The antihypertensive effect of (+/-)-3-(4-allyl-1-piperazinyl)-2,2-dimethylpropyl methyl 1,4-dihydro-2,6-dimethyl-4-(3-nitrophenyl)-3,5-pyridinedicarboxylate dihydrochloride (NKY-722, CAS 117241-46-0) was examined in conscious spontaneously hypertensive rats (SHR), normotensive Wistar rats (NWR) and anesthetized NWR, and its vasodilatory effect was investigated in the perfused NWR mesenteric vascular bed. NKY-722 and nicardipine administered intravenously (10-100 micrograms/kg) and orally (0.3-10 mg/kg) lowered blood pressure dose-dependently with an increase in heart rate in conscious SHR and NWR. The effects of NKY-722 were slower in onset and longer-lasting than those of nicardipine, and were more marked in SHR than in NWR. The effect of NKY-722 was roughly the same as that of nicardipine on intravenous administration. However, NKY-722 was 4-8 times more potent than nicardipine on oral administration. In anesthetized NWR, the hypotensive effects of NKY-722 administered via the femoral vein, portal vein and duodenum were examined in comparison with those of nicardipine. The findings suggest that NKY-722 is more efficiently absorbed from the gastro-intestinal tract and more resistant to the hepatic first pass effect than nicardipine. In the perfused NWR vascular bed, NKY-722 and nicardipine (0.01-1.0 microgram) attenuated the pressor response to KCl dose-dependently. The effect of NKY-722 was slower in onset and longer-lasting than that of nicardipine. In conclusion; NKY-722 has a potent, slow-onset and long-lasting antihypertensive activity, which is mainly attributed to its slow-onset and long-lasting vasodilatory action. NKY-722 is expected to be a useful antihypertensive drug.

A seroepidemiologic study of house dust allergy and pollinosis was performed among a general population aged 17 to 69 in Aichi Prefecture, Japan. The prevalence of blood containing IgE antibodies to four perennial antigens and three seasonal antigens was examined among 198 persons whose serum total IgE antibodies were above 40 IU/ml. The prevalence of blood containing IgE antibodies was from 9 to 18 percent among perennial antigens and from 6 to 30 percent among seasonal antigens. These results show that Dermatophagoides farinae, Cryptomeria japonica pollen and Anthoxanthumodoratum pollen are important antigens in this population.
The clinical and radiographic results of lumbar isthmic spondylolisthesis were compared between interspinous block-assisted anterior interbody fusion (block-assist group, n = 16) and anterior interbody fusion with no use of the block (nonassist group, n = 17) with an average follow-up of 7 years (range 1 1/3-13 years). Satisfactory relief of low-back pain, significantly early interbody union (union rate 88%, p < 0.05), and spontaneous fusion of pars defect (fusion rate 44%, p < 0.05) were obtained in the block-assist group. In the nonassist group, interbody union was markedly delayed (p < 0.05), the union rate was 53%, and spontaneous fusion of the pars was found in 12% of patients. The overall clinical results at final follow-up in both groups did not show a statistically significant difference, but the results tended to be superior in the block-assist group.
The superior colliculus (SC) plays an important role in the transformation of visual and auditory signals into motor commands to generate saccadic eye movements toward the targets. This study investigated the synaptic input patterns from the SC, into vertical eye movement-related superior and inferior rectus motoneurons, using intracellular recording and SC stimulating techniques in anesthetized adult cats. It was found that the responses obtained from these motoneurons consisted of complex components: a sequence of monosynaptic excitatory postsynaptic potentials (EPSPs), di- or trisynaptic EPSPs or inhibitory postsynaptic potentials (IPSPs) and polysynaptic EPSPs. The amplitudes of the latter two components almost coincided with the motor map of the SC, which represented the correlation between the sites of SC stimulation and the directions and amplitudes of evoked saccades. It was concluded that the SC elicits vertical eye movements through these di- or trisynaptic and polysynaptic pathways in the cat.
The omnipause neurons (OPNs), located in the nuculeus raphe pontis, cease firing during all rapid eye movements and otherwise fire at a steady rate in the awake state. Firing patterns of the OPNs were studied during the awake state, non-REM sleep, and REM sleep in chronically prepared adult cats, to examine whether the OPNs' firing participates in the genesis of eye movements during each state. During non-REM sleep, the OPNs continued their firing, and their firing patterns had little correlation with slow drifting eye movements appearing in this state. During REM sleep, the OPNs maintained a tonic firing and ceased their firing during rapid eye movements in all directions. These results suggest that cessation of OPN firing plays an important role in the generation of rapid eye movements during REM sleep as well as during the awake state in the cat.
