence and near the lateral wall of the third ventricle. Neuronal perikarya contained immunoreactive dense granules as well as developed cell organellae. They received neuronal inputs from other neurons through axoplasmic and axodendritic synapses. Immunoreactive neuronal processes containing dense granules and mitochondria were found as preterminal elements on non-immunoreactive neuronal soma and dendrites. Immunoreactive processes also make intimate contact with capillaries in the arcuate nucleus near the median eminence.

Immunocytochemical localization of substance P in the rat spinal cord with special reference to fibers within the ventral column of the rostral lumbar segments.

The distribution of substance P (SP) in the rat spinal cord was investigated by peroxidase-anti-peroxidase (PAP) immunocytochemistry. A dense network of SP-immunoreactive fibers and terminals was detected in the ventral column of the rostral lumbar cord with a different density and extent from the other segmental levels. These fibers and terminals were accumulated within and around the centromedial nucleus (CM) of the L1 and L2 segments, with some bundles of immunoreactive fibers between the CM and other areas; i.e. laminae V and X and the contralateral CM. They formed a dense network, such as in arborization of immunoreactive fibers and terminals on the transverse plane and in a comb-shaped structure on the horizontal plane. The origin of the SP in this network was examined. It was determined that neither a total transection of spinal cord at a low thoracic level or mid-lumbar level, nor at an ipsilateral or bilateral section of the 3-5 dorsal roots, containing L1 and L2 roots, induced any visible changes in the SP staining pattern. An intrathecal injection of colchicine revealed the presence of SP-immunoreactive neurons in the dorsal horn and intermediate gray matter at the spinal cord including the rostral lumbar cord. The present findings suggested that the majority of SP immunoreactivities in the above network are derived from local circuit interneurons of the spinal cord.

Induction of histidine decarboxylase activity in the spleen of mice treated with staphylococcal enterotoxin A and demonstration of its non-mast cell origin.

Histidine decarboxylase (HDC) activity increased 13-, 7-, and 2-fold in the spleen, lung and liver, respectively, but not in other tissues of C57BL/6 mice injected i.v. with 50 micrograms/kg of Staphylococcal enterotoxin A (SEA). But even in the spleen, increase in the histamine level was only 1.5 times that of untreated mice. In genetically mast cell-deficient WBB6F1 - W/Wv mice HDC activity in the spleen increased to the same extent as in wild type WBB6F1 - +/+ mice on SEA treatment, but the histamine level in the spleen also increased 20-fold, whereas it increased only 1.4-fold in +/+ mice. These results suggest that the increases in HDC and histamine resulted from interaction of SEA with non-mast cells in tissues.

Analysis of serum lactic dehydrogenase levels and its isoenzymes in ovarian dysgerminoma.

The usefulness of serum lactic dehydrogenase (LDH) levels and its isoenzyme analysis as potential malignant ovarian tumor markers was investigated in 74 patients, including 5 cases of ovarian dysgerminomas. In this study 41.7% of patients with ovarian adenocarcinomas of the common epithelial types and 29.4% of cases with metastatic ovarian carcinoma had elevated preoperative serum LDH levels. In contrast all 5 cases of ovarian dysgerminomas had extremely high levels of serum LDH prior to therapy, whereas all 11 cases of benign solid ovarian tumors had normal LDH levels. The analysis of LDH isoenzymes revealed that ovarian dysgerminomas had high levels of the two fast fractions of LDH, whereas other malignant ovarian tumors had elevated levels of the two slow fractions. The present findings indicate that serum LDH levels and electrophoresis of its isoenzymes may be of value in patients with ovarian dysgerminomas.
