The influence of Ca-antagonists and antispasmodic drugs on contractions by ACh and by DMPP were investigated. Verapamil (10(-5) to 10(-4) M), diltiazem (10(-5) to 10(-4) M) and D-600 (10(-5) to 10(-4) M) depressed both the contractions in a dose dependent manner. Papaverine (10(-5) to 10(-4) M) and Aspaminol (10(-6) to 10(-5) M) also depressed both the contractions in a dose dependent manner. These findings indicate that Ca2+, which initiates the contraction by ACh, is supplied from both the external medium and intracellular store sites.
The effect of some reductones such as ascorbic acid (AsA), triose reductone (TR), epinephrine (Ep) and their derivatives on cyclic 3', 5'-adenosine monophosphate phosphodiesterase (cAMP PDE) was studied in the presence or absence of Cu2+. AsA, TR, Ep and the reductones related to them inhibited cAMP PDE activity. Among the reductones, TR showed the highest inhibition. AsA, 5-methyl-3,4-dihydroxytetrone, pyrocatechol, p-hydroxyquinone and resorcinol had a relatively high inhibiting activity. The type of inhibition of AsA, TR and Ep was uncompetitive, competitive and noncompetitive, respectively. Cu2+ enhanced the inhibitory action of the reductones markedly and altered the type of inhibition of the reductones.
A method was established for quantitative determination of the composition and the amounts of cytochromes in the aerobic respiratory chain of Escherichia coli. Cytochromes were solubilized with 3%(W/V) Sarkosyl and the extract was analyzed by high-performance liquid chromatography on a gel permeation column of TSK gel-G3000SW. This analytical system required only about 0.5 mg of membrane protein and 20 min per assay. The effects of the gene dosage of F-prime factors and plasmids on cytochromes were analyzed with this system, and the system was applied to the analysis of mitochondrial cytochromes.
The patient was a 31-year-old female who complained of headache and gait disturbance and admitted to our hospital on January 14, 1983. Familiar history revealed that her mother died of hemorrhage from hemangioblastoma in area postrema. Neurological examination revealed bilateral papilledema, right cerebellar sign and ataxia. Other neurological and clinical examinations were normal. CT-scan showed a right cystic cerebellar lesion and a right intraventricular lesion suggestive of hemangioblastomas. Right vertebral angiography demonstrated two vascular tumors lying in the right cerebellar hemisphere and right trigone of the lateral ventricle. Operation was performed on January 28, 1983. The histological diagnosis were hemangioblastomas.
A 23-year-old woman was admitted to our hospital because of a soft, painful mass in the right parietal region for a month. Neurological examination revealed no abnormality. Laboratory data including serum Ca, P and alkaline phosphatase were normal. Skull x-ray film showed a partially osteolytic, not well circumscribed lesion in the right frontal, parietal and occipital bone. No remarkable sclerotic area was seen in the lesion. CT scan showed that the lesion was located in the diploë, destroying both inner and outer tables of the bone. Right external carotid angiogram revealed that the lesion is supplied by the branches of superficial temporal artery and middle meningeal artery. 99mTc bone scan showed increased uptake in the lesion. The patient underwent a right parietal craniectomy. The skull bone was invaded by the tumor, which was yellow-brown in color, thickened and fragile. We could easily perforate it through. The tumor didn't invade the dura mater or periosteum. It was extensively removed with an airtome and rongeurs. A cranioplasty was carried out using a plastic resin. The pathological specimen showed that the tumor was mainly composed of mature, regularly-aligned bone and intermingled fibrous tissue. Neither mitosis nor atypical cellular features was seen. These findings were compatible with the diagnosis of ossifying fibroma. The postoperative course was uneventful and the patient was discharged in a good condition. There was no evidence of local recurrence 10 months later. Ossifying fibroma is a rare, benign bone tumor that mainly involves the craniofacial bone. The differential diagnosis of ossifying fibroma versus fibrous dysplasia is difficult. These diseases can be differentiated on combined clinical, radiological and morphological grounds.

