Domains that confer intracellular sequestration of the Glut4 glucose transporter in Xenopus oocytes.

The Glut4 glucose transporter is poorly functional compared with other glucose transporter isoforms when expressed in Xenopus oocytes. To investigate the molecular basis for this poor functionality, we compared the biosynthesis and targeting of Glut1 and Glut4 in oocytes after microinjection of the corresponding mRNAs. Both Glut1 and Glut4 were present as lower molecular weight endoglycosidase H-sensitive and higher molecular weight endoglycosidase H-resistant. Subcellular fractionation indicated that Glut1 was targeted to the plasma membrane with a 6.6-fold greater efficiency than was Glut4. Confocal immunofluorescence microscopy confirmed the relative enrichment of Glut1 in the plasma membrane and the efficient intracellular sequestration of Glut4. As in mammalian cells, the endoglycosidase H-resistant form of Glut4 was concentrated in low-density intracellular vesicles, whereas Glut1 was distributed in intracellular vesicles of higher average density. The structural basis for the differential localization of Glut1 and Glut4 was investigated by determining the plasma membrane content of a series of chimeric Glut1/Glut4 molecules. These data indicated that two distinct regions of Glut4, encompassing residues 24-132 and the COOH-terminal cytoplasmic tail, confer intracellular sequestration on the chimeric transporter molecules. At least part of the sequestration effect of the more N-terminal domain was due to the incomplete maturation of chimeras containing this region, resulting in the accumulation of lower molecular weight endoglycosidase H-sensitive and endoglycosidase H-resistant forms, whereas the COOH-terminal cytoplasmic tail conferred sequestration of fully glycosylated chimeras in a low-density intracellular membrane compartment.

Intracranial extraskeletal myxoid chondrosarcoma with special reference to lamellar inclusions in the rough endoplasmic reticulum.

An extraskeletal myxoid chondrosarcoma in the pineal region was studied by light and electron microscopy. The immunohistochemical positivity for S-100 protein, vimentin and collagen type II favored a chondrocytic origin of the tumor. In addition to the well-described ultrastructural features suggestive of a cartilagenous nature, this tumor had unusual lamellar inclusions in the rough endoplasmic reticulum. To the best of our knowledge, this is the first report of these special inclusion bodies in a myxoid chondrosarcoma.

Initial flow of tissue conditioners--influence of composition and structure on gelation.

The effects of the molecular weight of polymer powders, the ethyl alcohol content of the liquids, the type of plasticizer and the polymer powder/liquid ratio on viscoelastic properties during gelation of tissue conditioners were studied with an oscillating rheometer. The results showed that: (i) The gelation time decreased exponentially with increases in molecular weight of the polymer powder and with powder/liquid ratio. There were linear relationships between the log of gelation time, and both the molecular weight of the polymer powder and the powder/liquid ratio. (ii) In general, gelation time decreased exponentially with increases in ethyl alcohol content. However, the relationship between the log of the gelation time and ethyl alcohol content fitted a second-order polynomial. (iii) The type of plasticizer affected gelation time. The order of gelation times was: benzyl benzoate < dibutyl phthalate < butyl phthalyl butyl glycolate. The initial flow of tissue conditioners could be controlled over a wide range by varying the molecular weight of the polymer powders, the ethyl alcohol content, the powder/liquid ratio and the type of plasticizer.

Phytochemical studies of seeds of medicinal plants. III. Ursolic acid and oleanolic acid glycosides from seeds of Patrinia scabiosaefolia Fischer.

Three isomeric pairs of ursolic acid (1, 3, and 5) and oleanolic acid (2, 4, and 6) glycosides were isolated as predominant constituents from seeds of Patrinia scabiosaefolia Fischer (Valerianaceae). Based on chemical and spectral evidence, their structures were established to be 3-O-[alpha-L-rhamnopyranosyl-(1-->2)-alpha-L-arabinopyranosyl] ursolic acid (1) and oleanolic acid (2), 3-O-[beta-D-glucopyranosyl-(1-->3)-alpha-L-arabinopyranosyl] ursolic acid (3) and oleanolic acid (4), and 3-O-[alpha-L-rhamnopyranosyl-(1-->2)-[beta-D-glucopyranosyl- (1-->3)]-alpha-L-arabinopyranosyl] ursolic acid (5) and oleanolic acid (6), respectively. Glycosides 1, 5, and 6 are new compounds and named as patrinia-glycosides A-I, B-I, and B-II, respectively. Glycoside 3 is a known but is the first naturally occurring product. Ursolic acid glycosides were first found from this plant specimen.

Postnatal development and sex-related difference of metyrapone reductase activities in liver microsomes and cytosol of the rat.