We studied DNA ploidy, point mutation of Ki-ras oncogene codon 12, and p21 expression using paraffin embedded materials from 42 cases of colorectal cancer. DNA ploidy was measured by the method of Hedley et al. flow cytometrically. Point mutation of Ki-ras oncogene was examined by the method of Bos et al. using a dot-blot screening procedure, and p21 expression was examined immunohistochemically. Incidence of aneuploidy, Ki-ras point mutation, and p21 expression was 71.4%, 26.2%, 40.5%, respectively. There was a very weak correlation between p21 expression and pathologic findings, but there was no correlation between pathologic findings and DNA ploidy, as well as Ki-ras point mutation. Patients who showed aneuploidy tended to have more point mutation of Ki-ras oncogene. There was no correlation between p21 expression and Ki-ras point mutation, as well as DNA ploidy. Although there was no correlation between Ki-ras point mutation and survival, a significant correlation between survival and DNA ploidy, as well as p21 expression was recognized. Patients who had tumors with diploidy or p21 expression tended to have better prognosis.
A method is described for the determination of inorganic tin by gas chromatography with flame photometric detection. The inorganic tins, stannous and stannic, were extracted with hydrochloric acid and n-hexane-benzene in the presence of 0.05% tropolone, and both inorganic tins were pentylated to tetrapentyltin with a Grignard reagent prior to gas chromatography. The absolute limit of detection for tetrapentyltin was 3 pg as tin. The recovery of stannous chloride added to rat urine samples was 80.2 +/- 2.4% (mean +/- S.D., n = 8). The application of this method to the study of urinary excretion of inorganic tin and organotin compounds in rats following oral administration of tin compounds is presented. The urinary excretion of tin compounds was observed over a period of 96 h following administration of stannous chloride or phenyltin compounds. Most of the inorganic tin was excreted into urine within 24 h after administration of stannous chloride. In the experiments on organotin administration, the level of the excretion as total tin for monophenyltin reached a maximum ca. 0-24 h after administration, whereas the maxima for di- and triphenyltin were found after 24-48 h and 48-72 h, respectively. The predominant excretion product of these tin compounds found in urine was monophenyltin.
The inhibitory effects of ceruletide (CLT), a cholecystokinin-8 (CCK)-like peptide, were investigated in the epileptogenesis in the amygdaloid kindled rats. Lower doses of CLT (20-80 micrograms/kg) inhibited the progression of kindling process. After acquiring C5 stage, a higher dose (160 micrograms/kg) was required to suppress the seizure susceptibility. These results, in light of several previous studies showing no serious side effects, suggest that CLT might be useful as an anti-epileptogenic agent for clinical usage.
We have developed a reverse dot-blot hybridization assay for detection of Salmonella using Salmonella-specific oligonucleotide probes designed from the base sequence of the 16S rRNA gene (rDNA). The target fragment of 16S rDNA was amplified, and labelled with biotin by the polymerase chain reaction. The amplified fragment was hybridized with the membrane-immobilized probe and the hybridization was detected by chemiluminescence. Amplified fragments from 24 different serovars of Salmonella hybridized with the probes, whereas those of species of Enterobacteriaceae, Pseudomonas aeruginosa, Bacillus subtilis, and Staphylococcus aureus failed to hybridize. By this assay, it was possible to detect in the order of 10(4) bacteria in fish meat homogenate in 10 h.

