Clinical and cytogenetic studies are reported in two infants with a stable ring G chromosome. Identification of the abnormal chromosomes was performed by the G-banding and the Q-banding methods.
The polarity of the summating potential wave induced by electrocochleographical examination has an important bearing upon the pathophysiology or the prognostic evaluation of sensorineural deafness. Electrocochleographical examination was performed on cases of sudden deafness, Ménière's disease, progressive sensorineural deafness, sensorineural deafness due to barotrauma, etc. When the polarity of the summating potential showed an increased negative wave, physiological blockage in sensorineural structures was revealed. In other words, under such conditions sensorineural structures have a potential for activity although hearing acuity was damaged. On the other hand, when the summating potential has an increased positive polarity wave, gradual decrease of the hearing acuity was revealed.

Electrocochleography was performed at first examination in 34 patients with sudden deafness. Recording was made with the use of the transtympanic needle electrode technique. A notable difference in the electrocochleographic findings was shown between the cases of complete recovery or remarkable improvement and the cases of slight improvement or no change. Electrocochleography may be prognostic of sudden deafness at initial onset.
It was confirmed that the substances injected into the facial nerve through the stylomastoid foramen, reached every part of the inner ear. In sensitized animals, by repeated provocative injections of antigen, acute reversible episodes, very similar to that of Ménière's disease, were observed. The injection of various solutions and the provocative injection into the sensitized animals, resulted in completely opposite reactions. In sensitized animals, the electrophysiological phenomena of the cochlea also showed fluctuations like Ménière's disease.
Experiments were undertaken to substantiate the hypothesis that the mechanism of the direct effect of ouabain on the renal excretion of electrolytes is the result of inhibition of the transport enzyme, (Na, K)-ATPase. In dogs hydrated with saline, an injection of 3H-ouabain into the unilateral renal artery produced a continuing marked increase in excretion of water and sodium from the kidney, but not from the counter kidney. At maximal diuresis--90 min after ouabain injection, both kidneys were removed to assay microsomal ATPase activity and determine radioactivity distributed in subcellular structures. It was demonstrated that 3H-ouabain was deposited in the microsome fraction obtained from the injected kidney in concentrations ranging from 10(-7) to 10(-6) M/kg wet weight, and (Na, K)-ATPase activity of this fraction was inhibited as compared with that of the microsomal fraction obtained from control kidneys. Since (Na, K)-ATPase activity of renal microsomes was significantly inhibited in vitro by more than 10(-7) M of ouabain, ouabain concentration in microsomes obtained from the injected kidney was considered to be sufficient to inhibit ATPase activity. These findings indicate that ouabain diuresis under the present condition is closely related to direct inhibitory effect of ouabain on (Na, K)-ATPase activity of microsomes in tubular cells.

Experiments were undertaken to substantiate the hypothesis that the mechanism of the direct effect of ouabain on the renal excretion of electrolytes is the result of inhibition of the transport enzyme, (Na+, K+)-ATPase. In dogs hydrated with saline, an injection of 3H-ouabain into the unilateral renal artery produced a continuing marked increase in excretion of water and sodium from the kidney, but not from the counter kidney. At maximal diuresis -- 90 min after ouabain injection, both kidneys were removed to assay microsomal ATPase activity and determine radioactivity distributed in subcellular structures. It was demonstrated that 3H-ouabain was deposited in the microsome fraction obtained from the injected kidney in concentrations ranging from 10(-7) to 10(-6) M/kg wet weight and (Na+, K+)-ATPase activity of this fraction was inhibited as compared with that of the microsomal fraction obtained from the counter kidney. Since (Na+, K+)-ATPase activity of renal microsomes was significantly inhibited in vitro by more than 10(-7) M of ouabain, ouabain concentration in microsomes obtained from the injected kidney was considered to be sufficient to inhibit ATPase activity. These findings indicate that ouabain diuresis under the present condition is closly related to direct inhibitory effect of ouabain on (Na+, K+)-ATPase activity of microsomes in tubular cells.
Experiments were undertaken on order to elucidate the mechanism of diuresis induced by dibutyryl cyclic AMP in dogs. Intravenous injections of dibutyryl cyclic AMP produced an increase in urine flow in dogs hydrated with saline. It was also noted that dibutyryl cyclic AMP produced an increase in myocardial contraction, renal blood flow and glomerular filtration rate; In addition, unilateral injection of dibutyryl cyclic AMP into the left renal artery produced diuresis bilaterally. Based on the above findings it was suggested that the diuretic action induced by dibutyryl cyclic AMP is associated with its hemodynamic effect.
The current status of nursery routines of prophylaxis against ophthalmia neonatorum were surveyed by mail questionnaire to 100 leading maternity hospitals. More than 20% of the respondents were not using silver nitrate, mainly because of chemical conjunctivitis. The clinical significance and incidence of chemical conjunctivitis were studied in 1,000 newborns whose eyes were handled differently. Rinsing after instillation of silver nitrate does not reduce the conjunctival irritation. Although 90% of the infants had conjunctivitis in the first hours of life, the majority cleared within 24 hours. Chemical conjunctivitis did not increase secondary infection, neither did it mask bacterial infection. Silver nitrate is effective in vitro against Neisseria gonorrhoeae and Staphylococcus aureus in a concentration of 0.1% and against Escherichia coli in a concentration of 0.01%.