A significant difference was observed between developmental patterns of metyrapone reductase activities in liver microsomes of male and female rats. The activity in liver microsomes of male rats was maintained at high levels until ten weeks of age after birth. In contrast, the activity in liver microsomes of female rats was markedly decreased until six weeks of age and reached low levels after eight weeks of age. As with the results of such different patterns, there were pronounced sex-related differences during adult period. Castration significantly decreased the activity in liver microsomes of male rats. This finding suggests that androgens have a role in sex-related differences of the activities in liver microsomes of male and female rats. Furthermore, sex-related differences of the activities were examined in microsomes and cytosol from the liver and kidney of the rat, mouse, hamster, guinea pig, and rabbit. All animal species tested showed sex-related differences in their activities. The rat and rabbit were found to resemble the guinea pig and hamster, respectively, in the occurrence of sex-related difference.

[Long-term results of isolated mitral valve replacement using bioprosthesis].

We evaluated late clinical results of 134 hospital survivors, of whom 152 patients underwent isolated mitral valve replacement with bioprosthesis from September, 1976 to July, 1989 for acquired valvular heart diseases. One hundred fifty-two bioprosthesis consisted of Hancock porcine aortic, 44, Carpentier-Edwards porcine aortic, 28 Ionescu-Shiley pericardial 46 and Carpentier-Edwards pericardial 34. The cumulative follow-up period was 1151.0 patient.year (pt.yr). The actuarial survival rate with Kaplan-Meier method was 82.4%, 79.9%, 76.0% at 5, 7 and 10 years respectively. The actuarial event free rate of primary tissue failure was 96.7%, 82.8% and 56.7% at 5, 7 and 10 years respectively. The linealized rate of bioprosthetic valve endocarditis was 0.3%/Pt.yr. Although 104 patients (77.6%) of 134 received postoperative anticoagulant therapy with Warfarin sodium about for one year. Thromboembolic complications occurred at rate of 1.3%/Pt.yr. The bioprosthesis in the mitral position is now used only in the selected patients because of the limited long term durability and necessity of permanent anticoagulant therapy.

Sex-related differences of acetohexamide reductase activities in the liver and kidney of various mammalian species.

Sex-related differences of acetohexamide reductase activities in cytosol and microsomes from the liver and kidney of the rat, mouse, guinea pig, hamster and rabbit were examined. Among animal species tested, the rat, guinea pig and rabbit exhibited sex-related differences in the activities. The activities in liver cytosol of the rat and rabbit were higher in the females than in the males, whereas the activity in kidney cytosol of the rabbit was higher in the male than in the female. Furthermore, the activities in liver and kidney microsomes of the rat and that in kidney microsomes of the guinea pig were much higher in the males than in the females. It is interesting to note that significant sex-related differences of acetohexamide reductase activities are found in some animal species other than the rat.

[ATP radionuclide ventriculography; its usefulness for the evaluation of ischemic heart disease].

To evaluate the usefulness for the detection of ischemic heart disease (IHD), ATP first pass radionuclide ventriculography (RVG) was performed in 10 patients. Coronary angiography was performed in all patients. Among these patients, 2 had single vessel disease, 2 had double vessel disease, 3 had triple vessel disease, and 3 had normal coronary arteries. RVG was performed at rest, then, ATP was infused at the rate of 0.16 mg/kg/min and ATP-RVG was done 4 minutes later. The left ventricular ejection fraction (LVEF) was calculated, and the regional wall motion (RWM) was graded as normal, hypokinesis, akinesis, and dyskinesis. In the patients with coronary artery disease, RWM decreased in 5 of 7 patients and LVEF decreased in 6 of 7 patients by ATP. However, RWM and LVEF did not decrease in the patients with normal coronary arteries. These results suggested that ATP-RVG might provide useful information for the evaluation of IHD.

[Surgical treatment of the primary cardiac rhabdomyoma in the neonate--a case report].

A successful surgical treatment of intracardiac rhabdomyoma in a 12-day-old female is reported. The neonate was first noted to have a cardiac murmur and she was diagnosed cardiac tumor by two dimensional echocardiography. It showed multiple cardiac tumors which occupied the right ventricular outflow tract and interventricular septum. Under the routine cardiopulmonary bypass the tumor of the right ventricular outflow tract was removed. Histological examination revealed a rhabdomyoma. Follow-up evaluation 7 years after operation showed normal growth and development of the patient. A repeated two dimensional echocardiogram revealed no sign of recurrence and the left ventricular cavity was not obstructed by the residual tumor in the interventricular septum. We reviewed 10 cases treated surgically in infancy including three neonate in Japan.

[Clinical use of an expanded polytetrafluoroethylene sheets (GORE-TEX SHEETS) as a pericardial substitute after aorto coronary bypass grafting].

