Blood flow of the ipsilateral and contralateral lower limbs after isometric contraction.

Calf and thigh blood flows in both of right and left legs were measured simultaneously after isometric contraction of the right thigh muscle. Isometric contraction was performed at a force of about 50% of maximal voluntary contraction for 15 sec. The thigh blood flow of the right leg increased immediately after voluntary contraction in all subjects, while the calf blood flow of right leg decreased conversely in spite of exercised ipsilateral leg.

Ventilatory response to hypercapnia by rebreathing in successive trials.

The ventilatory response to carbon dioxide at rest was determined repeatedly by the rebreathing method; three successive CO2 response lines, with 30-min intervals, were examined in 12 healthy male subjects. Overall mean slopes obtained at 30, 60 and 90 min were 2.95, 2.47 and 2.28 liters/min . mmHg, respectively, the difference being statistically not significant. However, the slopes of four high responders, who were subjects with higher slopes than that of the mean values obtained here, decreased significantly (p less than 0.05) at 60 min as compared with the 30 min test, but this did not occur in the other subjects. Better reproducibility in obtaining the CO2 response slopes in the high responders was observed in the first rather than the 60 or 90 min trails, suggesting incomplete recovery in the humoral agents released during the CO2 test. These results indicated that the CO2 response curve by rebreathing should be determined carefully, especially in the high responder, taking into consideration its individual variability and and interval in consecutive measurements.

Effects of physical training on the calf and thigh blood flows.

Blood flows of the calf and thigh at rest and after submaximal and maximal exercise were determined by the venous occlusion method in 6 healthy subjects before and after physical training for 5 weeks. It was found that the magnitude of the flow after training was significantly (p less than 0.05) decreased when measured immediately after submaximal exercise, while there were no significant differences found with respect to both calf and thigh blood flows measured immediately after maximal exercise before and after training.

Inhibition of dopamine-beta-hydroxylase, A copper enzyme, by bleomycin.

Bleomycin was found to be one of the most potent inhibitors of dopamine-beta-hydroxylase. Bleomycin-A2 at 8 X 10(-8) M inhibited the enzyme activity by 50%. Kinetic studies showed that the inhibition by bleomycin-A2 was of the competitive type with both the substrate and the cofactor, ascorbate, and was not affected by fumarate, a stimulator for the enzyme. The inhibition mechanism is possibly due to chelating action of bleomycin toward the copper atom at the active site of the enzyme together with some other kinds of binding, for the addition of the cupric ions or extensive dialysis completely reversed the inhibition and bleomycin Cu(II)-complex did not inhibit the enzyme.

Adrenal medullary secretory response to stimulation of the medulla oblongata in the rat.

In rats anesthetized with sodium pentobarbital, adrenal venous blood samples were collected and analyzed for epinephrine and norepinephrine by the fluorometric method. Stimulation of the inferior fovea or adjacent areas resulted in a significant increase in epinephrine secretion. Stimulation of some other areas of the floor of the fourth ventricle was associated with a significant decrease in norepinephrine secretion with or without significant decrease in epinephrine secretion. In no instance was observed a significant increase in norepinephrine secretion in response to the medulla oblongata stimulation.

Partial purification and biological activities and properties of chick growth factors.

Cellular stimulating factors on cell proliferation in the supernatants of chick embryo carcases and adult muscles were studied. There were plural stimulating factors in embryonic and adult muscular supernatants that promoted cell proliferation without any supplement of sera and other materials. Salting-out methods with ammonium sulfate, ethanol fractionation, and isoelectric precipitation were used to isolate the stimulating factors, and these three methods proved the presence of plural stimulants on cell proliferation in the supernatants of chick embryo and adult muscles. The stimulants had altered physico-chemical properties and biological activities due to embryological development. The embryonic stimulants enhanced the synthesis of DNA and protein remarkably, and RNA synthesis in whole cell systems slightly. The muscular stimulants enhanced protein synthesis without any stimulation of DNA and RNA synthesis. Partial purification of the stimulants from the ethanol fractions was performed by DEAE-cellulose chromatography and Sephadex gel chromatography.
