[Pulmonary manifestations of alveolar soft part sarcoma; bronchial arterial embolization for hemoptysis].

Alveolar soft part sarcoma is a rare soft tissue tumor arising in extremities. The universal tendency for late widespread metastasis to lung, bone and brain, make the treatment of this tumor a particular challenge. This time, we performed the intra-arterial infusion of MMC and bronchial arterial embolization by spongel for hemoptysis of this metastatic lung tumor. Rosenbaum has reported the characteristics of angiogram of the metastatic brain tumor. In our case, bronchial arteriography showed the manifest hypervascular pattern "rosary form," "pooling" and "B-P shunt." These findings should yield benefits for the diagnosis and treatment of this metastatic lung tumor.

[Prevention of immunodeficiency induced by cancer chemotherapy with BCG].

MFC (MMC, 5-FU and cytosine-arabinoside) therapy applied with the liver organism Bacillus Calmette-Guérin (BCG) for the treatment of postoperative patients with cancer of the digestive organs presenting at stage II and III. Immunological parameters included skin reaction by purified protein derivative (PPD), lymphatic blastogenesis test using phytohemagglutinin (PHA) and lymphatic subsets. The frequency of MFC therapy was significantly higher in the MFC plus BCG group than in the MFC group (p less than 0.001). At the completion of MFC therapy, both of PHA blastogenesis rate, OKT4/OKT8 and OKT3 were all within the normal limits. The PPD skin reaction was positive (18.2 +/- 4.0mm) as before the start of MFC therapy. These results suggest that BCG immunotherapy may potentiate the effect of chemotherapy.

Mechanisms of cross-resistance between radiation and antineoplastic drugs.

The mechanisms responsible for the cross-resistance between radiation and certain antineoplastic agents have been examined in human ovarian cancer cell lines. Cell lines established from patients at a time when they were resistant to combination chemotherapy regimens, which included cisplatin and an alkylating agent as well as cell lines with resistance induced in vitro to melphalan and cisplatin, all have increased cellular levels of glutathione (GSH) compared with drug-sensitive cell lines from untreated patients. In addition, cell lines with acquired resistance to melphalan and cisplatin, but not to doxorubicin, were cross-resistant to radiation. L-Buthionine sulfoximine (BSO), an irreversible inhibitor of gamma-glutamylcysteine synthetase, lowered GSH levels in all the resistant cell lines studied. Lowering of GSH levels to less than 10% of control values potentiated the in vitro cytotoxicity of melphalan and cisplatin. Furthermore, BSO was also shown to potentiate the cytotoxicity of melphalan in a nude mouse model system of ovarian cancer in which mice die of disseminated intra-abdominal carcinomatosis. The BSO administered in the drinking water decreased GSH levels by 96%. A single melphalan treatment of 5 mg/kg following GSH depletion produced a 72% increase in median survival time compared with treatment with melphalan alone. In addition, depletion of GSH levels in cell lines with acquired resistance to melphalan led to a marked sensitization of these cells to irradiation.(ABSTRACT TRUNCATED AT 250 WORDS)

[Clinical effect of terodiline hydrochloride (TD-758) in patients with urinary disturbances].

We evaluated the efficacy of terodiline hydrochloride (TD-758) in 23 patients with psychosomatic bladder, chronic prostatitis and chronic cystitis. The drug was administered at a dose of 24 mg once a day for 4 weeks. In 78% of the patients the symptoms such as urinary frequency and sense of residual urine improved. Only mild side effects such as thirst were noted. We concluded that terodiline hydrochloride was effective in this trial.

[A case of pancreatic cancer with invasion to the wall of the intrahepatic bile duct].

Presented here is the case of a 69-year-old woman with pancreatic cancer. Efforts were made to discover the origin of the cancer but without success. Tumor nodules were scarcely observed in the pancreas macroscopically at autopsy, while histological examination of the pancreas revealed the presence of a well-differentiated adenocarcinoma (duct cell carcinoma). The invasion to the wall of the intrahepatic bile duct was also observed microscopically.

The role of tubulin polymerization during spindle elongation in vitro.

We describe the effect of exogenous tubulin on reactivation of anaphase spindle elongation in isolated diatom spindles. In the absence of tubulin, spindle elongation is limited to the equivalent of the microtubule overlap zone, but in the presence of tubulin spindle elongation is several times the length of the overlap zone. Biotinylated neurotubulin is incorporated into the overlap zone and around the poles. Before spindles have elongated by the equivalent of the overlap zone, there are two regions of incorporated tubulin flanking this zone. After further elongation, there is one broad zone of incorporated tubulin in the spindle midzone. Spindle elongation and the pattern of tubulin incorporation into the midzone, but not the poles, are ATP-dependent and vanadate-sensitive. These results suggest that tubulin adds onto the ends of microtubules in the overlap zone, which then slide through the midzone as the spindle elongates.
