Mutation in the Jak kinase JH2 domain hyperactivates Drosophila and mammalian Jak-Stat pathways.

The Jak (Janus) family of nonreceptor tyrosine kinases plays a critical role in cytokine signal transduction pathways. In Drosophila melanogaster, the dominant hop(Tum-l) mutation in the Hop Jak kinase causes leukemia-like and other developmental defects. Previous studies have suggested that the Hop(Tum-l) protein might be a hyperactive kinase. Here, we report on the new dominant mutation hop(T42), which causes abnormalities that are similar to but more extreme than those caused by hop(Tum-l). We determined that Hop(T42) contains a glutamic acid-to-lysine substitution at amino acid residue 695 (E695K). This residue occurs in the JH2 (kinase-like) domain and is conserved among all Jak family members. We determined that Hop(Tum-1) and Hop(T42) both hyperphosphorylated and hyperactivated D-Stat when overexpressed in Drosophila cells. Moreover, we found that the hop(T42) phenotype was partially rescued by a reduction of wild-type D-stat activity. Finally, generation of the corresponding E695K mutation in murine Jak2 resulted in increased autophosphorylation and increased activation of Stat5 in COS cells. These results demonstrate that the mutant Hop proteins do indeed have increased tyrosine kinase activity, that the mutations hyperactivate the Hop-D-Stat pathway, and that Drosophila is a relevant system for the functional dissection of mammalian Jak-Stat pathways. Finally, we propose a model for the role of the Hop-D-Stat pathway in Drosophila hematopoiesis.

[The prevalence of hypertension, diabetes mellitus and hyperlipemia among 1369 subjects for physical examination in Changsha].

A survey of the prevalence of hypertension (HT) and diabetes mellitus (DM) and hyperlipemia (HL) was conducted among 1369 subjects for health check in Changsha. The results showed that the prevalence of HT and DM and impaired glucose tolerance (IGT) and HL were 17.2% and 4.6% and 5.9% and 27.4%, respectively. The prevalence of HT and DM and IGT increased with age. The higher the body mass index was, the higher the prevalence of HT and DM and HL was. These findings indicate that the aging and over weight are important risk factors to the prevalence of HT, DM and HL.

Burnout in Ghanaian hospitals: phase model findings in sub-Saharan Africa.

This replication shows the expected covariation of the phases of burnout with a set of 5 marker variables. As the phases progress I-->VIII, so do decreases or deficits occur on all the marker variables. All covariants far surpass usually-accepted levels of statistical significance, although the magnitudes are not as great as in much other research with the phases. The Ghanaian incidence of the phases is also compared with several panels of populations. Those comparisons at once indicate a substantial Ghanaian incidence of advanced phases, as well as a distribution comparable to North American worksettings and more favorable than a panel of global worksettings.

[Circadian variations of plasma SOD and MDA in health subjects].

Oxygen derived free radical system has been to be implicated in the pathogenesis of many diseases, such as cardiovascular disease, tumor, trauma and radiation injury. Many studies have suggested that oxygen free radical initiate a series of events that result in cell membrane alterations leading to the development of cell injury. The objective of the present study was to examine the circadian characteristics of some parameters of this system in the blood. One parameter was superoxide dismutase (SOD), a major oxygen free radical scavenger; the other was malondialdehyde (MDA), an important product of lipid peroxidation initiated by oxygen free radicals. Blood samples were collected at 6 h intervals beginning at 08:00 over a 24 h span in nine healthy volunteers (5 females, 4 males, average age 30 +/- 5 years). All participants were synchronized for one week with diurnal activity from 07:00 to 23:00 and nocturnal rest, with three meals served at 07:30, 12:00 and 19:00. Plasma SOD, MDA levels were measured by pyrogallol autoxidation assay, thiobarbituric acid spectrophotometry respectively, and the data were fitted with a 24 h cosine curve and analyzed further by a population-mean cosinor. The results showed that the plasma SOD levels exhibited a highly significant circadian rhythm (P < 0.001) in healthy subjects, with acrophase (phi) at 21:17, mesor (M) of 3975.3 +/- 680.5 u/g Hb, and amplitude (A) of 273.62 u/g Hb. A circadian rhythm of plasma MDA was also detected (P < 0.01), phi = 15:15, M = 4.23 +/- 0.86 mumol/L, A = 2.37 mumol/L). The evaluation of circadian natures of SOD and MDA may be important in the prevention, diagnosis and treatment of associated diseases.

[The effect of Tianma injection on cultured vascular smooth muscle cells proliferation].

