Detection of lung cancer in clinical specimens using a human monoclonal antibody HB4C5-clone 3.

Biopsies of 18 patients with lung cancer and cellular specimens of 21 lung cancer patients were analyzed with human monoclonal antibody HB4C5-clone 3 using avidin-biotin-peroxidase techniques. Analyses with the biopsies showed that HB4C5-clone 3 reacted with 16 of 18 biopsy specimens at a high rate of about 90% and reacted with all specimens of five adenocarcinoma tissues, five of six squamous cell carcinomas, two of three large cell carcinomas, both specimens of small cell carcinomas, and both of the other types of carcinomas. In all the reactive cases, the monoclonal antibody was reactive with greater than 50% of cancer cells. With cellular specimens of lung cancer patients, HB4C5-clone 3 reacted with 7 of 13 adenocarcinoma specimens, two of six squamous cell carcinomas, and one of two small cell carcinomas. The immunoreaction with HB4C5-clone 3 was observed in sites of the cytoplasm of cancer cells. From these data, HB4C5-clone 3 is considered to be of potential use in diagnoses of tissues and cellular specimens of lung cancer.

Rapid and sensitive chemiluminescent enzyme immunoassay for measuring tumor markers.

We developed a chemiluminescent enzyme immunoassay (CLEIA) to quantify such tumor markers as carcinoembryonic antigen (CEA), alpha-fetoprotein (AFP), CA19-9, and CA125. We used a novel chemiluminescent substrate, a derivative of 1,2-dioxetane phosphate (AMPPD), to measure alkaline phosphatase as a labeling enzyme to Fab' fragments of antibody. Regardless of the solid phase, i.e., polystyrene beads (6 mm diameter) or ferrite-coated particles (0.3 microns diameter), the standard curves within the dynamic ranges of the conventional RIA or enzyme immunoassay (EIA) were linear in all cases except for those with AFP. Use of the ferrite particles further shortens the immunoreaction, so the assay can be performed in 30 min. In addition, the relationships between concentrations of the marker and chemiluminescent signals for CA19-9, CA125, and CEA were linear up to concentrations about 10-fold greater than the ordinary dynamic ranges. Intra- and interassay CVs (averages for individual analyte) were 2.2%-4.9% and 2.0%-5.8%, respectively. In an analysis of serum samples, results of the CLEIA correlated reasonably well with those of RIA or EIA. The lower limit of detection by CLEIA with ferrite particles was 390 arb. units/L for CA19-9, 990 arb. units/L for CA125, 0.06 micrograms/L for CEA, and 0.03 micrograms/L for AFP. Thus, the sensitivity increased to between two- and 10-fold that of RIA or colorimetric EIA, depending on the analytes.

Three cases of multiple myeloma developing into melphalan-related chronic myelomonocytic leukemia.

From 1980 to 1987, three cases of chronic myelomonocytic leukemia (CMML) were encountered among 68 cases of multiple myeloma who survived more than three years from the diagnosis. The incidence (4.8%) of secondary myelodysplastic syndrome (MDS) is almost identical to previous reports, but case reports of chronic myelomonocytic leukemia were rare. In Japan, there are few reports of multiple myeloma patients who later developed secondary MDS or acute myelogenous leukemia (AML). In our cases, none of the 31 patients treated with cyclophosphamide developed secondary MDS, while three of 37 patients treated with melphalan developed CMML. This difference is not statistically significant.

A patient with primary plasma cell leukemia accompanied by an extensive polypoid infiltration of the gastrointestinal tract.

A 37-year-old male presented with primary plasma cell leukemia (PCL) with kappa-type Bence Jones proteinuria (BJP) and polyposis of the stomach and colon. His plasma cell leukemia was not preceded by a pre-existing multiple myeloma, and presented the complication of polyposis in both stomach and colon. Biopsy of mucosal lesions revealed marked accumulation of atypical plasma cells with positive cytoplasmic kappa-chain. No amyloidosis was present. His disease responded remarkably well to intermittent melphalan and prednisolone. This case represents an uncommon combination of primary plasma cell leukemia and polypoid gastrointestinal lesions with plasma cell infiltration.

T-zone lymphoma in association with systemic lupus erythematosus.

