Among the lymphograms of 25 F.U.O. patients whose diagnoses were unknown after several weeks of detailed examinations, abnormalities were noted in 4. Of the 4 abnormal cases, 2 were finally diagnosed by superficial lymph nodes biopsies as having malignant lymphomas. The other 2 had no final diagnoses; though biopsies of enlarged superficial lymph nodes were performed, they showed only reactive hyperplasia. No exploratory laporatomy was performed among the 25 cases. We conclude that lymphography is not absolutely indicated in the evaluation of F.U.O.

Phenylalanine hydroxylase was purified approximately 3000-fold to apparent homogeneity with a 13% yield and crystalized from L-phenylalanine-induced cells of Chromobacterium violaceum. The enzyme was shown to be composed of a single polypeptide chain with an estimated molecular weight of approximately 32,000. Some of the physical properties of the enzyme include: a Stokes radius of 26.0 A, a sedimentation coefficient of 2.71 S, a diffusion coefficient of 8.20 X 10(-7) CM2/S, a frictional ratio of 1.23, and an isoelectric point of pH 4.5. No detectable iron was found in the purified enzyme. Apparent Km values for L-phenylalanine and 2-amino-4-hydroxy-6,7-dimethyltetrahydropteridine were 140 and 54 muM, respectively.
Leupeptin (acyl peptidyl-L-argininal) is a potent inhibitor of trypsin and related proteases. We analyzed the association of leupeptim with bovine trypsin kinetically, assuming that it proceeds by a pathway which involves two steps: E + I in equilibrium K1 Complex I k-2 in equilibrium k+2 Complex II. The observed dissociation constant (K1) for the first step was 1.24 X 10(-3) M (at pH 8.2 15 degrees C) and the two first-order rate constants (k+2 and k-2) were 166 s-1 and 1.75 X 10(-3.s-1, respectively (at pH 8.2, 15 degrees C). The dissociation constant (Kd) for the whole process was calculated from these parameters to be 1.34 X 10(-8) M. This value is compatible with that determined directly by an independent static method (2.36 X 10(-8) M). We also measured Kd for the leupeptine complex of anhydrotrypsin, a trypsin derivative in which the active-site hydroxyl group is missing. The observed value was about 5 orders of magnitude larger than Kd and was rather similar to K1 in native trypsin. A elupeptin isomer which contains a D-argininal residue did not show strong affinity towards trypsin. These findings suggest that complex II consists of a covalent hemiacetal adduct formed between the serine hydroxyl group in the enzyme active site and the aldehyde group in the inhibitor. The pH dependencies of the dissociation constant and other parameters show that deprotonation of the charge-relay sustem in the active site is important for the formation and stabilization of complex II.
An asymptomatic, posterior mediastinal mass first appeared in a routine chest X-ray film of a 43-year-old woman. Computed tomography (CT) permitted the diagnosis of intrathoracic kidney to be advanced. Intravenous urography confirmed the CT diagnosis.

The nature of the oxygenated intermediate observed (Fujisawa, H., Hiromi, K., Uyeda, M., Okuno, S., Nozaki, M. and Hayaishi, O. (1972) J. Biol. Chem. 247, 4422--4428) during the reaction of protocatechuate 3,4-dioxygenase (protocatechuate:oxygen 3,4-oxidoreductase (decyclizing), EC 1.13.11.3) was investigated. 3,4-Dihydroxyphenylpropionic acid and 3,4-dihydroxyphenylacetic acid were used as substrates of the enzyme to slow down the rate of the reaction. The enzyme reactions were performed under conditions where the concentration of the organic substrate was lower than those of the enzyme and oxygen in the reaction mixture. The reactions were stopped before completion by the addition of hydrochloric acid or guanidine hydrochloride and then the organic compounds were extracted from the reaction mixture to be analyzed. The qualitative analyses by thin-layer chromatography revealed that there was no species other than the organic substrate and the enzymatic reaction end-product during reaction. The quantitative spectrophotometric analyses revealed that the organic substrate which had participated in the formation of the oxygenated intermediate existed as a species indistinguishable from the reaction end-product, indicating that the oxygenated intermediate was not a simple complex of oxygen, substrate and the enzyme, i.e., a ternary complex, but a species rather close to a binary complex of product and the enzyme.