The proliferation of vascular smooth muscle cells (VSMC) has been thought to play an important role in the development of hypertension and atherosclerosis. The aim of this study was to investigate the effect of Tianma Injection and its major component Gastrodin on the proliferation of cultured VSMC. VSMC were cultured and randomly divided into the control, Tianma and Gastrodin groups. The results showed that Tianma Injection significantly decreased cell numbers dose-dependently and time-dependently, and it inhibited 3H-TdR incorporation in VSMC. It also increased the content of PGI2 production in VSMC. However, no significant differences of the above measurements were observed between the Gastrodin and control groups. The suggest that Tianma Injection can inhibit VSMC proliferation and the effect possibly results from its increasing PGI2 production content, and that the inhibiting effect of Tianma is irrelated to Gastrodin.

[A comparative study on the chemical components of Salvia miltiorrhiza Bge. collected from Zhongjiang of Sichuan and from other habitats].

In this paper, the chemical components in the root of Salvia miltiorrhiza from three habitats (Zhongjiang of Sichuan, Henan and Shandong) were comparatively studied. The results show that the TLC chromatogram and contents of tanshinone IIA in the crude drugs are about the same, the sodium content appears to be lower in the sample from Sichuan, but the contents of other metallic elements are about the same in the samples from different habitats.

[Application of TLC to tannin measurement].

There has been little report on the use of in measuring tannin in traditional Chinese medicinal preparations by TLC. Gallic acid, the decomposd product of tannin, was in bright yellow and green color, showing a strong specificity. The measurement of gallic acid can provide reliable indexes for quality assay of TCM preparations.

[The effect of combining intra-aortic balloon pumping with tianma injection on myocardial ischemic-reperfusional injury in goats].

The objective of this study was to explore the effect of combined Intra-Aortic Balloon Pumping (I-ABP) and Tianma Injection on necrotic area in goat myocardial ischemic-reperfusional models which were established by two hours ischemia and a follow-up reperfusion. The results showed that the combined effect of IABP and Tianma Injection was more significant than that of IABP or Tianma separately. It is suggested that IABP and Tianma Injection have synergetic effect on the limitation of necrotic area, which possibly results from their mechanisms for decreasing the size of infarct.

[Application of computerized image analyzing system in morphometric study of vascular smooth muscle cells].

The authors used image analyser to measure several morphometric parameters of cultured aorta vascular smooth muscle cell (VSMC) and assess the relationships between these morphometric parameters and 3H-TdR incorporations. On this basis, the authors also explored the effect of Ligustrazine, a traditional Chinese medicine, on VSMC morphology in order to evaluate the role of computerized image analysing system in the study of VSMC proliferation. The results showed that (1) there were significant correlations between VSMC cellular area, nuclear area and 3H-TdR incorporations in control group; (2) Ligustrazine significantly decreased VSMC cellular area, nuclear area and nuclear perimeter, and a significant correlation was also noted between cellular area and 3H-TdR incorporation in Ligustrazine group. These suggest that computerized image analysing system is a rapid, accurate and reliable method to study the morphology and proliferation of VSMC.

Effects of configuration on the myocardial uptake of radioiodinated 3(R)-BMIPP and 3(S)-BMIPP in rats.

UNLABELLED: Radioiodinated 3(R)-(+)- and 3(S)-(-)-15-(p-iodophenyl)-3-(R,S)-methylpentadecanoic acid (BMIPP) were prepared and evaluated in rats to investigate the effects of absolute configuration of the 3(beta)-methyl group on myocardial uptake and release kinetics. METHODS: The 3(R)-(+)-BMIPP analog was synthesized by initial acylation of a thiophene template with the acid chloride of ethyl 3(R)-methylglutarate. 3(S)-(-)-BMIPP was obtained by separation from the mixture of diastereomeric amides prepared from reaction of the acid chloride of racemic BMIPP with the S-(-)-alpha-methylbenzylamine. The amide of synthetic 3(R)-BMIPP prepared from S-(-)-alpha-methylbenzylamine was identical to the chromatographically more polar isomer. Free acids were obtained by acid hydrolysis of the amides, fully characterized and then converted to the radioiodinated BMIPP isomers. RESULTS: Biodistribution studies in rats with the dual-labeled [(131)I]-3(S)-BMIPP/[(125)I]-3(R)-BMIPP mixture demonstrated greater myocardial uptake of 3(R)-BMIPP compared with the 3(S)-BMIPP isomer [60 min: 3(R)-BMIPP = 4.37 %ID/g; 3(S)-BMIPP = 3.44; p < 0.05; 180 min, 2.31 and 1.78 %ID/G, respectively, p < 0.01], although both isomers had similar myocardial washout curves (5-180 min). Percent ID/g values for other tissues which were examined (blood, lungs, thyroid) were similar. CONCLUSION: Higher myocardial uptake of the 3(R)-BMIPP isomer observed in these animal studies may suggest differences in carrier-mediated myocyte uptake of the two isomers. These studies suggest that [(123)I]-3(R)-BMIPP is a candidate for clinical evaluation and may show greater myocardial uptake than the 3(S)-BMIPP isomer and may thus require reduced injected dose.