Two patients with systemic lupus erythematosus (SLE) and T-zone lymphoma are described. On admission, both showed arthralgia, generalized lymphadenopathy, hypergammaglobulinemia and positive antinuclear antibody. Lymph node biopsies revealed diffuse infiltration of atypical T-lymphocytes in the expanded interfollicular area (T-zone), a finding characteristic for the T-zone lymphoma. Renal biopsy showed lupus nephritis and neoplastic lymphoid cell infiltration in the glomeruli of one patient, but only diffuse infiltration of neoplastic lymphoid cells and mature plasma cells were observed in the interstitium of the other patient. Both patients responded remarkably well to prednisolone (1 mg/kg/day).

[Treatment of 61 patients with acute myelogenous leukemia at first relapse].

Treatment results in the 61 adult patients with AML in first relapse were analyzed to establish a better strategy for this group of patients. These patients received reinduction chemotherapy during 1979-1988. Complete remission (CR) was obtained in 57.4% of the cases, and the probability of survival and remaining in CR at five years was 11.9% and 17.9%, respectively. The longer duration of initial remission was favorable factor for achieving second CR. Type of reinduction regimens which were different from those used in the initial induction phase did not influence the second CR rate. The use of different consolidation regimen appeared to favorably affect the survival and probability of remaining in CR.

Generation of hybrid hybridomas secreting human IgM class hybrid antiricin and antidiphtheria toxin antibodies.

Two lines of mouse-human hybridomas were fused to produce human bifunctional antibodies. A hybridoma that secretes human IgM (kappa) class antiricin monoclonal antibody was treated with actinomycin D to inhibit cell growth. Another cell line, secreting human IgM (lambda) class antidiphtheria toxin monoclonal antibody, was 6-thioguanine resistant (HAT sensitive). Six hybridomas secreting IgM with both kappa and lambda light chains were obtained. Two of them secreted antibodies with antiricin specificity (kappa and lambda light chains) and antidiphtheria toxin specificity (kappa and lambda light chains). Antidiphtheria toxin antibodies secreted from other hybrid hybridomas did not contain kappa light chain, but lambda chain. A hybrid hybridoma clone 5-29 secreted complete hybrid IgM with lowered affinity to diphtheria toxin. These results suggested unbalanced recombination of immunoglobulin polypeptide chains in hybrid hybridomas.

Histone H2B as an antigen recognized by lung cancer-specific human monoclonal antibody HB4C5.

Histone H2B was demonstrated to be an immunoreactive material recognized by the human monoclonal antibody HB4C5, which had been already established to be specific for lung cancers. The inhibitory effect of histone H2B on the activity of HB4C5 antibody to immunostain the cytoplasmic antigen in lung adenocarcinoma tissue indicated that histone H2B at least had antigenic determinant comparable to the cytoplasmic antigen. A mouse anti-histone H2B monoclonal antibody could immunostain the cytoplasm of lung adenocarcinoma cells in sliced tissue sections in the same manner as the human monoclonal antibody HB4C5. Enzyme-linked immunosorbent assay of HB4C5 antibody on plastic immunoplates coated with histone H2B also showed specific reactivity of this antibody with histone H2B, and the reaction was effectively inhibited when extra histone H2B or mouse anti-histone H2B monoclonal antibody was added to the reaction mixture. These results consistently lead us to a conclusion that histone H2B possesses antigenicity to the human monoclonal antibody HB4C5.

Characterization of a lung cancer-associated human monoclonal antibody HB4C5.

A human monoclonal antibody HB4C5, which is known to be associated with lung cancers, was characterized. Two kinds of light chains with 30-kD and 32-kD molecular mass were found to comprise the molecule of this IgM antibody. Various combinations of the two light chains could be contained in this IgM antibody molecule, which might have resulted in the heterogeneity of this antibody with different reactivities. The presence of the 30-kD light chain was indispensable for the antibody activity of cancer recognition. Monomeric 180-kD units of the antibody, having 32-kD light chains exclusively, which showed marginal activity of cancer recognition, could be removed by immunosorbent column chromatography with histone H2B-Sepharose 4B. F(ab')2 of the HB4C5 antibody containing the active 30-kD component as the dominant light-chain constituent could be purified by hydroxyapatite chromatography by HPLC, facilitating clinical applications of this lung cancer-associated monoclonal antibody of human origin.

[A study of the adhesive resins used in adhesion bridges].

