[Hormonal and functional changes and the quality of life in the late phase after gastric surgery for cancer].

The effects of gastrectomies and those reconstructions for gastric cancer on gut hormone release, GI tract transit time, RISA absorption test and triolein digestion and absorption test were investigated clinically. The patient's quality of life was evaluated by the reconstructive methods. In the cardiectomy type, gut hormone response was markedly increased. The transit time was significantly shortened. In the Billroth I and the interposition types, gut hormone response was slightly decreased. The transit time was normal. In the Billroth II and the Roux-en-Y types, gut hormone response was decreased. The transit time was prolonged. Serum glucose was markedly increased in the Roux-en-Y type. Absorption in protein was depressed in Billroth II and that in fat in Roux-en-Y. Quality of life was best in Billroth I, Billroth II, interposition, Roux-en-Y and cardiectomy in descending order. Postoperative changes of gut hormones and metabolism and the quality of life are influenced by the type of reconstructive procedure. For this reason, for the surgical treatment of cancer of the stomach, it would seem that partial, not total, gastrectomy is more appropriate if meeting the conditions of radical cancer treatment. Moreover, it seems to be indicated that a reconstructive procedure which allows food to pass through the duodenum is the procedure to choose.

'Superactivation' of alkaline phosphatase activity by cycloheximide in rat hepatoma cell cultures.

We have found that rat hepatoma cells (R-Y121B) retain alkaline phosphatase activity, and that this enzyme activity is increased by cycloheximide. Actinomycin D also increased the enzyme activity. This increase due to actinomycin D was partially inhibited by cycloheximide. The characteristics of alkaline phosphatase of the cells treated or untreated with cycloheximide or actinomycin D were similar to each other; they were heat labile and the enzyme reaction was strongly inhibited by L-homoarginine, but weakly by L-phenylalanine. The increase in alkaline phosphatase activity with cycloheximide has been termed a 'superactivation' of alkaline phosphatase.

[Antitumor effect of FU-O-G, new antitumor agent, following long term administration].

FU-O-G, the O-glucuronide methylester of 5-fluorouracil (5-FU), is a compound with a unique chemical structure. It is an antitumor agent of the prodrug type which exerts its activity by enzymatically liberating 5-FU in tumor tissues. It has been reported that the antitumor activity of this compound is superior to those of 5-FU and Tegafur (FT-207) in the treatment of various transplantable tumors. In this study, long-term administration of FU-O-G to mice bearing relatively slow-growing tumors such as Lewis lung carcinoma, mammary tumor FM3 A and hepatoma MH-134 was carried out. Consequently, FU-O-G was shown to be remarkably effective against those tumors, whereas 5-FU and FT-207 were hardly effective. Long-term daily administration was shown to be more effective than intermittent dosage in the treatment of Lewis lung carcinoma. In combination therapies of FU-O-G with other antitumor drugs, FU-O-G exhibited a synergistic effect against Lewis lung carcinoma when combined with Carboquone (CQ) or Nimustine hydrochloride (ACNU).

[Quantitation of left-sided valvular regurgitation by gated cardiac blood pool scan: evaluation of a new approach using the Fourier amplitude ratio].

Utilizing the Fourier amplitude ratio (FA ratio), we evaluated a new approach for quantitating valvular regurgitation by the gated cardiac blood pool scan. The subjects consisted of 36 patients with documented left ventricular regurgitation of grade I-IV (group 1), 24 patients with myocardial infarction including severe cardiac dysfunction (group 2), and 15 normal persons (group 3). From each pixel of the gated cardiac blood pool scan, a time-activity curve was obtained and the first-harmonic Fourier analysis was performed. Two functional images; namely, the phase and amplitude images, were generated. To generate the FA ratio, the region of interest was drawn around each ventricle in the amplitude image. The FA ratio was calculated using the following formula; FA ratio = left ventricular amplitude value/right ventricular amplitude value. The FA ratio was 2.66 +/- 1.75 (mean +/- S.D.) in group 1; 1.29 +/- 0.27 in group 2; and 1.11 +/- 0.19 in group 3. The FA ratio for group 1 was significantly higher than those of groups 2 and 3 (p less than 0.001). There was no significant difference between the FA ratios of groups 2 and 3. In patients with regurgitation, the FA ratio was 1.50 +/- 0.37 in patients with grade I, 1.97 +/- 0.34 in grade II, 2.73 +/- 0.95 in grade III; and 6.65 +/- 2.14 in grade IV. There was a good agreement between the FA ratios and quantitative angiographic estimates of regurgitation.(ABSTRACT TRUNCATED AT 250 WORDS)
