Carnitine palmitoyltransferase: activation and inactivation in liver mitochondria from fed, fasted, hypo- and hyperthyroid rats.

The sensitivity of carnitine palmitoyltransferase to malonyl-CoA is lost when liver mitochondria are preincubated in a KCl-containing medium. This loss of sensitivity is slowed down in mitochondria from hypothyroid rats and accelerated in mitochondria from fasted and hyperthyroid rats. Glucagon seems to enhance the effect of fasting. The loss of sensitivity is significantly slowed down by 50-500 nM malonyl-CoA and accelerated by small amounts of palmitoyl-CoA in the preincubation medium.

Effects of thyroid state and fasting on the concentrations of CoA and malonyl-CoA in rat liver.

Liver CoA is markedly higher in hyperthyroid rats as compared to hypothyroid rats, and in fasted rats as compared to fed rats. In hyperthyroid rats the CoA is increased mainly in the cytosol, while in fasted rats the increase is mainly in the mitochondria. Malonyl-CoA is markedly higher in hypothyroid rats than in euthyroid and hyperthyroid rats. With fasting, malonyl-CoA drops 80-85% in all thyroid states. These findings are discussed in relation to the regulation of fatty acid oxidation in the liver.

Effect of gamma interferon on spreading of mouse peritoneal macrophages on solid surfaces.

The influence of a homologous gamma interferon preparation (MuIFN-gamma) on in vitro spreading of freshly-seeded unelicited mouse peritoneal macrophages (MPM) was tested. Light microscopy and scanning electron microscopy observations gave comparable results. Addition of MuIFN-gamma enhanced the spreading on solid surfaces, especially 3-5 hours after seeding. Maximal effect was achieved with 10 units of IFN. Addition of specific anti-MuIFN-gamma or treatment of the preparation at pH 2 eliminated both antiviral activity and effect on spreading of MPM.

The effect of nutritional and thyroid state on the distribution of fatty acids between oxidation and esterification in isolated rat hepatocytes.

Small amounts of glycerol (0.25 mM) simultaneously decrease oxidation and increase esterification of fatty acids in isolated rat hepatocytes, mainly by raising the intracellular concentration of alpha-glycerophosphate. These effects were obtained in all thyroid states as well as in the fed and fasted states. In normal hepatocytes, the rate of oxidation showed a poor correlation with the content of malonyl-CoA. The effects of glycerol on fatty acid metabolism were most pronounced in hepatocytes with a high capacity for esterification and a previous low concentration of alpha-glycerophosphate (glucagon-treated hepatocytes from euthyroid fed rats). Esterification capacity was influenced by the nutritional and thyroid state (decreased in fasting and in hyperthyroidism). It was not influenced by glucagon in short-term experiments. alpha-Glycerophosphate may not regulate fatty acid metabolism in vivo, but a high esterification capacity probably traps acyl-CoA for esterification, thus preventing fatty acids from being oxidized. The metabolism of alpha-glycerophosphate seems to be accelerated by hyperthyroidism and slowed down by fasting. The effect of fasting seems not to depend on thyroid hormones for its effects on fatty acid metabolism in the liver.

Assessment of the anticholinergic effects of antidepressants in a single-dose cross-over study of salivation and plasma levels.

In order to evaluate the anticholinergic effect of antidepressant drugs, 11 healthy volunteers were given single oral doses of reference drug, test drugs or placebo on a double-blind basis at weekly intervals. The doses corresponded to average daily patient medications. Spontaneous whole mouth and parotid salivation, and plasma levels of drug and possible metabolites were measured 2, 6 and 10 h after drug administration. Moderate, statistically significant inhibition of salivation was found when nortriptyline, imipramine-N-oxide and mianserin were given. Less pronounced, but still statistically significant inhibition occurred after ingestion of nomifensine and zimelidine. The zimelidine effect was exclusively due to the metabolite norzimelidine, and the inhibition after imipramine-N-oxide was mainly due to the metabolite imipramine, but imipramine-N-oxide itself also had slight activity. Isocarboxazide and lithium had no effect on salivation. From these results and reported values of pharmacokinetic variables, the average level of anticholinergic activity during long-term treatment may be predicted: for mianserin and (nor-)zimelidine moderate inhibition of salivation, although less pronounced than with nortriptyline; for nomifensine no clinically significant effect; and for imipramine-N-oxide a negligible contribution from the unmetabolized drug.

Monoclonal antibodies to the h1 agglutinogen from Staphylococcus aureus 17A. Serological testing with type strains.

Four monoclonal antibodies to the h1 agglutinogen were produced by conventional means, and slide agglutination of S. aureus type strains was performed with protein A affinity purified IgG1 antibodies. In accordance with Oeding's serotype system the type strains 17A and 670 were strongly and consistently agglutinated. In addition, however, several of the remaining twelve type strains investigated showed varying reaction patterns. Our results indicate that the h1 agglutinogen may be more widely distributed among S. aureus strains than previously assumed.

Carnitine acetyltransferase. Effect of malonyl-CoA, fasting and clofibrate feeding in mitochondria from different tissues.

