Efferent connections of area 20 in the cat: HRP-WGA and autoradiographic studies.

Following injections of horseradish peroxidase-wheat germ agglutinin conjugate (HRP-WGA) and tritiated leucine into area 20 of the cat, terminal labeling was observed in visual areas 19, 21, the splenial visual area, the lateral suprasylvian area as well as in premotor, association and limbic related cerebral cortical regions. Labeled terminals in the subcortex were distributed in the caudate nucleus, the claustrum, the putamen, the anterior ventral nucleus, the intralaminar nuclei, the caudal division of the intermediate lateral nucleus, the lateralis posterior-pulvinar complex, the parvocellular C laminae of the dorsal lateral geniculate nucleus and the ventral lateral geniculate nucleus. In HRP-WGA preparations, retrogradely labeled somata were observed in these regions with the exception of certain subcortical structures. The projections are discussed with respect to the possible role area 20 plays in the cortical control of pupillary constriction.

Reducing terminals of urinary chondroitin sulfate.

Human urinary chondroitin sulfate isolated from the cetylpyridinium chloride-complex of the non-dialyzable fraction of the pooled urine was subjected to ethanol fractionation, successive enzymic digestion with neuraminidase and mucopolysaccharidases, and anion exchange chromatography. The gas liquid chromatographic analyses of the acetyl and butaneboronic acid ester derivatives of the reduced terminal sugar units after treatment with sodium borohydride plus hydrolysis revealed that 42% of the urinary chondroitin sulfate was bound to peptide through xylose. The reducing terminal sugar units of the peptide-free form consisted of 34.6% of xylose, 22.4% of galactose, 16.4% of glucose of unknown origin and 26.6% of glucuronic acid. These observations showed that the xyloside, galactoside and glucuronide linkages at non-terminal sites of carbohydrate chains of chondroitin sulfate were cleaved in tissues. It was thus suggested that the endo-types of beta-xylosidase, beta-galactosidase and beta-glucuronidase, which act on proteochondroitin sulfate are present in tissues.

Improved analytical method for the hexuronic acids at the reducing terminals of glycosaminoglycans.

When endo-uronidases act on glycosaminoglycans, the reaction products have hexuronic acid residues at the reducing terminals. An analytical method for hexuronic acids at the reducing terminals was devised for hyaluronate oligosaccharides having hexuronic acid residues at the reducing terminals. The procedure is as follows: Hexuronic acid residues at the reducing terminals of hyaluronate oligosaccharides were tritiated with reduction using NaB[3H]4 and the products were hydrolyzed with trifluoroacetic acid and nitrous acid. As a result, the tritiated and reduced hexuronic acid residues, that is aldonic acids, were liberated from the reducing terminals. After passing them through anion and cation ion-exchange resins, the aldonic acids were lactonized. The lactones were developed on paper chromatography, and their radioactivities determined on the paper. The method is also useful for discrimination between glucuronic acid and iduronic acid at the reducing terminals of glycosaminoglycans.

Splenic infarction, a complication of transcatheter hepatic arterial embolization for liver malignancies.

Splenic infarction was recognized in five of 37 patients who had hepatic neoplasms and who were being examined after transcatheter hepatic arterial embolization by CT during a follow-up study. CT images of the spleen showed multiple low density areas of a wedge or fused-wedge shape in most patients and of a rod or round shape in others. These low density areas decreased in size in time, and in two patients they disappeared in three months. Three of the five patients complained of a dull pain in the left upper quadrant that did not require any treatment. CT is effective in the diagnosis and assessment of splenic infarction. Awareness of this complication will help to avoid it when hepatic arterial embolization is being performed.

