Narcotic withdrawal like mouse jumping produced by amphetamine and L-DOPA.

Upward jumping in mice that resembled the narcotic-withdrawal syndrome was produced by a combination of amphetamine (4 mg/kg) and L-DOPA (400 mg/kg) administration. Neither drug alone caused that jumping. Pretreatment with haloperidol (0.16 mg/kg) or pimozide (0.08 mg/kg) but not with phentolamine (10mg/kg) effectively blocked amphetamine-dopa jumping. After DOPA there was an increase in the brain levels of DOPA, dopamine and norepinephrine. Amphetamine, ineffective in itself, enhanced DOPA-induced elevation of brain DOPA and dopamine without affecting brain norepinephrine levels.

Acquistion and extinction of a conditioned hyperthermic response to a tone paired with morphine administration.

Following a 7-day baseline period, five male hooded rats were injected daily with a constant dose (20 mg/kg) of morphine sulfate. Immediately following the injections they were taken to a sound-attenuated room and exposed to a 5,000 cps tone for 90 min. On certain predetermined days of the morphine treatment, rectal temperatures were measured before each injection and 30 min following the onset of the tone. Also, after the 11th, 19th, and 57th morphine-tone pairing, the rectal temperatures were measured before and after a saline injection which was paired with tone. On these days tone presentations by themselves were sufficient to cause a reliable increase in rectal temperature resembling that seen following an injection of morphine. When tone presentations were continued, without further pairing with morphine, the hyperthermic response to the tone was gradually extinguished. However, a significant increase in rectal temperature in response to the tone was again observed following a second set of five morphine-tone pairings.

Influence of experimental dietary conditions on hepatic enzymes of glutamic acid metabolism in rats.

The lowering levels of dietary protein induced a significant fall of some hepatic enzymes associated with glutamic acid metabolism. The changes were later normalised during dietary rehabilitation of the protein-deprived rats. The levels of these enzymes were found to be increased as compared to those observed after starvation, on feeding a carbohydrate-free, protein-rich diet or by the dietary supplementation by glutamic acid.

Aggression in mice after p-chloroamphetamine.

Two or 3 days after a single high-dose (20 mg/Kg) of p-chloroamphetamine (PCA) aggression was reliably observed in male mice. This agression was not produced when the same dose of d-amphetamine was injected or when PCA was injected one hour before testing. However, PCA produced an increase in locomotor activity one hour, but not 24 hours, after injection.

Reduction of morphine-withdrawal body shakes by a conditional stimulus in the rat.

Narcotic dependence in rats was established, in five days, by administering intravenous injections of morphine sulfate in increasing doses. From day six onward, dependence was maintained by administering a morphine injection (15 mg/kg) every 6 h throughout the day, and each of these injections was paired with a tone. After 16 morphine-tone pairings, test trials, in which the tone was presented in the absence of morphine, were instituted. These tone presentations significantly reduced morphine-withdrawal "wet shakes" in rats which received morphine-tone pairings, but not in rats which received control treatments.

The cardiovascular effects of ergometrine in the experimental animal in vivo and in vitro.

The cardiovascular effects of ergometrine have been examined using animal models in vivo and in vitro. In rats in vivo the pressor effects of a standard dose of ergometrine were found to be dependent upon the anaesthetic used for the procedure. The pressor action of ergometrine was greater when urethane, an anaesthetic which produces both sympathetic and parasympathetic acitivity, was used. The in-vitro experiments, using isolated rabbit aorta and portal vein, failed to explain the phenomenon; they suggested that ergometrine has a direct action on the alpha-adrenoceptor in rabbits. No evidence was found to suggest that ergometrine inhibits noradrenaline uptake or its release from the nerve terminals.
