[Purification of soluble egg antigen (SEA) 38 kDa molecule of Schistosoma japonicum and preliminary evaluation of its immunodiagnostic efficacy].

OBJECTIVE: To study the applicability of soluble egg antigen (SEA) 38 kDa molecule of Schistosoma japonicum in the immunodiagnosis of schistosomiasis. METHODS: The target immunodiagnositic antigen SEA 38 kDa molecule was separated and purified by SDS-PAGE and electroelution technique and evaluated by SDS-PAGE and Western blotting. To compare their immunodiagnostic efficacy, the purified SEA 38 kDa and SEA were used to examine the sera from 182 cases including 31 acute schistosomiasis patients, 31 chronic schistosomiasis patients, 60 normal human controls, 30 paragonimiasis patients and 30 clonorchiasis patients. RESULTS: The positive rates of purified SEA 38 kDa and SEA for acute schistosomiasis patients, chronic schistosomiasis patients, normal human controls, paragonimiasis patients and clonorchiasis patients were 90.3%, 90.3%, 1.7%, 0, 0 and 90.3%, 83.9%, 3.3%, 0, 0, respectively. The P/N ratio of purified SEA 38 kDa and SEA in immunodiagnosis of schistosomiasis by ELISA had been compared by two-sample t-test. It was demonstrated that there existed significant differences between purified SEA 38 kDa and SEA in the three groups of acute and chronic schistosomiasis patients and normal persons(P < 0.001). The P/N ratio of SEA 38 kDa in detecting the sera from schistosomiasis patients was obviously higher than that of SEA, but when SEA 38 kDa was used to examine normal human sera, the result was converse. CONCLUSION: The purified SEA 38 kDa has higher sensitivity and specificity and immunodiagnostic potential.

The effect of liposome encapsulated daunorubicin on rabbit eyes after extracapsular lens extraction.

PURPOSE: To investigate the effect of liposome encapsulated daunorubicin (DNR) on rabbit eyes when it was used in prevention of posterior capsule opacification (PCO). METHODS: The liposome encapsulated DNR was prepared by modified freeze-thawing method. Each eye was injected with 0.1 ml liposomes (0.2 mg/ml and 20 micrograms/ml DNR) into the capsular bag during the extracapsular lens extraction (ECLE) in 10 rabbit eyes respectively. The phosphate buffer solution (PBS) was injected as control. Besides biomicroscope observation and histology examination of all eyes, the concentration of DNA in aqueous humor was also determined by high performance liquid chromatography (HPLC). RESULTS: The morphology of liposome encapsulated DNR were similar to the blank liposome with round or ellipse shape. The encapsulated effeciency of liposome encapsulated DNR was 45.1%. The inflammatory response was much more severe both in 0.2 mg/ml and 20 micrograms/ml DNR group than the control after liposome injection. All eyes in DNR group were showed severe corneal edema and opacity and lasted for 4 to 8 weeks. Histopathological study revealed that lens epithelial proliferation occured by 4 weeks and Soemmering's ring developed by 8 weeks in control; However, in DNR group eye only a few LECs remained in the bag and there were obvious corneal edema and eosinophiles infiltration. Corneal endothelial cells were severely lost. CONCLUSION: The results suggest that liposome encapsulated DNR could inhibit metaplasia and proliferation of remnant lens epithelial cells after ECLE in rabbit eyes, but the severe toxicity must be avoided by further study.

[Shunt tube implantation combining amniotic membrane transplantation and implantation of Molteno implant for glaucoma after penetrating keratoplasty].