Some properties of synthetic fucopyranosylserines and -threonines.

alpha Fuc-Ser, alpha Fuc-Thr, beta Fuc-Ser, and beta Fuc-Thr were synthesized stereoselectively in good yields, and their properties were investigated. beta-Fucosylserine (and -threonine) was synthesized by Koenigs-Knorr reaction of per-O-acetylfucopyranosyl bromide with N-carbobenzoxy-L-serine (and -threonine) benzyl ester using mercuric cyanide, followed by hydrogenolysis and deacetylation. alpha-Fucosylserine (and -threonine) was synthesized by the halide-ion catalyzed reaction of per-O-benzylfucopyranosyl bromide with the serine (and threonine) derivative described above, followed by hydrogenolysis. These substances each gave a single peak on an amino acid analyzer. The anomeric configurations of these compounds were determined by 1H-NMR spectroscopy, optical rotation measurement and enzymatic degradations of these compounds. On acid hydrolysis, alpha Fuc-Ser and alpha Fuc-Thr were more rapidly hydrolyzed than the corresponding beta-series compounds. Under mild alkaline conditions in the presence of sodium borohydride, per-protected beta Fuc-Thr released about 50% of fucose but free beta Fuc-Thr did not However, free alpha Fuc-Thr and beta Fuc-Thr were cleaved to give acetylated fucose during acetylation with acetic anhydride-pyridine. alpha-L-Fucosidases, which were purified from abalone and rabbit livers by affinity chromatography, acted very well on alpha Fuc-Ser and alpha Fuc-Thr, but not on the beta-series compounds.

Fucose↗

Enzymic determination of urinary free L-fucose.

Conditions necessary for the precise measurement of free L-fucose in urine by an enzymic method using L-fucose dehydrogenase have been studied. The normal urinary levels of L-fucose were 16.4 /+- 9.1 micrograms/ml in children (22.2 /+- 6.5 micrograms/mg of creatinine) and 17.7 /+- 8.5 micrograms/ml in adults (16.6 /+- 5.7 micrograms/mg of creatinine). There was a close correlation between the concentration of free L-fucose and that of creatinine. A close correlation was also found between the concentration of free L-fucose and alpha-L-fucosidase activity. Thus, it was suggested that the free L-fucose in urine reflected the metabolism of L-fucose or L-fucose-containing glycoconjugates. In a preliminary screening test, several urine samples which showed high concentrations of free L-fucose, namely cases of "fucosuria" were found.

Linear and rod-shaped peptic ulcers.

Experience with the routine use of the pharmacologically aided, double-contrast examination to demonstrate various primary and healing ulcers over a six month period is assessed. Thirteen linear or rod-shaped ulcerations were diagnosed. It appears that gastric relaxation and the demonstration of fine mucosal relief are necessary to detect ulcers of linear or rod form.

Improved radiographic detection of multiple gastric ulcers.

In more than 1,500 double contrast upper gastrointestinal examinations performed January through November 1974, 26 cases of multiple gastric ulcers were diagnosed. This represents 18.6% of the 140 patients who had a demonstrable gastric lesion, and 23.4% of the 111 patients who had either a single gastric ulcer or scar. This technique was developed in a country where gastric carcinoma is common, but in the United States its greatest usefulness will probably be in the detection of subtle mucosal defects such as multiple gastric ulcers, linear ulcers, and erosions. The sensitivity of the routine pharmacologically aided double contrast upper gastrointestinal examination is confirmed by the high incidence of multiple gastric ulcers found.

Evaluation of routine double contrast views of the anterior wall of the stomach.

Since June 1974, we have performed over 1,500 consecutive double contrast upper gastrointestinal examinations. The double contrast study of the anterior wall has been a useful adjunct to conventional mucosal relief and dosed compression studies. Nevertheless, the low diagnostic yield of the anterior wall examination does not appear to justify including it as part of the routine double contrast upper gastrointestinal study.

Demonstration of erosive gastritis by the double-contrast technique.

The value of the routine pharmacologically-aided double-contrast examination of the upper gastrointestinal tract in demonstrating superficial gastric erosions was assessed. Ten cases of erosive gastritis were diagnosed during a six-month period. It appears that erosive gastritis is more common than generally believed. Diagnosis requires relaxation of the stomach and demonstration of fine mucosal relief by the double-contrast technique.
