Late results of postoperative long-term cancer chemotherapy for the gastric cancer patients subjected to curative resection.

Postoperative long-term cancer chemotherapy (PLCC) with a combination of Mitomycin-C (MMC), FT-207 and PSK (an immuno-stimulant) was prescribed for gastric cancer patients subjected to curative resection. The 5 year survival rates for patients with stage III and stage IV cancer were 58.3 per cent and 50.0 per cent in the PLCC groups, 48.0 per cent and 15.4 per cent in MMC groups, and 46.3 per cent and 13.3 per cent in no chemotherapy groups, respectively. In stage IV, the survival rate in PLCC group was significantly higher than that in MMC or no chemotherapy group (p less than 0.05). In the PLCC group, there was a tendency toward a dose-dependent effect in each group, and 5 year survival rate of stage III group administered over 60 mg of MMC, 60 g of FT-207 and 270 g of PSK was 70 per cent, such being remarkably higher than 46.3 per cent in those given no chemotherapy (p less than 0.07). There was no drug related death and only a slight leukopenia and hepatotoxicity occurred in some patients.

Combination chemotherapy enhances survival of patients with unresectable gastric cancer.

Of 177 Japanese patients with a gastric cancer which could not be resected and seen at our institution during the period from 1964 to 1979, 153 were investigated with regard to the efficacy of anticancer agents, in terms of prolongation of life. The average survival time was 23 weeks in the combination chemotherapy group (57 cases), 17 weeks in the single drug chemotherapy group (42 cases) and 13 weeks in no chemotherapy group (54 cases). Three and 6 month survival rates in the overall patients were 57.1 per cent and 16.7 per cent for single drug chemotherapy group, and 37.0 per cent and 11.1 per cent for no chemotherapy group, while in the combination chemotherapy group, the rates were higher at 64.9 per cent and 29.8 per cent, respectively (combination chemotherapy vs. no chemotherapy group, p less than 0.05). In patients with peritoneal dissemination, hepatic metastasis and carcinomatous ascites, there was a significant difference in survival rates between those prescribed combination chemotherapy and those given no chemotherapy (p less than 0.05). Of 57 in the combination chemotherapy group, 6 and 9 month survival rates were 45.5 per cent and 22.7 per cent in the postoperative long-term cancer chemotherapy (PLCC) group (22 cases), such being higher than other combination chemotherapy group (35 cases), 22.9 per cent and 11.4 per cent, respectively. There was a significant difference in the survival rates between the two groups (p less than 0.05).

Contraction of triton-treated culture cells. A calcium-sensitive contractile model.

Using five species of mammalian and avian cells, the authors succeeded in preparing Triton-treated culture cells that contract upon addition of MgATP. The contraction of these Triton cell models was inhibited by N-ethylmaleimide-modified myosin subfragment-1, a specific inhibitor of actin-myosin interaction. Triton cell models adhered more strongly to the substratum than glycerinated ones. Triton cell models of mouse 3T3 and human MRC-5 cells showed Ca2+-sensitive contraction. They required Ca2+ of 1 microM or more for the contraction. Other Triton cell models and all glycerinated cell models did not require Ca2+ for the contraction. The Ca2+-dependent contraction of 3T3 and MRC-5 cell models was inhibited by chlorpromazine, an inhibitor of calmodulin. The Ca2+-sensitivity of the contraction was lost by pretreatment of these cell models with adenosine 5'-O-(3-thiotriphosphate) in the presence of Ca2+. These results agree with a hypothesis that Ca2+-calmodulin-dependent phosphorylation of myosin light chain regulates actin-myosin interactions in non-muscle cells.

[Cefotaxime concentration in the prostatic tissue].

Concentrations of cefotaxime (CTX) in serum and prostatic tissue were studied in human and rat. CTX kept high level of 40 to 100 micrograms/g in prostatic tissue at least as long as 2 hours following intraperitoneal injection in a dosis of 100 mg/kg of body weight in rat. Patients intravenously injected of 1 g of CTX showed the activity of 6.8 +/- 4.1 micrograms/g in prostatic tissue.

[Effect of Moristerol (anti-cholesterolemic agent) on benign prostatic hypertrophy].

Moristerol was orally administered in 20 cases of benign prostatic hypertrophy. One capsule of Moristerol contained 200 mg of soysterol . In all the cases, 9 capsules of Moristerol were given per day in three divided doses for 6 weeks. Clinical improvement was observed in 11 of the 20 cases (55%). It consisted mainly of subjective complaints such as improvement of dysuria, narrow and weak urinary stream and feeling of incomplete voiding. As objective findings, a statistically significant decrease of residual urine volume was noted after treatment, although shrinkage of the enlarged prostate was not clear upon palpation or roentgenography. In addition to the improvement of symptoms, serum lipid peroxide which might be considered to be a provocative cause of benign prostatic hypertrophy, was also significantly decreased by Moristerol administration. No marked side effects were seen in this study.

[Adjuvant chemotherapy for aged patients with gastric carcinoma].

The effectiveness of postoperative adjuvant chemotherapy on aged patients (more than 70 years old) with resectable gastric carcinoma was examined in our Department. Of the 178 patients, 75 patients received the drugs on various regimen and 103 patients had no chemotherapy a control group). No significant difference was noted in patient characteristics of age, sex and curability. In advanced gastric carcinoma such as stage III and IV, a significant increase in long-term survival after surgery was obtained with combination chemotherapy or immunochemotherapy compared with the control group (P less than 0.05). On the other hand, there was no significant difference in a survival rate of the patients with stage I and II.

Serial determinations of carcinoembryonic antigen for early detection of recurrent gastric cancer.

In attempts to predict the recurrence of gastric cancer, postoperative changes in serum carcinoembryonic antigen (CEA) levels are monitored in our clinic by radioimmunoassay (Dainabot, Japan). Recurrences are suspected when serum CEA levels are 4 ng/ml, in the postoperative period. Out of 34 patients in whom there were increases in serum CEA, 18 were confirmed to have a recurrence and 15 of these 18 patients were assessed accurately by serial postoperative levels of CEA, two patients died of a recurrence after elevation of serum CEA levels. Thus, recurrence was predicted in 17 out of 34 patients (50 per cent) and in 12 out of 17 patients there was a metastasis to the liver. In 14 out of 34 patients there are no signs of recurrence 9 to 25 months after serum CEA elevations.

Isolation and characterization of circumferential microfilament bundles from retinal pigmented epithelial cells.

Chicken retinal pigmented epithelial cells have circumferential microfilament bundles (CMBs) at the zonula adherens region. We have isolated these CMBs in intact form and characterized them structurally and biochemically. Pigmented epithelia obtained from 11-d-old chick embryos were treated with glycerol and Triton. Then, the epithelia were homogenized by passing them through syringe needles. Many isolated CMBs were found in the homogenate by phase-contrast microscopy. They formed polygons, mostly pentagons and hexagons, or fragments of polygons. Polygons were filled with meshwork structures, i.e. they were polygonal plates. Upon exposure to Mg-ATP, isolated CMBs showed clear and large contraction. The contraction was inhibited by treatment with N-ethylmaleimide-modified myosin subfragment-1. After purification by centrifugation with the density gradient of Percoll, CMBs were analyzed by SDS PAGE. The electrophoretic pattern gave three major components of 200, 55, and 42 kdaltons and several minor components. Electron microscopy showed that the polygons were composed of thick bundles of actin-containing microfilaments, and the meshworks were composed primarily of intermediate filaments.
