[Second-look operations in malignant ovarian cancers].

We have been performing second look operations as a measure to improve the results of ovarian cancer therapy. Remarkable advances have been made recently in anticancer drugs; we have been using such agents in combination with the present method not only after conventional trial laparotomies, but also wherever possible in cases having undergone one year of chemotherapy, especially cases presenting neither subjective nor objective symptoms. Of the cases where a therapeutic effect of the drugs was observed, the patient's life was prolonged whenever all residual tumors could be removed as a result of this method. As none of the tumor markers currently in use has been found to be effective in early detection, the second look operation would seem to be indicated both for early detection of reoccurrence and judgment of therapeutic results.

[Treatment of advanced ovarian cancer with cis-dichlorodiammine platinum (CDDP) as single agent or cddp in combination with adriamycin (ADM)].

Twenty-five patients with advanced ovarian cancer were treated with Cis-dichlorodiammneplatinum (CD-DP) 50 mg/m2 as single agent or CDDP 50 mg/m2 in combination with adriamycin (ADM) 50 mg/m2. Both CDDP and CDDP-ADM combination were administered intravenously. Thirteen patients received CDDP alone and twelve patients received CDDP-ADM combination. The single agent activity of CDDP produced a complete response (CR) of 23.1% and a partial response of 15.4% with an overall response rate of 38.5%. The combination of CDDP and ADM gave a response of 58.3% (CR 25.0%, PR 33.3%). Thus, the combination of CDDP and ADM produced a response rate superior to that of CDDP alone. These clinical findings were supported by our investigation of novel experimental therapies in 7, 12 dimethylbenz (a) anthracene induced rat ovarian cancer.

A case of K cell deficiency with diabetes mellitus and Graves' disease.

The following immunological functions were studied in a case of insulin dependent diabetes mellitus with Graves' disease: (1) Lymphocyte subpopulations, (2) mitogen response, (3) immunoglobulin producing cells by the plaque forming cell assay, (4) cell-mediated cytotoxicity, and (5) natural killer activity were normal. The patient lacked antibody-dependent cell-mediated cytotoxicity. No conclusion could be drawn as to whether effector cells mediating natural (NK cell) and antibody-dependent cell-mediated cytotoxicity (K cell) are identical or different. However, the existence of such a case strongly suggests that the K cell is distinct from the NK cell. To resolve the question of whether or not a loss of antibody-dependent cell-mediated cytotoxicity activity plays an important role in the development of diabetes mellitus and Graves' disease, further studies on a large number of cases are necessary.

The effect of sperm concentrations during preincubation and at insemination on fertilization of rat eggs in vitro.

Rat epididymal spermatozoa, at a concentration of 40-65 X 10(4) cells/ml, were preincubated for 5-5, 5 hr and then added to eggs from superovulated, immature rats. By 10-12 hr after insemination, 86% of the eggs had undergone fertilization, but many of them (41%) were polyspermic. When sperm were preincubated at almost the same concentration and for the time stated above, but then diluted to 1-3 X 10(4) cells/ml before insemination, the number of fertilized eggs and the incidence of polyspermy were both reduced, being 56% and 5%, respectively. When a sperm concentration of 1-3 X 10(4) cells/ml was used both during preincubation and at insemination, the fertilization rate of 60% or more, with polyspermy not in excess of 10%, a sperm concentration of 30-100 X 10(4) cells/ml during preincubation and then diluted to approximately 1-3 X 10(4) cells/ml at insemination appears to be optimal.

Effect of polychlorinated biphenyls and polychlorinated quaterphenyls in Cynomolgus monkey (Macaca fascicularis).

Female Cynomolgus monkeys (Macaca fascicularis) with P-KC-400, Y-PCB, PY-PCB or polychlorinated quaterphenyls (PCQ) received a daily dose of 5 mg for 20 weeks, and some monkeys received a daily dose of 10 mg of Y-PCB or 0.5 mg of PCQ. The chemical compositions of the polychlorobiphenyls (PCB) used for the oral administration were as follows: P-KC-400, PCB from which polychlorodibenzofurans (PCDF) have been removed from Kanecklor 400, largely contains tri- and tetrachlorobiphenyls and no PCDF. Whereas, Y-PCB and PY-PCB, PCB with constituents similar to PCB ingested by yusho patients, largely contain penta- and hexachlorobiphenyls, in addition, PCDF of 400 ppm was present only in Y-PCB, but not in PY-PCB. There were immunosuppression, enlargement and histopathological changes of the liver (such as interstitial inflammation, and proliferation of epithelial cells of biliary duct, etc.) in the groups fed P-KC-400 and PY-PCB (free of PCDF). In the group fed Y-PCB (with PCDF), there were more apparent decreases in body weight, immunosuppression, fatty liver and histopathological changes than in the groups P-KC-400 and PY-PCB. In addition, there were hair loss, acneform eruptions, edema of the eyelid, congestion and abscess of the Meibomian gland, and cornifications of the skin, characteristic dermatological findings of yusho disease.

Synthesis and spasmolytic activities of 2-(1,2-benzisoxazol-3-yl)-3-[[omega-(dialkylamino)alkoxy]phenyl]acrylonitriles.

Several 2-(1,2-benzisoxazol-3-yl)-3-[[omega-(dialkylamino)alkoxy]phenyl]acrylonitrile derivatives were synthesized and screened for potential spasmolytic activity. The effect of structural variation of these molecules on biological activities was systematically examined. Among these compounds, (Z)-2-(1,2-benzisoxazol-3-yl)-3-[2-(2-piperidinoethoxy)-phenyl]acrylonitrile (1d), (Z)-2-(1,2-benzisoxazol-3-yl)-3-[2-(2-morpholinoethoxy)phenyl]acrylonitrile (1f), and their analogues (3c,d) having a methoxy substituent at C5 of the benzoisoxazole ring showed potent antispasmodic activities in the in vitro and in vivo studies.

Physicochemical properties and states of sulfhydryl groups of uricase from Candida utilis.

Highly purified uricase [urate: oxygen oxidoreductase, EC 1.7.3.3] was obtained from Candida utilis by affinity chromatography with xanthine-agarose conjugate followed by chromatography with Sephadex G-200 in the presence of dithiothreitol. The uricase molecule had a molecular weight of 120,000 and was composed of four identical subunits with a molecular weight of 30,000. The amino acid composition was determined and the N-terminal amino acid was identified as methionine. Other physiochemical properties obtained were as follows; isoelectric point (5,6), alpha-helix content (12%; b0 = -76 +/- 15 degree), specific activity (25.8 units/mg protein) and Km value (1.0 x 10(-5) M) for uric acid at pH 8.5 and 25 degree C. The uricase preparation did not contain detectable amounts of the following metals; Fe, Cu, Mg, Mn, Co, Ni, Zn, and Cd. There were three cysteine residues in the subunit molecule and these were classified into two types by a chemical modification study; one is accessible to a reagent and the remaining two become accessible only after denaturation. The former may be located in the vicinity of the active center of the molecule, although it does not participate directly in the catalysis.
