Changes in aminolevulinate synthase and aminolevulinate dehydratase activity in cirrhotic liver.

Delta-Aminolevulinate synthase and delta-aminolevulinate dehydratase activities were determined in liver biopsy specimens obtained from 12 patients with hepatic cirrhosis. Delta-Aminolevulinate synthase activity was determined by the incorporation of [1,4-14C]succinyl coenzyme A into delta-aminolevulinate. The mean activity of delta-aminolevulinate synthase was significantly higher in cirrhotic liver specimens (mean +/- SE, 193.7 +/- 34.5 picomoles delta-aminolevulinate per milligram protein per 30 minutes) than in controls with minimal histologic changes (32.7 +/- 13.6, p less than 0.01). Furthermore, the mean activity of delta-aminolevulinate synthase was higher in micronodular cirrhosis (281.6 +/- 58.8) than in the other types of cirrhosis (131.0 +/- 23.1, p less than 0.05). Levels of indocyanine green retention at 15 min correlated with the activity of hepatic delta-aminolevulinate synthase (p less than 0.05). The mean activity of delta-aminolevulinate dehydratase, in contrast, was significantly lower in cirrhotic liver specimens (9.4 +/- 1.3 nanomoles porphobilinogen per milligram protein per hour) than in controls (22.0 +/- 2.6, p less than 0.05). These results suggest that the extent of liver injury or the degree of portosystemic shunting, or both, influence the rate of hepatic heme biosynthesis.

[Case of three peculiar tumors of gastric epithelioid leiomyosarcoma, retroperitoneal paraganglioma and pulmonary chondroma].

A 27-year-old woman with an unusual combination of three independent tumors, gastric epithelioid leiomyosarcoma, retroperitoneal paraganglioma and pulmonary chondroma is reported. There are 15 similar cases collected by Carney (1979) from the literature; ours is the first Japanese case. Patients with this tumor triad tend to be young females, all were of blood group A. The prognosis of these patients is generally good without associated systemic hypertension. Our patient is well 18 months after surgical removal of the three tumors.

[Distribution of 67Ga-citrate in 3'-methyl-4-dimethylaminoazobenzene-induced hepatoma of rats].

The distribution of 67Ga-citrate in the hepatoma of rat induced by 3'-methyl-4-dimethylaminoazobenzene was studied. 67Ga uptake ratio resected specimen, autoradiography and histological specimen were compared each other. 67Ga uptake ratio of the tumor was increased 1.6 to 7.2 times (average 4.4) to control group. Regardless of the size of the tumor, macroautoradiographically observed distribution of 67Ga-citrate in the hepatoma was higher in the peripheral zone than in the central zone. Histologically the degeneration of tumor cell was low or absent in the peripheral zone of tumor, whereas it was intense in the central zone. 67Ga-citrate was highly accumulated in the zone which the degeneration was low or absent. We, however, could not demonstrate the site where 67Ga-citrate was incorporated.

Synthesis and structure-activity relationship of spiro[isochromanpiperidine] analogues for inhibition of histamine release.

Two types of 1'-alkylspiro[isochroman-3,4-piperidines] and 1'-alkylspiro[isochroman-4,4'-piperidines] were prepared and examined for their biological activity. Several of the compounds inhibited the compound 48/80 induced histamine release from isolated rat peritoneal mast cells. The structural requirements for this activity in the present series are discussed.