Blood flow in contracting arterioles with non-circular cross-sections due to endothelial protrusions is investigated. The flow field was determined for a viscous Newtonian fluid with a numerical computation from electron micrographs of arteriolar crossections in skeletal muscle at different stages of contraction. The results indicate that the flow field in such vessels deviates from Poiseuille flow for circular tubes of equal cross-sectional area in several respects: the resistance is elevated, the ratio of maximum and mean velocity is increased above the value of 2 for Poiseuille flow, and the wall shear stress is nonuniform with a maximum at the tip of local endothelial projections and near zero at endothelial cell junctions. The average wall shear stress is less than in circular vessels with equal lumen crossection and pressure gradient.
We constructed a full-length cDNA coding for 3-methylcholanthrene-inducible rat liver cytochrome P-450MC by the method of Okayama and Berg. The isolated clone pAU157 contained the cDNA insert of 2.7 kb in length. Sequence analysis of the cDNA insert revealed that the amino acid sequence of cytochrome P-450MC was composed of 523 amino acid residues, including the initial 22 N-terminal amino acids whose sequence was determined with the purified protein. The primary structure was found to contain two highly conserved regions as pointed out from comparisons of the reported amino acid sequences of cytochrome P-450 species. The predicted molecular weight of the apoprotein was 59,300 daltons. Therefore, we concluded that the amino acid sequence determined here is for cytochrome P-450MC, probably corresponding to cytochrome P-450c.
A 37 kb fragment of DNA from an F-prime factor, F100-12, which showed a gene dosage effect on b-type cytochromes, was cloned with a cosmid vector, pHC79. Gel filtration of cytochromes and product analysis of the hybrid plasmids indicated that this fragment contained cybB, the structural gene for cytochrome b561. A chromosomal DNA fragment carrying the cybB gene was cloned by the plaque hybridization technique with Charon 4A as a vector. The gene was subcloned into pBR322 and was located in a 1.3 kb DNA fragment. It was concluded that the cybB gene is located on the chromosome of Escherichia coli K12.
The amounts of cytochrome b556 in the cytoplasmic membranes of several Escherichia coli K12 strains having F-prime factors and a lambda transducing phage were determined. The amount was amplified about two-fold in strains having F100-12 and F152, but not in strains having F100-11, F8 and lambda psu+2glnS+. The strain TK3D11, which lacks the kdp-gltA region (deletion D-01) of the E. coli chromosome, did not synthesize cytochrome b556 at all. From these results, the gene cybA encoding cytochrome b556 was located in the kdp-gltA region. In the cytochrome b556-deficient mutant, a novel b type cytochrome, cytochrome b561 which is a product of the gene cybB, was identified. It seems to function as a physiological electron transferring cytochrome in place of cytochrome b556 in this mutant.
The distribution of TRH receptors in the membrane fraction of the CNS in ataxic mutant mice (C3Hf/Nem-rol and C57BL/6j-tg) was studied. TRH binding sites in cerebellum and frontal lobe of the ataxic form and the non-ataxic heterozygotes of Rolling Mouse Nagoya were decreased in comparison with the controls, whereas those in the spinal cord of Rolling Mouse Nagoya and cerebellum of Tottering Mouse were increased in the ataxic mice over the controls. Kinetic studies were performed on cerebrum and cerebellum of the different ataxic mutant mice. Such species differences in the distribution of the TRH receptors have to be considered in the action of TRH in individual ataxia cases.
In an attempt to elucidate molecular pathogenesis of ataxia without cytological abberations in the cerebellum, Rolling Mouse Nagoya (C3Hf/Nem-rol) was used to study distribution of GABA receptors in membrane fractions. Among muscimol binding sites of various regions in the ataxic CNS, those in pons and medulla were significantly decreased (P less than 0.001) compared with control and non-ataxic heterozygote CNS, followed by cerebellum at a lower degree of significance (P less than 0.01). The kinetic studies demonstrated that dissociation constants of high- and low-affinity binding sites of muscimol of each control and those of ataxic mutant mouse were similar, i.e., KH = 41 nM and KL = 1.1 microM, respectively. GAD in the various regions was assayed, and it showed higher activity in the thalamus and hypothalamus, and lower activity in the cerebellum, of the ataxic mutant mouse as compared to that of the control mouse.