[Inhibition of sodium selenite on transformation of human B lymphocytes by Epstein-Barr virus].

We detected the effect of non-toxic doses of sodium selenite (Se) on the expressing effect of nuclear antigen (EBNA) of Epstein-Barr virus (EBV) in Raji cell line by microfluorescence spectrophoto-quantitation (MFS) and studied the inhibing transformation in B lymphocytes from human umbilical cord blood (BL) with EBV in vitro by Se. We found that Se (0.01-0.50 microgram/ml) can lower EBNA-MFS value 8.4%-21.8% in Raji cell line. The transformation effect of BL with EBV was markedly inhibited by Se (0.1-1.0 microgram/ml) in pretreatment and cotreatment. The inhibition rates of BL transformation were 73.4%-92.1% and 60.3%-77.2% respectively. EBV is associated with nasopharyngeal carcinoma (NPC). The Se levels of serum and hair in NPC patients were markedly lower than those in healthy persons. The results suggest that supplement Se to persons for active expression of EBV gene and NPC patients may help protect againt and treat NPC and diseases associated with EBV.

[The protective role of heat shock gene expression on hydrogen peroxide induced pulmonary alveolar macrophages].

The aim of this study is to determine whether heat-induced heat shock gene expression in rat pulmonary alveolar macrophages (PAMs) would protect PAMs against hydrogen peroxide (H2O2)-mediated cell killing. In response to sublethal heat shock at 42 degrees C for 2 h, the cells synthesized heat shock protein 70 kD (HSP70) and other different molecular weight heat shock proteins, which were detected with gel electrophoresis after [35S]-methionin labeling cellular protein and Western blotting analysis in PAMs. Northern blot analysis showed the induction of HSP70 mRNA with heat shock treatment. Futhermore, with heat shock, there was a significant increment of survival cells after H2O2 (1,2,3 mmol.L-1, 45 min) exposure. This increase was blocked both by Cycloheximide and by Actinomycin D, and was associated with inhibition of synthesis of HSP70 protein and transcription of HSP70 mRNA. These results strongly suggest that expression of heat shock gene (especially HSP70 gene) plays an important role in the intracelluar mechanism of cytoprotection against H2O2 in rat PAMs.

Preserved vasodilatory response to nitroglycerin in saphenous vein bypass grafts.

BACKGROUND: Uncertainty exists regarding the effects of nitroglycerin on atherosclerotic segments of coronary arteries, and information on vasoreactivity of saphenous vein bypass grafts is sparse. Intravascular ultrasound enables identification of atherosclerosis in angiographically normal segments and allows continuous determination of alterations in cross-sectional lumen areas. METHODS AND RESULTS: Patients with documented coronary atherosclerosis were studied. Vessel morphology and lumen area at baseline and after 100 to 200 micrograms nitroglycerin were assessed at 10-second intervals for 60 seconds in vessel segments without angiographically apparent lesions. Coronary artery saphenous vein bypass grafts from 11 patients were compared with native coronary arteries in 16 different patients. Atherosclerosis was present in all vessel segments studied. There was a rapid increase in lumen area compared with baseline after intravascular nitroglycerin in both native coronary arteries and saphenous vein bypass grafts. Maximum lumen area dilatation was 19.6 +/- 12.2% in saphenous vein bypass grafts and 19.7 +/- 13.1% in native coronary arteries. An earlier peak response in saphenous vein bypass grafts (34.5 +/- 6.9 seconds) compared with native coronary arteries (44.7 +/- 8.5 seconds; P = .003) was found. Vessel wall area remained constant during vasodilation, but there was a significant reduction in measured wall thickness (P = .034). CONCLUSIONS: In patients with documented coronary artery disease, intravascular ultrasound reveals substantial atherosclerosis in angiographically normal vessel segments. In these vessel segments, both native coronary arteries and saphenous vein bypass grafts exhibit prompt vasodilation with the intravascular administration of nitroglycerin. The vasodilatory capacity in response to nitroglycerin seems to be preserved in transposed, denervated, and devascularized saphenous veins.

Impaired signaling in alloantigen-specific CD8+ T cells tolerized in vivo: employing a model of Ld-specific TCR transgenic mice transplanted with allogenic hearts under the cover of a short-term rapamycin treatment.

