Effects of high level of vitamin C on tissue antioxidant status of guinea pigs.

Two groups of weanling male Hartley guinea pigs maintained on vitamin E deficient diet were supplemented with 0.4 I.U./100 g body weight/day of vitamin E and either 2 (Group A) or 10 (Group B) mg/100 g body weight/day of vitamin C for 5 weeks. As compared to Group A, the degree of erythrocyte hemolysis and liver TBAR level of Group B were significantly increased while plasma vitamin E and erythrocyte GSH levels were significantly decreased. In another experiment, two groups of guinea pigs were given 0.8 I.U./100 g body weight/day of vitamin E and 2 (Group C) or 30 mg/100 g body weight/day (Group D) of vitamin C. Levels of plasma vitamin E and erythrocyte GSH of Group D were significantly lower than those of Group C: however, erythrocyte hemolysis and liver TBAR were not affected by the level of vitamin C supplementation. The results suggest that the high levels of vitamin C supplementation lowered tissue antioxidant potential of animal when vitamin E was marginally adequate, and the hemolytic and peroxidizing effect of high level of vitamin C may be counteracted by increasing the level of vitamin E.

Effects of Fructus gardeniae extract on hepatic function.

This article discusses the effects of Fructus Gardeniae extract on hepatic function. Fructus Gardeniae extract manifested no hepatotoxic effects on rats, as shown by alkaline phosphatase, aspartate aminotransferase, and lactate dehydrogenase studies. Fructus Gardeniae extract failed to activate the UDP-glucuronyltransferase system; whereas in hyperbilirubinemic state the enzyme was activated, presumably by substrate induction. Fructus Gardeniae extract increased the activity of UDP-glucose dehydrogenase, which would result in an increase in availability of UDP-glucuronic acid intracellularly, BSP clearance study showed an unexpected impairment of hepatic uptake of the dye after extract treatment. The action mechanism involved in lowering of serum bilirubin level by Fructus Gardeniae extract may well be complex; it is probably acting on a locus other than glucuronyl transferase.

Tie-1 tyrosine kinase is an independent prognostic indicator for invasive breast cancer.

Receptor tyrosine kinases are known to be involved in the growth, progression and metastasis of solid tumors. We investigated the relationship between tie-1 expression and progression of invasive ductal breast carcinoma with immunohistochemical analysis. Tie-1 protein was detected in the microvessel endothelial cells and cytoplasm of tumor cells. The tumor size and stage were significantly associated with the expression of tie-1, which portends a worse 5-year disease-free status (39.3% v 59.2%, p = 0.07) and overall survival rate (67.3% v 93%, p = 0.02) than those without tie-1 expression. Multivariate analysis demonstrated that larger tumor size, presence of lymph node metastasis and tie-1 expression were independent prognostic parameters, both in 5-year disease-free survival and overall survival. Patients with lymph node metastases and tie-1 expression had the worst 5-year disease-free survival (0%) and overall survival (42.4%) compared to those without tie-1 expression (50.2%, 85%). In lymph node negative patients, those without tie-1 expression had better 5-year disease-free survival and overall survival (72.9%, 100%) compared to those with tie-1 expression (65.5%, 87.7%). We conclude that tie-1 expression is an independent prognostic factor for invasive ductal breast carcinoma, adversely affecting survival of breast cancer patients with positive nodes to a significant extent.