UNLABELLED: We suspected that the composition of stiffened adhesive resins may cause adhesion bridges to drop out. In this study, by using a scanning type electron microscope, we observed the breaking sides of the adhesive resin after the tension test. We also examined the surfaces of the resin that we cut and ground after it had hardened, and the surfaces of the adhesion bridges that dropped out. RESULTS: 1) RESULTS of the tension test. No significant differences between the adhesive resins were observed. We noticed no significant differences between the metal surface treatment methods. 2) Observations of the breaking side after the tension test. When we examined Panavia EX, we observed flaking of the metallic side and the tooth enamel. We noticed that the surface was smooth regardless of the method of preprocessing the metal. But the surface of the adhesive resin was bumpy, and we found bubbles of different sizes on the resin surfaces. When we examined Super-Bond C&B, we observed flaking of the metallic side and the tooth enamel, and the surfaces were smooth regardless of the method of preprocessing the metal. Unlike Panavia EX, however, we found various hollows, big and small, like the bubbles on the smooth surfaces of Super-Bond C&B. 3) Observations of the abrasive surface of stiffened adhesive resins. With an ambient temperature of 15 and 28 degrees, we found many minute hollows like bubbles on the abrasive surfaces of Panavia EX and Super-Bond C&B. 4) Observations of the surface of the bonding materials of the adhesion bridges which dropped out. We found bumpy surfaces on the adhesive resin which remained on the metallic side of the adhesion bridges, whichever position the adhesion bridges dropped out of.

Identification of a receptor for protein import into mitochondria.

Anti-idiotypic antibodies, prepared using a chemically synthesized signal peptide of a mitochondrial precursor protein, recognized a mitochondrial integral membrane protein (p32). Fab fragments derived from both anti-idiotypic antibodies and monospecific antibodies against purified p32 inhibited protein import into mitochondria. Moreover, anti-p32 antibodies specifically immunoprecipitated a precursor-p32 complex after detergent solubilization of mitochondria. Immunoelectron microscopy and subfractionation of mitochondria indicate that p32 is located in contact sites between the outer and inner mitochondrial membranes.

Isolation and characterization of the gene for a yeast mitochondrial import receptor.

We have previously identified an integral membrane protein (p32) from Saccharomyces cerevisiae as a receptor for protein import into mitochondria, and have localized it to the mitochondrial outer membrane at contact sites. Here we report isolation of the corresponding mitochondrial import receptor gene, termed MIR1. The deduced amino-acid sequence of p32 shows roughly 40% identity with proteins of bovine heart and rat liver that have been suggested to be mitochondrial phosphate carriers. Haploid cells carrying a disrupted MIR1 allele were unable to grow on a non-fermentable carbon source but grew in media containing glucose, indicating that the MIR1 protein is essential for mitochondrial function. Compared with wild type, amounts of some mitochondrial proteins were markedly reduced in cells containing a disrupted MIR1 allele, whereas levels of others were unchanged. This indicates that yeast contains more than one pathway for protein import into mitochondria.

Insulin-like growth factor I and transforming growth factor alpha as autocrine growth factors in human pancreatic cancer cell growth.

The roles of insulin-like growth factor I (IGF-I) and transforming growth factor alpha (TGF-alpha) as autocrine factors in the proliferation of MIA-PaCa 2 cells (human pancreatic cancer cells, PC cells) were investigated. Furthermore, the mechanism(s) of inhibition of PC cell growth by a phorbol ester in relation to these two kinds of growth factor was also studied. PC cells grew autonomously when Dulbecco's modified essential medium supplemented with 4% fetal calf serum was changed to serum-free medium (0.3% bovine serum albumin-Dulbecco's modified essential medium). In addition, serum-free conditioned medium from PC cells dialyzed against fresh Dulbecco's modified essential medium had a stimulatory action on the growth of the same kind of cells when compared with that induced by nonconditioned medium. These observations suggest that a factor(s) produced and released by PC cells stimulates their own growth. Analysis of conditioned medium from PC cells revealed the presence of immunoreactive (IR)-IGF-I and IR-TGF-alpha. The molecular size of IR-IGF-I was similar to that of authentic IGF-I. On the other hand, IR-TGF-alpha was present as multiple forms when analyzed using gel chromatography. Authentic IGF-I and TGF-alpha added to culture medium stimulated PC cell growth by 1.45- and 1.5-fold above control value, respectively. A monoclonal antibody to IGF-I receptor was able to inhibit PC cell growth. PC cell proliferation was markedly inhibited by 12-O-tetradecanoyl-13-acetate (greater than 0.16 nm), whereas cell growth of human fibroblasts was stimulated by it. 12-O-Tetradecanoyl-phorbol-13-acetate also reduced the binding of 125I-TGF-alpha, but not 125I-IGF-I, to PC cells. Decrease in TGF-alpha binding was mainly due to the reduced affinity of receptors to the ligand. These results suggest that IGF-I and TGF-alpha are involved in PC cell proliferation as autocrine factors. Further, the inhibition of PC cell growth by phorbol ester could be, at least partly, due to the decreased binding of TGF-alpha to the cells.