Peripheral tolerance of T cells is necessary because thymic deletion is not complete, and tissue-specific Ags exist outside the thymus. We have reported that persistent Ag is required to maintain peripheral tolerance in vivo. We suspect that the TCR signaling pathway in in vivo tolerized cells is compromised due to continuous exposure to the Ags. In this study, the TCR signaling events in these cells were investigated using TCR transgenic mice (2C mice) whose T cells are predominantly Ld alloantigen-specific CD8 cells. The 2C mice were thymectomized and then rendered tolerant to Ld Ag by allogenic heart transplantation plus short-term treatment with rapamycin. We found that 1) the in vivo tolerized CD8 cells have compromised intracellular Ca2+ flux upon mitogen stimulation; and 2) their cellular tyrosine proteins fail to be phosphorylated properly upon TCR cross-linking. These results indicate that the signaling pathway in the in vivo tolerized CD8 cells is indeed defective. We also found that 1) the tolerized CD8 cells have no characteristic surface markers; and 2) the allograft is probably the place where the rejection response is initiated according to the appearance of an early activation marker of T cells on graft-infiltrating cells.

Rapamycin inhibits aldolase A expression during human lymphocyte activation.

Rapamycin (RAPA) strongly inhibits lymphocyte activation and proliferation, but does not affect most of the activation-related gene expression at the mRNA level. In order to understand the mechanism of action of RAPA and to gain further insights in lymphocyte signalling which is impaired by RAPA, we screened for RAPA-sensitive genes using differential hybridization. The expression of human aldolase A gene was found to be inducible during T and B cell activation, and the induction was repressed by RAPA at both the mRNA and enzymatic levels. The other two important immunosuppressants, cyclosporin A and FK506, also inhibited the mitogen-induced upregulation. However, none of these three drugs inhibited the constitutive expression. There was no fluctuation of aldolase A expression during the cell cycle, and RAPA failed to block the first cell cycle after synchronization in Jurkat cells. However, the second cycle was hampered by RAPA, and this was correlated with the inhibition of aldolase A expression during this later stage. Since aldolase A is a key enzyme in glycolysis and lymphocytes mainly depend on glycolysis for energy supply, the data from this study suggest that aldolase A might be one of the downstream targets of RAPA. The inhibition of the enzyme upregulation might deprive the cells of additional supply of energy, and prevent the cells from entering an optimal status for proliferation.

Biochemical characterization of the reverse transcriptase of a human intracisternal A-type particle (HIAP).

The discovery of novel human intracisternal A-type particle (HIAP) that may be associated with the autoimmune disease Sjŏgren's syndrome has been previously reported. Although the HIAP retrovirus has been shown to be antigenically related to HIV-1, the viruses were distinguishable by different hydrodynamic mobilities through a sucrose gradient by morphology and intracellular location, and by differing divalent cation requirements for their in vitro reverse transcriptase (RT) reactions. In this report, additional biochemical characteristics are provide that further differentiate the HIAP RT from HIV-1 RT. Data are also presented that distinguish the HIAP RT from the known cellular DNA polymerases.

Transcutaneous ultrasound augments lysis of arterial thrombi in vivo.

BACKGROUND: External ultrasound has a synergistic effect on thrombus disruption with thrombolytic agents in vitro. We hypothesized that transcutaneous ultrasound could augment thrombolysis in vivo. METHOD AND RESULTS: Thrombus formation was induced electrically in 48 pairs of iliofemoral arteries of 24 rabbits; arterial occlusions were documented angiographically. In 17 of 24 rabbits, 25000 units/kg streptokinase was then administered intravenously. The pairs of iliofemoral arteries were randomized to receive ultrasound treatment or no ultrasound treatment. Low-frequency (26 kHz) ultrasound (continuous wave, 18 W/cm2) was applied transcutaneously over the area of occlusion. In 7 of 24 rabbits, 14 thrombotically occluded iliofemoral arteries were exposed to ultrasound alone without streptokinase. The results were evaluated through the use of angiography (TIMI grade flow) and histopathology. After 30 +/- 10 minutes of activated sonication combined with intravenous streptokinase, 10 of 17 iliofemoral arteries (59%) treated with transcutaneous ultrasound were widely patent angiographically, with TIMI grade 3 flow. Histologically, the patent arteries had only minimal focal moral thrombus. The angiographic patency rate was significantly lower in the control groups: 1 of 17 arteries (6%) treated with streptokinase alone (P = .0012) and 1 of 14 arteries (7%) treated with ultrasound alone (P = .0036). CONCLUSIONS: In vivo transcutaneous ultrasound significantly augments lysis of thrombi with streptokinase in rabbit iliofemoral arteries.