PURPOSE: To investigate the therapeutic effect of aqueous humor shunt implants with amniotic membrane transplantation on intractable glaucoma. METHODS: Glaucoma was induced in rabbits by the injection of alpha-chymotrypsin into the posterior chamber of the eyes. The rabbits were divided into four groups. Group A: control group, Group B: single shunt tube group, Group C: shunt tube with amniotic membrane transplantation group, Group D: shunt tube with amniotic supporter and amniotic membrane transplantation group. The intraocular pressure(IOP), histology and filterable ability of the tissue around the tubes were studied. The therapeutic effect of the three methods for the glaucoma was compared. From 1998 to 1999, 42 eyes of 41 patients with uncontrolled glacoma after penetrating keratoplasty were randomly assigned into two groups. One group (12 eyes) underwent implantation of shunt tube combining transplantation of amniotic membrane. The other group underwent implantation of a single plate Molteno implant. Clinical records were reviewed to ascertain postoperative IOP, visual acuities, number of medications. RESULTS: The IOP elevated after the operation and reached at the peak on the third day for all groups and then dropped slowly. The IOP was 33.34 +/- 5.54 mmHg (1 mmHg = 0.133 kPa) for Group A and 27.88 +/- 8.86 mmHg for Group B three months after the operation. There was no statistical difference between the two groups (P = 0.274). The IOP was 22.33 +/- 3.73 mmHg for Group C and there was statistical significant difference between Group C and Group A (P = 0.02) and no difference between Group C and Group B (P = 0.113). The IOP was 15.74 +/- 2.94 mmHg for Group D and there was statistical significant difference between Group D and Group A (P = 0.001) and Group B (P = 0.036). There was no difference between Group D and Group C (P = 0.09). The study of horseradish peroxidase penetrability indicated that there was peroxidase in the tissues around the tube with amniotic membrane transplantation and no peroxidase for simple shunt tube. The fibrous tissue near the tube was denser in simple shunt tube group than that in containing amnion groups. In clinic, the basic data existed no statistical difference between the two groups before surgery. The IOP was 54.42 +/- 9.65 mmHg in shunt tube with amnion group and 43.28 +/- 10.57 mmHg in simple plate Molteno implant group (P = 0.535) before operation. There was significant relativity of the visual acuity before and after the operation in two groups (r = 0.916, P = 0.002 and r = 0.962, P = 0.000). Most patients of the two groups had to use one or two anti glaucoma drugs. The overall success rates were 58.3% for shunt tube with amnion group and 66.7% for Molteno implant group (P = 0.73) within the follow-up period. Kaplan-Meier success analysis indicated one year cumulative success rates of 52.25% for shunt tube with amnion group and of 65.3% for Molteno implant group (P = 0.42). CONCLUSION: Amniotic membrane can inhibit proliferation of the scar around the shunt tube. Shunt tube implantation conbining amniotic membrane transplantation can significantly lowered the IOP of glaucoma. Combined using amnion supporter can expend shunt area and significantly reduce IOP of glaucoma. The effect of shunt tube implantation combining amniotic membrane transplantation has the similar result of single plate Molteno implant for the glaucoma after penetrating keratoplasty.

A comparison of the effects of two kinds of glass-ionomer cement on human gingival fibroblast attachment, proliferation and morphology in vitro.

The aim of this study was to compare the response of cultured human gingival fibroblasts to two kinds of glass-ionomer cement in vitro with regards to understanding the biocompatibility of these materials and their suitability for use as subgingival restorative materials. The glass-ionomer cements tested were Ketac Fil, and GC Fuji II. These cements were mixed according to the manufacturers' directions and condensed into tissue culture wells or cavities prepared on the root surfaces of extracted teeth. Healthy root slices and untreated tissue culture wells were used as controls. Several features such as attachment, proliferation and morphology of cells were studied. The results indicated that cells could not attach to the glass-ionomer cements unless they had been washed with distilled water and tissue culture media. No significant difference in initial attachment (1.5h) and long term attachment (48h) between the groups tested was noted. No significant difference was observed between levels of proliferation for human gingival fibroblasts grown on Ketac Fil, Fuji II, root slices and culture wells. No significant alterations of the cytomorphology could be detected in glass-ionomer cement compared with root slices. These results indicate that provided the surface is adequately washed, Ketac Fil, and Fuji II may be acceptable subgingival root surface restorative materials.

[Advances in the research of the peritoneal lymphatic stomata in human].

Peritoneal lymphatic stomata are small openings of the subperitoneal lymphatic vessels on the free surface of the mesothelium. The peritoneal cavity is connected with lymphatic system via these small openings which are considered to be the main passage-way that can absorb matter from the peritoneal cavity. The lymphatic stomata are claimed to be involved in many clinic procedures, such as ascites elimination; ultrafiltration failure on the continuous ambulatory peritoneal dialysis; metastasis of tumor cells from the peritoneal cavity, and so on. It was reported that the cellular factor-NO(i.e. endothelium-derived relaxing factor, EDRF) can enhance the patency of the stomata and lymphatic absorption of the stomata by stimulating guanylate way, then increasing the concentration of the cGMP, decreasing the concentration of the [Ca2+] and as a result diastole the lymphatic stomata. Some traditional Chinese medicines, which can enhance absorption of ascites, have a regulative function on the stomata by enhancing the NO concentration.

[Expression of IL-2 receptors on the surfaces of L1210 cells and bioactivity assay for IL-2-toxin fusion proteins].