Stimulation of proliferation and immunoglobulin M production by lactoferrin in human-human and mouse-mouse hybridomas cultures in serum-free conditions.

The effects of growth factors, such as insulin, transferrin, lactoferrin, ethanolamine, and selenium, on proliferation and IgM production of human-human hybridomas HB4C5 cells in a serum-free enriched RDF (eRDF) medium were studied. Among them, lactoferrin markedly stimulated proliferation and IgM production of the cells. Another iron-binding protein, transferrin, stimulated proliferation of HB4C5 cells as well as lactoferrin, but its stimulatory effect on IgM production was negligible. The proliferation and IgM production of HB4C5 cells gave some detectable delays in conventional ITES-eRDF medium at low cell densities, but the delays were avoided by the addition of lactoferrin. However, eRDF medium supplemented with lactoferrin could not support proliferation and IgM production of the cells at high cell densities. For proliferation and IgM production of HB4C5 cells, eRDF medium supplemented with 25 micrograms/ml lactoferrin, 10 microM ethanolamine, 35 micrograms/ml transferrin, and 2.5 nM selenium (LETS-eRDF) gave the best result. Lactoferrin stimulated proliferation of human-human and mouse-mouse hybridomas producing IgG or IgM, but stimulation of Ig production was detected only in IgM producers. These results suggest that cell proliferation, IgM production, and IgG production of hybridomas are regulated by different mechanisms.

Purification and characterization of immunoglobulin production stimulating factor derived from human B lymphoblastoid HO-323 cells.

An immunoglobulin (Ig) production stimulating factor (IPSF) for hybridomas was found in spent medium of the human B lymphoblastoid cell line, HO-323. The IPSF was purified by serial use of DEAE chromatography, ultrafiltration, gel filtration and HPLC-DEAE chromatography. Purified IPSF was estimated to be a 410 k macro molecule by gel filtration, and contained three types of isomers which were separated from each other by native polyacrylamide gel electrophoresis. All of the isomers were, however, assumed to have the same protein components by SDS polyacrylamide gel electrophoresis. The IPSF was effective for human-human and mouse-mouse hybridomas producing IgM, but not for IgG producers in the experimental condition used here. Human-human hybridoma HF10B4, cultured in IPSF-containing medium, produced 20 times more IgM than in IPSF-free medium under serum-free conditions. The IPSF showed very little proliferation stimulating activity on HF10B4 cells.

Purification and some properties of proliferation suppressing factor from human lung adenocarcinoma PC-8.

Extracts by 3 M KCl from a human lung adenocarcinoma cell line, PC-8, suppressed the proliferation of phytohemagglutinin activated lymphocytes. The proliferation suppressing factor (PSF) was purified from the extracts by ammonium sulfate precipitation, gel filtration and ion exchange chromatography. The purified PSF had a molecular weight of 50 kDa by SDS-PAGE and was acid- and heat-labile. Since the PSF also suppressed the proliferation of three established human cell lines, it is unlikely that the PSF hinders the interaction between lymphocytes and mitogens.

A new human fusion partner, HK-128, for making human-human hybridomas producing monoclonal IgG antibodies.

A hypoxanthine-aminopterin-thymidine (HAT) sensitive human fusion partner cell line, HK-128 was established from a human plasmacytoma line, LICR-LON-HMy2 (HMy2). The HK-128 cells showed a 100% cloning efficiency. Fusion efficiency of HK-128 was so high that one hybridoma cell was produced by fusion of 10(5) cells of HK-128 with lymphocytes, obtained from lymph nodes of breast cancer patients. About 90% of the resulted hybridomas were IgG producers. The remainder revealed IgM producing activity, which was lost by long term culture. This result indicates that the HK-128 cell line has an advantage for making hybridoma cells producing IgG. Among ca. 7,000 hybridomas obtained by fusion of HK-128 with lymphocytes of a breast cancer patient, we could establish a hybridoma cell line which produced IgG specifically reacting to a human breast cancer cell line, MCF-7.