Malonyl-CoA sensitivity of carnitine acetyltransferase (EC 2.3.1.7) activities in isolated intact mitochondria from rat liver, heart and white and brown adipose tissue, and rabbit liver and heart were studied with a radioisotopic assay using [3H]acetyl-CoA. Malonyl-CoA inhibited [3H]acetylcarnitine formation in intact rat liver mitochondria (overt activity), while acetyltransferase in heart and white and brown adipose tissue were malonyl-CoA-insensitive. Acetyltransferase from rabbit liver and heart was only weakly inhibited by malonyl-CoA. Acetyltransferase in intact rat liver peroxisomes was malonyl-CoA-insensitive. Lysis of the mitochondria with (+)palmitoylcarnitine increased acetyltransferase activities (total activity) in all mitochondria studied and abolished malonyl-CoA-sensitivity, showing the necessity of intact mitochondrial membranes for malonyl-CoA-sensitivity. A differential increase in overt and total mitochondrial carnitine acetyltransferase was observed in rat liver upon fasting and clofibrate feeding. Fasting increased overt activity more than total activity and diminished malonyl-CoA-sensitivity. Clofibrate increased mainly total activity and also reduced malonyl-CoA-sensitivity. A similar pattern of increased activities was observed with palmitoyl-CoA and octanoyl-CoA as substrates.

Reduction of 4-nitroquinoline 1-oxide to 4-hydroxyaminoquinoline 1-oxide in lysates of cytomegalovirus-infected cells.

The rates of virus inactivation by 4-nitroquinoline 1-oxide (NQO) and 4-hydroxyaminoquinoline 1-oxide (HAQO) were compared and samples of cytomegalovirus (CMV)-infected cell lysates to which NQO had been added were examined for the presence of HAQO. These experiments demonstrated that (i) CMV inactivation by HAQO was more rapid than with NQO, (ii) virus inactivation by either NQO or HAQO failed to demonstrate a photodynamic component, and (iii) NQO-treated stocks contained HAQO, indicating reduction of NQO to HAQO. The results support the concept that metabolism of NQO to HAQO enhances the genotoxic effect of NQO.

Comparison of peripheral anticholinergic effects of antidepressants: dry mouth.

Twenty-one healthy volunteers were given single doses of placebo or antidepressants corresponding to average daily doses of patient medications. Spontaneous whole mouth and parotid salivation was measured two, six, and ten hours after drug administration in session 1; in session 2 after ten hours. The results indicate that a subdivision of antidepressants into four groups can be made according to inhibitory effect on salivation: (a) isocarboxazide and lithium citrate with no effect, (b) zimelidine and nomifensine with slight effect, (c) imipramine oxide and mianserin with moderate effect and (d) maprotiline, nortriptyline, clomipramine, imipramine, and amitriptyline with pronounced effect. Research implications and predictive value for new antidepressants are discussed.

Regulation of palmitate esterification/oxidation by glucagon in isolated hepatocytes: the role of alpha-glycerophosphate concentration.

Lipolysis was measured as the disappearance of [3H]glycerol previously incorporated into triacylglycerol, diacylglycerol and phosphatidic acid. There was no effect of glucagon on the lipolysis of any of these lipids. A transient increase in cellular alpha-glycerophosphate was induced by addition of glycerol during incubation. This resulted in an immediate and temporary decrease in oxidation and increase in esterification of palmitate while the uptake of palmitate from the incubation medium was unchanged. The change in alpha-glycerophosphate was also correlated with a transient drop in acyl-CoA and acylcarnitine. The lactate/pyruvate ratio was increased by the glycerol addition, but was still elevated for some while after the transient change in alpha-glycerophosphate. Similar effects were obtained by addition of dihydroxyacetone instead of glycerol. It is concluded that fatty acid esterification/oxidation can be changed by variations in the concentration of alpha-glycerophosphate, and that glucagon acts on lipid metabolism by decreasing the level of this metabolite.

Traffic accidents involving psychiatric patients. Characteristics of accidents involving drivers who have been admitted to Danish psychiatric departments.

A nationwide study of psychiatric in-patients driving motor vehicles and involved in accidents causing bodily injury in the years 1972--74 was undertaken. Compared with drivers in general involved in accidents causing bodily injury, drivers with a psychiatric record were characterized by an over-representation of women. A preponderance of psychiatric drivers in the age group 25--54 was observed. About 3% of drivers with a psychiatric record were involved in two or more accidents during the period studied. A greater proportion of the psychiatrically diseased group drove stolen vehicles, without valid licences, and were found not to have used safety belts at the time of the accident. Occurrence of injury was more frequent among the diseased drivers. They were more frequently alcohol intoxicated and with a higher blood alcohol level. The most commonly encountered diagnostic groups were personality disorder and transitory reactions.

Traffic accidents involving psychiatric patients. Description of the material and general results.

The present investigation analyzes the characteristics of traffic accidents involving psychiatric patients and comprises all persons who during the period 1970-74 have been admitted to a psychiatric in-patient institution and who during the period 1972-74 have been involved in a traffic accident causing bodily injury. Accidents with psychiatric patients involved constitute about 5.7% of all traffic accidents reported in Denmark. Compared with the total group of accidents, they are characterized by an over-representation of women and of persons in the age group 25-54 years. Accidents with psychiatric patients are over-represented during the period July-September, and in areas densely built up. The accidents tend to be more complicated, and the psychiatric patients are more likely to get injured and be hopitalized due to the accident. The investigation is based exclusively on routinely collected data. The limitations with which the results are encumbered and the problems concerning methodology and comparison of different studies are discussed.
