Fibronectin enhances the phagocytic activity of cultured rabbit keratocytes.

To understand corneal stromal wound healing, we investigated the effect of fibronectin (FN) on the phagocytic activity of rabbit corneal keratocytes. The phagocytic activity was measured under a dark-field microscope using polystyrene latex beads coated with either FN, bovine serum albumin, or both. The percentage of phagocytosing cells increased with incubation time. The phagocytic activity was significantly enhanced in a dose-dependent manner by coating beads with FN. Transmission electron microscopic observation demonstrated that latex beads were internalized into the cytoplasm and were surrounded by limiting membranes. These findings indicate that cultured keratocytes have phagocytic activity and that FN shows opsonic effects on the phagocytic activity, suggesting that FN in the injured corneal stroma may play a role in stromal wound repair by opsonic effects.

Phlebolithiasis associated with intramuscular hemangioma. X-ray diffractometric, X-ray microanalytical and scanning electron microscopic investigations.

Phlebolithiasis associated with intramuscular hemangioma is relatively rare. Combined crystallographic, microanalytical and morphological studies have not been done before. We investigated a case of phlebolithiasis by scanning electron microscopy, X-ray microdiffractometry and energy-dispersive X-ray microanalysis. The calculus revealed a lamellar structure with irregular distribution of radipaque and radiolucent portions. The radiopaque portions contained apatite which could be demonstrated in prismlike as well as globular granular structures. In contrast to other authors, brushite was not detected by us. The radiolucent protions adjacent to the stone surface contained sulfur and showed undefined diffraction peaks supposing the presence of organic crystals. The possible milieu factors govering calculus formation are discussed.

Demonstration of structural variation in rat incisor dentin as determined by the x-ray Laue method.

Rat incisor dentin was studied by an x-ray micro-Laue method to investigate the relationship between crystal orientation and structural variation in the dentin. The results showed that crystal orientation was very poor in the labial dentin, but that the crystals were well-oriented in the lingual dentin. In the lingual dentin, the axis of crystal orientation was parallel to the tooth axis. It is suggested that the difference in crystal orientation between the lingual and labial dentin is due to the different mode of calcification between the two.

Plasminogen activator activity levels in patients with Behçet's syndrome.

We studied the plasminogen activator levels in plasma samples of 37 patients with Behcet's syndrome. A significant decrease of plasminogen activator activity level was shown by the euglobulin lysis time method, the amidolytic activity level (Testzym method), and the fibrin plate method. Such a decrease may be associated with the severity of Behcet's syndrome.

A Fukuyama type of congenital muscular dystrophy associated with atypical gyrate atrophy of the choroid and retina. A case report.

A three-year-ten-month old boy with the Fukuyama type of congenital muscular dystrophy (FCMD), early onset of muscular hypotonia, severe mental retardation, a slow progressive course and a characteristic fundus appearance similar to that of gyrate atrophy is described. Hyperornithinaemia and iminoglycinuria were not noted. To our knowledge, this is the first reported case of FCMD associated with atypical gyrate atrophy of the choroid and retina.

Destruction of implanted gastric tumors in rats by acridine orange photoactivation with an argon laser.

The phototoxicity of acridine orange and argon laser irradiation on Walker carcinosarcoma 256 stomach tumors was studied. Wistar strain rats bearing stomach tumors 4-6 mm in diameter 5-10 days after their implantation were injected intraperitoneally with 40 mg/kg of acridine orange 2 hr before irradiation. Then the forestomach was opened and the tumors were exposed to the argon laser at 488 nm at an intensity of 15 mW/cm2 for 20 min. Tumors in rats treated with acridine orange were brightly fluorescent during irradiation. No marked temperature rise was detected during irradiation. Argon irradiation significantly prolonged the survival of rats treated with acridine orange. Histologically, complete or partial tumor necrosis was observed, with sparing of surrounding mucosa, in rats treated by the combination therapy. Phase-contrast and electron microscopy showed that cytotoxicity was mediated by changes in the cell, nuclear and lysosomal membranes. Neither the dye nor laser alone had any effect.

Plasma steroid levels and clinical effects after topical application of betamethasone.

A study was made of 41 patients (45 eyes) with post-operative irritative conditions to evaluate the clinical efficacy of topical betamethasone (BM) at various concentrations. The results obtained were as follows. First, serum concentrations of BM after treatment with 0.01% and 0.05% BM solution were much lower than those found after a 0.1% solution had been applied. However, no reduction in the serum cortisol was observed in the former. Second, the clinical evaluation was made in patients after cataract extraction. Marked and moderate improvement of post-operative inflammatory changes was observed in 90% of the patients treated with 0.05% and 0.1% solution and in only 30% of those treated with 0.01% solution. This study clearly shows the efficiency of 0.05% solution of BM with minimal suppressive effects on adrenal function.

Free amino acid concentrations in blood cells of two brothers with gyrate atrophy of the choroid and retina with hyperornithinaemia.

In this study we selected blood cells as accessible tissues and measured the free amino acid concentrations in erythrocytes, lymphocytes, and granulocytes of two patients with gyrate atrophy with hyperornithinaemia. We found that ornithine concentrations were elevated in blood cells, to different degrees depending on the type of cell, and that aspartic acid concentrations were low in erythrocytes. We also investigated the fluctuation of free amino acid concentrations in blood cells up to 24 h after the oral administration of ornithine. The ornithine concentration in the blood cells of the patients reached a peak 2 h after the oral administration, and remained high for at least 4 h, although after 2 h the concentration curves differed in the three types of cells.

Milbemycins, a new family of macrolide antibiotics. Fermentation, isolation and physico-chemical properties of milbemycins D, E, F, G, and H.

Strain Au-3, a mutant of Streptomyces hygroscopicus subsp. aureolacrimosus, obtained with ultraviolet irradiation, was a high-yield strain of milbemycins D, E, F, G and H. Fermentation studies on the strains were conducted in shake flasks and 30-liter jar fermentors. Isolation of the metabolites was performed by adsorption on resinous adsorbent followed by elution with aqueous MeOH and separated by silica gel column chromatography. Milbemycin D was obtained as colorless needles after recrystallization and milbemycins E, F, G and H were purified to homogeneity by column chromatography. Physico-chemical characterization revealed that milbemycins D, E, F, G and H were new antibiotics possessing the 16-membered macrocyclic lactone with a 6,6-membered spiroketal ring system.