The histologic effects of posterior lumbar surgery and retained extradural foreign bodies on the cauda equina were investigated in rats over time. The following four groups of rats were provided: Group 1, sham operation (laminar exposure alone); Group 2, laminectomy alone; Group 3, laminectomy with retained extradural silk thread; Group 4, laminectomy with extradural kaolin; and the control. Histological study was performed on the transverse sections of laminectomized (L5) and nonlaminectomized (L4) areas. In Group 1, adhesive changes involving the cauda equina, consisting of dilatation of nutrient vessels, and eosinophilic exudative changes between the adhering cauda rootlets were seen the day after surgery, but the fibrinous adhesion was resolved spontaneously in all rats after 1 week without permanent sequelae. In Groups 2 and 3, postoperative adhesion of the cauda equina roots was conspicuous, but it resolved by 6 weeks. In Group 4, obvious adhesion persisted through the 12th week after surgery. Destruction of the myelin sheath and axonal loss of the cauda equina occurred in the first week after surgery in all the experimental groups except in the sham group. The severity and extent of the neural degeneration were mostly parallel to the severity of inflammatory changes in the epidural areas. At the nonlaminectomized area (L4) in each group, the arachnoid and cauda equina tended to congregate the day after surgery, but these changes were all transitory. The severity and persistency of the arachnoiditis and neural degeneration directly corresponded to the magnitude of the inflammation and wound healing processes. This seems to imply that careful surgical intervention with no use of foreign bodies is necessary to reduce postoperative adhesive arachnoiditis.
The regional cerebral blood flow (rCBF) was measured with positron emission tomography (PET) in ten normal right-handed volunteers with the purpose of comparing rCBF changes related to movements of the dominant (right) and non-dominant (left) hand. The hand movement task consisted of sequential opposition of the thumb to each finger. The rCBF measured was compared with a rest state. Movements of the dominant hand and the non-dominant hand, increased CBF significantly in the contralateral motor area (MA) and the premotor area (PMA) with small increases in rCBF in the supplementary motor area (SMA). However, movements of the non-dominant hand also elicited significant ipsilateral increases in rCBF in the MA and PMA (6.3% and 5.0%, respectively). Superior part of the prefrontal area (PFA) of the left hemisphere showed significant CBF increases to both left and right hand movement. Our findings indicate that rCBF changes in the motor areas and the PFA of one hemisphere are not related simply to movement of the contralateral hand. Non-dominant hand movement may in addition require activation of ipsilateral motor areas. That is, there appears to be functional asymmetry in the MA and PFA in humans even in this relatively simple and symmetric motor task.
The distribution of beta 1- and beta 2-adrenoceptor subtypes in mice was determined in crude membranes prepared from the submandibular gland, lung, spleen, heart and kidney, all of which are major target tissues of sympathoadrenal neurons. Scatchard analysis using [125I]-(-)iodocyanopindolol gave linear plots and the order of the density of total beta-adrenoceptors was lung > submandibular gland > spleen > kidney > heart. Competition binding studies using beta 1-selective antagonist CGP20712A indicated that all tissues examined possessed both high and low affinity sites which approximately stand for beta 1- and beta 2-adrenoceptor subtypes respectively despite the notable difference in the ratios of high and low affinity sites depending on the tissues. The order of the occupancy of high affinity sites was submandibular gland (71%), heart (55%), spleen (18%), kidney (17%), lung (14%). These results will be helpful to elucidate the regulatory mechanism which underlies beta-adrenergic functions in mice.
A polyclonal antibody against the Na+,K(+)-ATPase holoenzyme was prepared. This antibody recognized native Na+,K(+)-ATPase and inhibited its activity. The peptide fragments corresponding to various regions of the Na+,K(+)-ATPase alpha-subunit, which were synthesized from the cDNA, were immunoprecipitated with the antibody, and the M32-D75, M158-D197 and M470-V552 fragments (the latter included K508, a putative ATP binding site) were identified as the epitopes. The M267-I442 fragment, which included a phosphorylation site at D376, and the C-terminal one-third of the alpha-subunit from M615 to the C-terminus, were not detected using this antibody. These results suggest that at least three regions of the Na+,K(+)-ATPase alpha-subunit, M32-D75, M158-D197 and M470-V552, cover its exposed regions, and that some of them are essential for ATPase activity.
The effects and pharmacological mechanisms underlying the inhibiting effect of ceruletide, a cholecystokinin (CCK)-related peptide, on apomorphine-induced turning behavior in 6-hydroxydopamine lesioned rats were investigated. For this purpose, selective CCKA and CCKB receptor antagonists were used. Ceruletide (50-400 micrograms/kg, s.c.) dose dependently suppressed apomorphine-induced rotational behavior. The antiapomorphine effect of ceruletide was reversed by the selective CCKA receptor antagonist, devazepide, but not by the selective CCKB receptor antagonist, L-365,260. This suggests that the suppression ceruletide exerts on hyperactive nigrostriatal dopamine neurons is mediated by CCKA receptors.
