A brief history of printing of lithographic medical books in modern times.

A large number of lithographic medical books came out within about 100 years after the introduction of lithography technique from the west. There are about 1000 extant lithographic medical books among which 500 still extant as lithographic edition in our country. The development of lithographic medical book could be divided into three stages: the embryonic stage, the prosperous stage and the devlined stage. In spite of the shortcomings that the printing was not exquisite and the textual proofreading was not precise, the value of these lithographic medical books could not be underrated in preserving the medical documents, spreading the medical knowledge and promoting the confluence of TCM and western medicine.

[Effect of age on mineralization expression of periodontal ligament cell clones].

OBJECTIVE: To evaluate the effect of age on the expression of mineralized tissue progenitors in the peridontium and the effect of basic fibroblast growth factor (bFGF). METHODS: The periodontal ligament (PDL) tissues that derived from three old and 4 young donors were cultured and cells were cloned. The mineralizing ability were detected with or without bFGF treatment in the presence of dexamethasone. The cell cloning technic was employed in present study. RESULTS: The results indicated that the frequency of mineralized tissue progenitors (old donor: 4.1%) and the numbers of cell clones that mineralized with bFGF treated in the PDL cultures derived from old donors (3.4%) is considerably less than that of cultures obtained from young donors (17.7% and 16.9%). bFGF enhanced the mineralization of cell clone but not the parent population. CONCLUSION: It is concluded that the poor stem cell and progenitors in PDL and the low reaction of progenitors to growth factors in PDL are responsible for the limited regenerative capacity of periodontium in old patient.

[The role of basic fibroblast growth factor on expression of mineralization of periodontal ligament cell population and clones].

OBJECTIVE: To determine the effect of basic fibroblast growth factor (bFGF) on mineraliztion of PDL cell clone. METHODS: The PDL tissue that derived from three human donors was cultured and cells were cloned. The mineralizing ability were detected with or without bFGF treatment in the presence of dexamethasone. Cloned progenitor populations were assayed for their constitutive capacity to bind CAP and express ALP. RESULTS: The results show that bFGF increases clone mineralization (11%), but not in PDL cell population. The cell clones that mineralized only in the presence of bFGF have higher CAP binding ability and lower ALP expression compared with other mineralized clone cells. CONCLUSION: bFGF enhances mineralizing expression of PDL clone. The clones that mineralized only with bFGF treatment are different cell types from the bFGF-undependent mineralized clones.

[An experimental study on mechanism of intraocular pressure decline after allogeneic transplantation of trabecular meshwork].

OBJECTIVE: To study the mechanism of intraocular pressure (IOP) decline after allogeneic transplantation of trabecular meshwork (TM) in primate experimental glaucoma. METHODS: The experimental open-angle glaucoma models were created in 8 rhesus monkey eyes by repeated pangoniophotocoagulation on functional TM using argon laser. After 2, 3, 5 months respectively, avidin-gold or self-erythrocytes, which acted as tracers of the aqueous humor outflow, were perfused into the anterior chamber under normal pressure and volume conditions. 30 minutes later, 2.5% glutaraldehyde was perfused into the anterior chamber under the same conditions. Allograft TMs and photocoagulated area (serving as the control) were observed by light, transmission, scanning electron microscopy and ultrahistochemistry study. RESULTS: The IOP of 5 eyes of 6 experimental eyes became normal. A blood reflux into Schlemm's canal was seen by gonioscopy in 4 eyes. The surface of TMs from graft showed the normal uveal TM cords and intertrabecular spaces. Erythrocytes were found in the intertrabecular spaces and Schlemm's canal. Avidin-gold and vacuolar containing gold were observed in the cells of TM and Schlemm's canal. The TMs of photocoagulated area appeared to be fewer and narrower intertrabecular spaces. The uveal cords of this zone were confused. The content of acid mucopolysaccharides in the grafts was closer to normal than that in the photocoagulated areas. CONCLUSION: TM of allogeneic transplantation not only can survive but also function as an aqueous humor outflow pathway. We consider that this operative procedure can be used for treatment of some cases with glaucoma cautiously.

[Cell density and expression of proliferating cell nuclear antigen (PCNA) in the lens epithelium of children and aging patients with cataract].

OBJECTIVE: To determine the cell density and expression of proliferating cell nuclear antigen (PCNA) in the lens epithelium of children and aging patients with cataract. METHODS: The lens epithelial cells (LECs) of the central area were examined in two groups, children (less than 12 years old) and senile persons (51 - 80 years old). The densities of LECs in the two groups were calculated with samples stained with hematoxylin and eosin (HE) staining, and the PCNA was tested immunohistochemically. The integral optical density (IOD) of PCNA expression was measured through a medical color image analysis system. RESULTS: The densities of LECs in children and senile persons were 5,020.25 +/- 246.01 cells/mm(2) and 4,340.00 +/- 240.95 cells/mm(2) (P < 0.05), the IODs were 2.89 +/- 0.57 and 2.13 +/- 0.63 (P < 0.05) respectively. CONCLUSION: The LEC counting and IOD values in children eyes are significantly higher than that of senile persons, suggesting that they be one of the important factors in the formation of posterior capsular opacification (PCO) or after cataract in children eyes.

