Enzymatic dehydrogenation of tryptophan residues of human globins by tryptophan side chain oxidase II.

Enzymatic dehydrogenation of tryptophan residues in human alpha- and beta-globins has been demonstrated with tryptophan side chain oxidase II, a hemoprotein from Pseudomonas. In 1 M ammonium acetate, pH 4, the enzyme catalyzed the conversion of Trp-14 of alpha-globin to alpha, beta-dehydro-Trp. Under identical conditions, Trp-15 of beta-globin was converted to alpha, beta-dehydro-Trp, whereas Trp-37 was unmodified. No modification was obtained in 0.4% sodium dodecyl sulfate or 8 M urea. The dehydrogenase component of tryptophan side chain oxidase II was also active toward globins in the presence of ferricyanide, whereas tryptophan side chain oxidase I was inert. Upon analyses of the tryptic fragments of the modified beta-globin, two forms of alpha, beta-dehydro-Trp-15 distinguishable by ultraviolet absorption and fluorescence were identified. Each of them rapidly equilibrated with another form on irradiation at 300-380 nm. We tentatively assigned them to two isomers of alpha, beta-dehydro-Trp distinguishable by the configuration around its beta-carbon atom. alpha, beta-Dehydro-Trp in globins and certain polypeptides used herein showed a blue fluorescence at 420 nm. In [alpha, beta-dehydro-Trp 15]beta-globin, the fluorescence was exceptionally intense, the fluorescence yield being comparable to that of NADH. From several lines of evidence, the result was ascribed to the excitation energy transfer from Trp-37 to alpha, beta-dehydro-Trp-15 in the modified beta-globin.

[Studies on alteration in biventricular dimensions during stepped elevation in pulmonary arterial pressure with special reference to right ventricular bellows action].

A new method for inserting ultrasonic crystals into the interventricular septum (IVS) of dogs was performed to measure the right and left ventricular septum-to-free wall dimensions (RVD, LVD) in a line. The advantage of this technique is minimal damage of the pericardium and no damage of the ventricular wall. The segmental lengths in the right and left ventricles (RVSL, LVSL) were also measured. Stepped elevation of pulmonary arterial pressure induced by ligation of pulmonary arterial branches caused an increase in RVD and a decrease in LVD. IVS shifted to the left and transseptal pressure gradient was reduced. RVD and RVSL increased and percent shortening in RVD and RVSL during ejection decreased with pulmonary hypertension. Although the change in RVD and RVSL basically related each other, those changes in RVD was more evident than in RVSL. Thus, the measurement of RVD and RVSL is indispensable for investigating the right ventricular function. There was a high correlation between the stroke volume and the shortening in RVD (expressed as percent of the control value, respectively) during ejection in pulmonary hypertension (F = 13.88 greater than F 1(20)(0.005) = 9.94), therefore we speculate that right ventricular bellows action is essential in blood ejection from the right ventricle. Furthermore the stroke volume decreased as the shortening in RVSL during ejection decreased (F = 3.17 greater than F 1(20)(0.1) = 2.97), therefore shortening in RVSL is thought to be an important factor causing bellows action, however further investigation is necessary.

Effects of Q metabolites and related compounds on mitochondrial succinate and NADH oxidase systems.

The effects of Q metabolites (Q acid-I, Q acid-II) and related compounds (dihydro Q acid-I, dehydro Q acid-II, QS-n, and their esters) on mitochondrial succinate and NADH oxidase systems were investigated. The activity restoring succinate oxidation in acetone-treated beef heart mitochondria was found to decrease with descending order of carbon number (n) of the side chain of the Q metabolites; activity was restored with Q acid-I (n = 7) to one-third as much as that with Q-7 and Q-10, but Q acid-II (n = 5) did not restore any activity. Of the related compounds with a carboxyalkyl group (QS-n), QS-16-QS-18 (n = 16-18) were found to be most active, and their activities were also correlated with n. The relationship between the restoration of activity and the partition coefficient was considered. NADH oxidation in pentane-treated beef heart submitochondrial particles could be restored with esters of low molecular weight quinones to the same extent as with Q-10, but not with the metabolites.

