A case of AFP producing rectal cancer was represented in this paper. The patients, 68 years old man, was admitted because of anal bleeding and preoperative examinations revealed rectal cancer without liver metastasis and hepatocellular carcinoma. Serum AFP measured preoperatively was 2750-3500 ng/ml and CEA was 23-33 ng/ml. The patient underwent amputation of the rectum and there were no abnormal masses in the liver at surgery. After operation serum AFP and CEA levels were normalized. Histology of the rectal tumor showed a tubular adenocarcinoma of moderately differentiated type. In the specimen, AFP producing tumor cells were detected by immunoenzymatic labelling (PAP) methods.

The effects of rifampicin (RMP) on isoniazid (INH) metabolism in rabbits were examined from the viewpoint of extensive hepatitis. After the RMP pretreatment, no remarkable changes were observed in the plasma levels of INH as well as its metabolites, acetylisoniazid (AcINH), acetylhydrazine (AcHz) and diacetylhydrazine (DAcHz) with the exception of hydrazine (Hz). After an oral administration of INH or Hz hydrate, the stochastic examination showed that the AUC0-8hr values of Hz plasma levels in RMP pretreated groups were significantly less than those in the control rabbits. RMP treatment was also shown to induce rabbit liver cytochrome P-450 activity. Histological studies demonstrated that Hz causes more remarkable hepatic necrosis in rabbits pretreated with RMP than in the control rabbits. These observations could suggest that Hz is a key intermediate of INH-hepatitis through the transformation of some hepatotoxic species by microsomal oxidation that is facilitated by RMP.
We have studied ouabain-resistant, external sodium-stimulated, lithium efflux (Li-Na countertransport) in red blood cells from 21 borderline hypertensives with at least one hypertensive first degree relative (BH-F), 19 borderline hypertensives without family history of essential hypertension (BH-NF), and 35 age-matched normotensive subjects. The data indicate the finding of an increased Li-Na countertransport in all BH (F+NF), but with a significant overlap between BH values and control ones: Li-Na countertransport is significantly higher only in BH-F but it is normal in BH-NF. Moreover, there is a significant correlation of Li-Na countertransport to total peripheral resistance but not to mean blood pressure in all hypertensive patients. It is suggested that in BH the increase of erythrocyte Na flux is mediated by the Na-Na exchange diffusion, and its abnormality may be associated to the hereditary trait of essential hypertension rather than the high blood pressure per se, probably resulting in the development of hypertension, through the increased vascular smooth muscle tone.
The head shell of bacteriophage lambda expands by about 20% in diameter when it packages the DNA molecule in vivo. The expansion reaction is essentially a conformational change of the major head protein molecules to a state of lower free energy and can also be triggered in vitro by treatment with 4 M-urea. In order to investigate the conformational change, we have measured the circular dichroism, fluorescence and difference absorption spectra of the lambda head shell before and after the expansion by the treatment with urea. The far-ultraviolet circular dichroism spectra and the fluorescence spectra show that the expansion is not accompanied by a great change in the secondary structure (29% alpha-helix, 23% beta-structure) and the environment (non-polar) of the tryptophan residues of the major head protein molecule. On the other hand, by measurements of the circular dichroism and difference absorption spectra in the near-ultraviolet region as well as by chemical modification experiments with tetranitromethane, we have found that one or two tyrosine residues of the major head protein are transferred from a polar, solvent-exposed to a non-polar, solvent-unexposed environment during the expansion. Judging from these results, the conformational change seems to be mainly intermolecular or interdomainal rather than intradomainal.
During the period from July to November 1981, 42 out of 128 young mink of a flock were found to possess antibodies against the viruses A/Bangkok/1/79 (H3N2) and A/Kumamoto/37/79 (H1N1), which were currently prevailing human influenza viruses. Seroconversion against A/Bangkok/1/79 was found in 12 mink from August to November. HI antibody titers of greater than 1: 128 were found in 8 out of 42 mink at the first examination in July and August. These findings suggest that infection with these human influenza viruses was present in this flock during the period from birth (the beginning of May) to autumn, the non-prevalent season in man. Attempts at virus isolation were unsuccessful.
The degree of polarization of the intrinsic fluorescence of purified myosin was estimated. On addition of ATP, polarization of the fluorescence of myosin increased when excited at wavelengths longer than 300 nm. In kinetic studies, coupled with the decay of the increased intensity of fluorescence of myosin, the increased polarization of the fluorescence decreased when the ATP was depleted. The decay of the increased polarization of fluorescence of myosin was specific to MgATP. According to the theory of polarization of the fluorescence of proteins, it is likely that some tryptophan residues of myosin, which are responsible for the increase in the fluorescence intensity and polarization when myosin interacts with substrates, reduce their local freedom of rotation.
The Raman spectra of purified myosin, C-protein and myosin-C-protein complex in aqueous solution, as well as that of pelleted filaments, were analyzed. The analyses confirmed that C-protein adopts a predominantly non-alpha-helical structure, while myosin adopts a predominantly alpha-helical structure. Neither 10 mM MgCl2 nor filament formation induced large conformational changes of purified myosin. Thermal denaturation of purified myosin, however, induced large changes in the Raman spectrum owing to conformational changes. It is likely that the binding of myosin and C-protein is not accompanied by large conformational changes of the proteins.
Hemodynamics in supine position were studied echocardiographically in 56 young patients with borderline hypertension and 56 age-matched normotensive subjects. In hypertensive patients, the cardiac index (CI) did not increase, but the total peripheral resistance (TPR) increased significantly (p less than 0.005). The hypertensive patients were classified into 2 groups, according to the level of the CI. In patients in group A ("normal" CI), the CI, heart rate and the mean circumferential fiber shortening velocity (mVCF) were normal, but the TPR was increased significantly. In patients in group B ("high" CI), the CI, heart rate and the mVCF increased significantly (hyperkinetic state), but the TPR was normal. Plasma renin activity (PRA) was significantly higher in patients in group B than the normal subjects, but the level of PRA in patients in group A was normal. These findings support the hypothesis that sympathetic nervous activity increases in patients in group B, but not in those in group A. Therefore, this study provides evidence that the TPR is abnormal in patients with borderline hypertension, and an impaired neurogenic activity seems to be important in the early stage of hypertension, as in borderline hypertension associated with a hyperkinetic circulatory state (group B).
To evaluate the therapeutic efficacy of a new calcium antagonist, nicardipine in hypertension, the cardiovascular effects of chronic therapy were determined by echocardiography in 10 patients with uncomplicated mild to moderate hypertension. Significant declines were recorded in mean blood pressure (average; 11.7 +/- 0.8%) and total peripheral resistance (22.0 +/- 2.0%), accompanied by an increase in cardiac index (13.6 +/- 2.2%). In spite of the considerable degree of blood pressure reduction, heart rate and body weight were unchanged. The magnitude of the decline in total peripheral resistance was directly correlated to both the decrease in mean blood pressure (r = 0.701, p less than 0.02) and the increase in cardiac index (r = -0.953, p less than 0.01). These findings suggest that the hypotensive effect of this new calcium antagonist is due to peripheral vasodilation, and it appears to represent an effective adjunct for the treatment of mild to moderate hypertension.