[Anti-invasion activity of several plant-originated anticancer drugs with different mechanism of action].

The antitumor and anti-invasion activities of taxol, harringtonine, homoharringtonine and camptothecin on highly metastatic melanoma B16-BL6 and human fibrosarcoma HT-1080 cells were reported in this paper. Results demonstrated that taxol, harringtonine, homoharringtonine and camptothecin exhibited significant inhibition of cell growth of B16-BL6 and HT-1080 cells. Taxol, harringtonine and homoharringtonine were also found to be effective for the inhibition of cell invasion and migration of B16-BL6 cells, but camptothecin showed basically no effect at the indicated concentration.

[Expression of human granulocyte-macrophage colony stimulating factor(hGM-CSF) by recombinant vaccinia virus and its effect on immunogenicity].

hGM-CSF was inserted into the down stream of P11K promoter of pJSB1175, which consists of HSV-1gD under the promoter P7.5K. The constructed plasmid pJSB1175D/GM-CSF was cotransfected with vaccinia virus Tiantan Strain into TK-143. After selection and screening, recombinant virus RVJSB11D/GM-CSF was harvested. It was proved that the recombinant virus could express GM-CSF and HSV-1gD at the same time by MTT assay and IFA in vitro. In vivo study showed that the expression of GM-CSF by RVJSB1175D/GM-CSF did not interfere with the HSV-1gD antibody production.

[YY1 and its repressive effect on human papillomavirus 16 early promoter P97 existed widely among human epithelial cell lines].

Cellular transcription factor YY1 represses the activity of early promoter P97 of HPV 16. Removal of the specific binding sites for YY1 in HPV 16 LCR can increase the P97 activity. To analyse the expression of YY1 and the existence of the activation effect on P97 due to removing YY1 binding sites in the LCR region, we extracted nuclear proteins from four cervical cancer cell lines and two human keratinocytes. EMSA assays were carried out in vitro. At the same time, we transiently transfected the luciferase reporter plasmids, which contain HPV 16 reference and mutated LCR sequences, respectively, into the different cells and determined the luciferase expressions. The results showed that all the analysed cell lines contained biologically functional YY1 proteins. No remarkable difference in the concentration of native YY1 proteins could be detected among the tested cells. Removal of YY1 binding sites in HPV 16 LCR could elevate P97 activity in several analysed cell lines, including normal human primary kerationcytes from foreskin. It suggests that YY1 regulatory system exists widely in human epithelial cell lines. We also found that the transcription activator NF1 was more highly expressed in C33a cell than in HT3 cell, indicating an unevenly distribution of functional proteins in various cell lines.

[Establishment and primary study of an assay for the detection of EB virus specific cytotoxic T lymphocytes].

In our study, a non-radioactive cytotoxicity assay by using a Balb/C (H-2d) mice, P815(H-2d) cells and lactate dehydrogenase (LDH) system was established, in order to detect EB virus specific cytotoxic T lymphocytes(CTLs). In this study, we detected both the changes of primary CTLs and secondary stimulated CTLs. Spleen CTL activity assay showed that the levels of primary and secondary CTLs in recombinant EBV-LMP1 vaccinia virus group were more higher than that in control group, at the same time, obvious enhancement of EBV-LMP1 specific CTL activity was observed in experimental group immunized by Baculovirus system expressing EBV-LMP1 protein when compared with the normal mice group. This work proved that the LDH assay can be used to detect cytotoxic T lymphocytes.

[Effects of artemisia capillaris on blood glucose and lipid in mice].

The effects of Artemisia capillaris Thunb (AC) on blood glucose and lipid in mice were studied and its effective mechanism was explained. The results showed that hyperglycemic effect in aloxan-induced mice was inhibited (P < 0.01); that hyperglycemica in DM mice was decreased (P < 0.05); but the change of fasting blood glucose (FBG) in normal and hyperlipidemia mice was not significant (P < 0.05); that the level of total cholesterol (TC) and/or atherosclerotic index (AI) was decreased (P < 0.01-0.05) and the content of high density lipoprotein cholesterol (HDL-C) was increased in the above different model mice (P < 0.01-0.05). These results indicated that the AC would have the similar hypoglycemic effect as biguanide-drugs, improve endogenous and exogenous metabolic derangement in blood lipid. These suggest that AC could be used to protect and treat DM and its chronic complication.

[A study on the spectra of L-fucose].

The concentration of L-fucose will extraordinarily increase in urine in patients with early stage of cancer. It seems that this is a significant clue for early phase diagnosis of cancer. Unfortunately the sensitive detection of L-fucose is difficult. Thus we carried out researches on IR-spectrum, Raman spectrum, ultraviolet absorption spectrum and ultraviolet fluorescence spectrum. We compared the results from patients and healthy subjects. It is confirmed that the spectroscopic method, especially ultraviolet fluorescence spectrophotometry, may be suitable for early stage screening of some kinds of cancer. This test is rapid, inexpensive and painless.