Purification, characterization, and comparison of extracellular hemoglobins in coelomic fluid and blood vessel plasma of Travisia japonica (Polychaeta, Annelid).

1. Two kinds of extracellular gigantic hemoglobins in blood vessel plasma and in coelomic fluid of Travisia japonica (polychaeta, annelid) were purified. 2. Their properties including electron microscopic observation, gel filtration, amino acid analysis, SDS polyacrylamide gel electrophoresis, and immunological properties were studied and compared. No significant difference was found between them. 3. They had a mol. wt of ca. 3 X 10(6) and a shape consisting of 12 subunits with a mol. wt of ca. 2.4 X 10(5) arranged in two superimposed hexagonal rings with a diameter of 22-25 nm and a thickness of 15-16 nm, as for other annelid gigantic hemoglobins described in the literatures.

Vitamin D resistant hypophosphatemic osteomalacia associated with osteosarcoma of the mandible: report of a case.

A case of vitamin D resistant hypophosphatemic osteomalacia associated with osteosarcoma of the mandible is presented. The patient complained of lumbar, knee and foot pain and muscle weakness of two years' duration. Serum phosphorous was 1.0-1.6 mg/dl, tubular reabsorption of phosphorus was 47 to 58%, TmPO4/GFR was o.7-1.2 mg/dl. Aminoaciduria was noted. Bone biopsy confirmed the diagnosis of osteomalacia. He partially responded to the treatment with 1 alpha()H) D3 and sodium phosphate. After removal of sarcoma of the mandible, symptoms remitted and pertinent laboratory data became normal except serum alkaline phosphatase for more than one year without treatment. It is suggested that an impaired response of the tubule and bone to active vitamin D3, caused in some way by the osteosarcoma might be one of the causes of osteomalacia in this case.

Radiolabeled alpha-bungarotoxin derivatives: kinetic interaction with nicotinic acetylcholine receptors.

The binding interactions of purified tritiated [3H]-alpha-Bgt and monoiodinated and diiodinated derivatives of alpha-bungarotoxin with membrane-bound nicotinic acetylcholine receptors (nAChR) from Torpedo californica electroplax and rat brain have been characterized by several kinetic and equilibrium techniques. By all criteria, [3H]-alpha-Bgt and 125I-labeled monoiodinated alpha-Bgt ([125I]-alpha-Bgt) exhibited comparable specificities and affinities for nAChR. In contrast, affinity of nAChR for 125I-labeled diiodinated alpha- Bgt ([125I2]-alpha-Bgt) was reduced, and [125I2]-alpha-Bgt-nAChR complexes showed anomalous biphasic dissociation kinetics. [125I]-alpha-Bgt and [125I2]-alpha-Bgt binding was inhibited most potently by native alpha-Bgt as opposed to iodinated toxins. [3H]- alpha-Bgt was the radiotoxin most resistant to inhibitory influences. The use of well-characterized, chemically modified alpha -Bgt derivatives may identify ligand binding microheterogeneities and tissue-specific receptor subclasses.

Effects of neonatal undernutrition and cold stress on behavior and biochemical brain parameters in rats.

This study was conducted to investigate the separate and combined effects of neonatal undernutrition (U) and cold stress (S) on the behavioral and cerebral development of postweaning rats. A severe U was imposed by feeding dams a low protein diet. Postweaning all pups were fed a control diet. S consisted of daily exposure to 5 degrees for 3 minutes from day 2 to 11. Behavioral data show that U animals, stressed (S) + nonstressed (NS), exhibited a significant deficit in reversal learning of T-maze at 21 days, an enhanced passive avoidance response, but no difference in active-avoidance at 35 days when compared to controls of the same age. S had no effect on behavior development. At death (110 days), the brains were dissected into five sections and assay for acetylcholinesterase (AChE) and cholinesterase (ChE) activities. Brain weights of U animals (NS + S) were significantly lower in all sections except dorsal cortex (DC). AChE and ChE activities were significantly higher in all sections (except DC) of U animals relative to controls. S resulted in lower cerebellar weight and ChE:AChE ratios in some sections. Our results suggest a delayed behavioral maturation in U animals and an association between early postweaning behavior and brain parameters in adult rehabilitated animals.
