[Recurrence-free phases and survival times in children with cytochemically varying acute lymphatic leukemias. Note on the prognostic evaluation of leukemias].

The recidive-free phases, survival times and the number of long surviging patients were recorded in 64 children with cytochemically differentiated ALL. A significantly more favourable course of ALL of PAS type could be observed as compared with the undifferentiated type and partially with the esterase type, too. According to the present findings the cytochemical examinations of ALL can be recommended in the initial phase of leukaemia as a prognostic evaluation.

[Behavior of thymus-arised lymphatic cells in the peripheral blood in acute leukoses].

On the basis of their ability of the spontaneous formation of rosettes by unsensibilised sheep erythrocytes the proportion of thymic lymphatic cells in 7 acute and 3 chronic lymphatic leukaemias was determined. Further observations on the content of T-cells in the peripheral blood were performed in leukoses, whereby the remission was begun with prednisone and cytostatics, under the immune stimulation with BCG during the remission and in 3 children with lymphogranulomatosis before and after splenectomy. The results of the acute stage of leucosis refer to the possibility of a prognostic evidence with the subclassification of the ALL. The T-cell-leukaemias revealed a particularly unfavourable course of disease. Cytochemically they were acute lymphatic leukaemias of undifferentiated type. In leucoses with smaller proportion of T-cells the blasts above all revealed a granular PAS-reaction.

Thyrotropin releasing hormone (TRH): restoration of oxotremorine tremor in mice. Comparison with quipazine, a serotoninergic and dopaminergic stimulant.

Both TRH and quipazine (2.5-25 mg/kg) were found to restore and to intensify the oxotremorine-induced tremor in mice when injected i.p. 60 min after oxotremorine 0.5 mg/kg s.c. This phenomenon does not seem to be due to an increase in body temperature or muscle tone. Also other dopaminergic drugs, e.g. amphetamine, methylphenidate, nomifensine and apomorphine had a significant but lesser effect than TRH or quipazine. Haloperidol and methysergide both antagonized the effect of quipazine but not that of TRH. Neither propranolol, phenoxybenzamine, alpha-methyl-p-tyrosine, nor p-chlorophenylalanine inhibited the activity of TRH or quipazine. The restoration of oxotremorine-induced tremor could be prevented by atropine but not by methylatropine. It is concluded that quipazine exerts its effect by direct stimulation of serotoninergic and dopaminergic receptors, whereas TRH receptors may represent separate entities and TRH may function as a neurotransmitter or neuromodulator.

[Results of polychemotherapy and immunotherapy in acute leukemias].

For the purpose of evaluating the efficiency of an unspecific immunostimulation in acute leukaemias the results of treatment obtained from two groups of patients (a total of 55 children) were compiled. In the first group an unspecific immunostimulation with vaccination (BCG, diphtheria-tetanus-pertussis, measles) could be observed after the induction of remission during a cytostatic maintenance therapy. In the second group a polychemical therapy and the CNS-irradiation was applied according to the treatment scheme developed by the working team of Donald Pinkel. The group of patients treated with unspecific immunostimulation involved a high percentage of surviving children. In total there was no essential difference between the treatment results of both schemes of therapy during our period of observation. As before, the treatment of hyperleukocytic forms of leukaemias will cause particular difficulties.

[Cytochemistry of leukocytes in children. II. Changes in cytochemical reactions of neutrophil granulocytes in the course of leukoses].

A report is presented on the changes of cytochemical reactions of neutrophilic granulocytes in children during acute leukaemia. Various hydrolases revealed a significant increase of activity during the acute stage of disease. The enzyme activity begins to decrease in remission, but regains its normal value only in the activity of alkaline granulocytic phosphatase. The glycogen content of granulocytes is reduced at the beginning of the disease and during the recidive as well as in the granulocytes of children affected with chronic myeloid leukaemia. The enhanced enzyme activity of granulocytes refers to the fact that the defence function of cells in acute leukaemias is not completely cancelled. Only a decreased glycogen content of cells refers to a disturbance in the metabolism of granulocytes.

[Cytochemistry of leukocytes in childhood. III. Changes of cytochemical reactions in lymphocytes in infections and during immunological reactions].

The present paper gives a report on changes of enzyme reactions, of the glycogen content, and the nucleolus picture in lymphocytes which are based on cytochemical investigations of blood smears taken from 110 children with different diseases. In 20 new-born babies the cytochemical responses of lymphocytes after triple immunization with Di-Pe-Te immunization matter were observed. The findings reveal significant changes to be found predominantly in the activity of alpha-naphthylacetate esterase, PAS-reaction and the nucleolus picture of lymphocytes in immunological reactions. No hints for specific immunological functions of lymphocytes could be detected. The changes may refer to B-lymphocytes and to T-lymphocytes.

[Cytochemistry of leukocytes in children. IV. The use of cytochemical tests in the differentiation and prognostic evaluation of acute leukemias].

From 63 children with acute leukaemia the bone-marrow smears were cytochemically examined before the beginning of therapy. The activity of peroxydase was examined according to Sato and Sekya, that of acid phosphatase according to Löffler and Berghoff, that of alpha-naphthyl-acetate-esterase according to Gomori; the evidence of glycogen was examined by means of the PAS-diastase response according to McManus. Among the 63 cases of leukaemia we found 6 cases of paramyeloblastic leukaemia, 2 cases of parapromyelocytic leukaemia, and 3 cases of myelomonocytic leukaemia. 52 cases of leukaemia could not be further differentiated in morphological respect. They represented an immature paraleukoblastic leukaemia. A division according to leading cytochemical criteria was made for them. The therapeutic possibility of influencing the various groups was checked by means of prolonged observations. Children affected with paraleukoblastic leukaemia of the phosphatase type had a significantly low rate of remission similar to the myeloid leukaemia. Paraleukoblastic leukaemia of the PAS type, esterase type and the undifferentiated type revealed no essential differences. The rate of remission, however, was highest in leukaemia of the PAS type amounting to 100%. In one part of patients the prolonged cytochemical observations in 8 children with recidives showed that the cytochemical type under chemotherapy was changed.