Reoperation after cardiovascular surgical procedures are often accompanied by the possibility of injuring the heart the great vessels, and extracardiac grafts during resternotomy. To reduce the risk associated with resternotomy and to facilitate dissection of intrapericardial adhesion, we have used GORE-TEX SHEET as pericardial substitute since January, 1989. In case GORE-TEX SHEET are implanted in the mediastinum, prolongation of the foreign body reaction, mediastinitis, increasing postoperative hemorrhage should be in mind. In this study, the biocompatibility of GORE-TEX SHEET was examined clinically. The following postoperative indices such as duration of fever, change of the WBC counts, WBC fraction, CRP, mediastinal hemorrhage are evaluated. These results showed no statistical significance between GORE-TEX SHEET group and autologous pericardial group by the randomized study. We recommend use of GORE-TEX SHEET as a pericardial substitute in cases of major cardiovascular operation using median sternotomy.

[Surgical treatment of membranous ventricular septal aneurysm with posterior ventricular septal defect--a case report].

A 45-year-old woman was hospitalized for examination of a cardiac murmur. She had undergone surgery for a membranous ventricular septal defect at the age of 22. Cardiac catheterization showed a membranous ventricular septal aneurysm and a residual shunt. At reoperation, we found the membranous septal aneurysm to be accompanied with a posterior ventricular septal defect. The septal defect was closed with a Dacron patch, and the septal aneurysm was excised, with direct closure. Membranous ventricular septal aneurysm is generally associated with membranous ventricular septal defect. Cases of membranous ventricular septal aneurysm with posterior ventricular septal defect are rare.

Shrinkage of tissue conditioners with time--effect of the particle size in powder and the EtOH content in liquid.

The dimensional change of tissue conditioners with time was measured until 4 weeks after mixing. Shrinkage of the materials was evaluated in relation to particle size in powder and the EtOH content in liquid. The dissolution of EtOH and plasticizer were also measured to investigate the relation to dimensional change. Shrinkage with time was recorded. The smaller the particle size in powder and the more EtOH in liquid, the greater the shrinkage rate. The smaller the particle size in the powder and the more EtOH contained in the liquid, the greater the dissolution of EtOH. The dissolution of BPBG was below 1/100 comparing that of EtOH and no difference was recognized by particle size and EtOH content. On the basis of these results it is suggested that the dissolution of EtOH is related to shrinkage of tissue conditioners with time and that the component particle size in the powder and EtOH content in the liquid have a significant influence on dissolution of EtOH associated with shrinkage.

Genetic evidence for an activator required for induction of pectin lyase in Erwinia carotovora subsp. carotovora by DNA-damaging agents.

In Erwinia carotovora subsp. carotovora 71, the induction of pectin lyase (Pnl), the bacteriocin carotovoricin (Ctv), and cellular lysis (Lss) requires a RecA function. We obtained mutants wherein a pleiotropic defect, i.e., the lack of induction with mitomycin C, is not restored by the recA+ DNA. From a genomic library of strain 71, a cosmid (pAKC280) that restored induction of Pnl, Ctv, and Lss by mitomycin C was isolated. The activator function, designated Rdg for regulator of damage-inducible genes, was localized by subcloning and insertional mutagenesis to a 2.6-kb region within a 6.7-kb EcoRI fragment. An rdg-lacZ operon fusion was inducible by mitomycin C in RecA+ but not RecA- derivatives of E. carotovora subsp. carotovora 71 and Escherichia coli. A RecA+ E. coli strain carrying only a PnlA+ plasmid was not inducible for Pnl production; however, when both a PnlA+ plasmid and a Rdg+ plasmid were present, the transcription of pnlA and the production of the enzyme were activated by mitomycin C. The size of the pnlA transcript produced in E. coli was identical to that of the transcript produced by E. carotovora subsp. carotovora 71, suggesting that the same promoter and termination sequences were being utilized in these bacteria.

Uncoupling action of antibiotic sporaviridins with rat-liver mitochondria.

The effects of the glycoside antibiotic sporaviridins (SVDs) on oxidative phosphorylation of rat-liver mitochondria were examined. SVDs released state 4 respiration, dissipated transmembrane electrical potential, and accelerated ATPase activity. These facts demonstrated that SVDs are potent uncouplers of oxidative phosphorylation. During the uncoupling caused by SVDs, large amplitude swelling and oxidation of intramitochondrial NAD(P)H occurred, suggesting that SVDs greatly enhanced nonspecific permeability of the inner mitochondrial membrane. It is suggested that the uncoupling action of SVDs might be caused by dissipation of proton electrochemical potential due to an increase in the permeability of inner mitochondrial membrane.