OBJECTIVE: To confirm that L1210 cells surfaces express IL-2 receptors (IL-2R) and to establish experimental models for studying the cytotoxic effect of IL-2-toxin fusion protein in vivo and in vitro. METHODS: IL-2R on mouse leukemia L1210 cells was analyzed with immunofluorescence stain and FACS. The cytotoxic effects in vitro of these fusion proteins IL-2-PEZNM and IL-2-K-PEZNM were assayed by MTT method; mice ascitic L1210 leukemia was induced by i.p. L1210 cells and the therapeutic effect of IL-2-K-PEZNM was observed. RESULTS: 86.3% of L1210 cells expressed IL-2R on their surfaces and the relative fluorescence intensity was 51.55. Two fusion proteins IL-2-PEZNM and IL-2-K-PEZNM showed dose-related cytotoxicities to L1210 cells in vitro with the IC50 of 0.51 and 0.67 microgram/ml respectively. IL-2-K-PEZNM (5 micrograms/d) showed significantly inhibitory effect on L1210 cells and prolonged the survival time of mice by 78.6%. CONCLUSIONS: L1210 cells as a model system to measure the cytotoxic activity of IL-2-toxin fusion proteins both in vitro and in vivo is simpler, stabler, and safer model than those with other cells such as Hut102 and activated T lymphocytes.

[A clinic analysis of 62 male patients with lupus nephritis].

To study the difference between male and female patients with lupus nephritis(LN), we made a retrospective analysis. The clinical characteristics of 62 male patients with LN were compared with 62 female patients with LN. The result indicated the distribution of LN onset ages was different between male and female patients with LN. The incidence of LN was higher in the male than in female patients with SLE. Male patients were often misdiagnosed or diagnosed long after the onset of the disease. Arthritis and positive rate of ANA were less commonly occurred in female patients with LN. Other clinical and pathologic features of LN were similar in male and female patients. The response to treatment and prognosis of male patients with LN was relatively poor. So male patients with LN should be paid more attention to.

[High dose intravenous immunoglobulins in treating systemic lupus erythematosus-associated thrombocytopenia].

OBJECTIVE: To study the effect of high dose intravenous immunoglobulins(IvIg) in systemic lupus erythematosus (SLE)-associated thrombocytopenia. METHODS: A case-control study was undertaken to assess the effectiveness of different therapeutic regimens (oral prednisone, IvIg, bolus methylprednisolone(MP), and IvIg plus bolus MP) in improving SLE-associated thrombocytopenia. RESULTS: For SLE-associated thrombocytopenia, IvIg, bolus MP, and IvIg plus bolus IvIg significantly shorten the time to reach a peak platelet count, which was (6.2 +/- 4.4) days, (7.3 +/- 4.9) days, and (3.8 +/- 2.4) days respectively in comparison with oral prednisone, which was (31.0 +/- 17.8) days (P< 0.01); if high dose IvIg and MP in combination, the time was also shorter than bolus MP alone (P < 0.05). The elevation of platelet count in groups receiving IvIg, bolus MP, and IvIg plus bolus MP were significantly higher than as in group receiving oral prednisone therapy(P < 0.05). CONCLUSIONS: High dose IvIg is a useful therapy in improving SLE-associated thrombocytopenia.

The juxtamembrane domain of cadherin regulates integrin-mediated adhesion and neurite outgrowth.

Axons are guided along their trajectories during development by many different systems of adhesion, attraction, and repulsion. Thus, many distinct, and potentially competing, receptor systems respond to environmental cues, and the information must be coordinated inside the growth cone to ensure that extension follows the appropriate path. In this brief review we bring together two studies, each of which has defined different aspects of a pathway through which N-cadherin regulates beta1-integrin function allowing for coordinated responses to environmental cues during neurite extension. First we review progress in defining the binding to cells and the subsequent effects on adhesion and neurite outgrowth of the chondroitin sulfate proteoglycan, neurocan. Neurocan binds to a cell surface glycosyltransferase associated with N-cadherin (but not integrin), initiating a signal which results in loss of cadherin and integrin-function-suggesting that these two adhesion receptor systems engage in cross-talk, allowing coordinate regulation. Second, we review the use of "Trojan" peptides, peptides which mimic specific sequences in the cytoplasmic domain of N-cadherin attached to a cell permeation sequence, to reveal protein-protein interactions critical to cadherin-integrin cross-talk. One peptide mimicking a 20 amino acid sequence in the juxtamembrane region of N-cadherin has the same effect as neurocan, blocking both cadherin- and integrin-mediated adhesion and neurite outgrowth. Both neurocan and the peptide cause the release of the non-receptor tyrosine kinase Fer from the cadherin complex and its binding to the integrin complex. These data define an epigenetic pathway through which environmental cues are capable of coordinately regulating the activity of two developmentally important adhesion systems.
