[Quantitative DNA determination and its role in the prognosis of malignant tumors].

We summarize our own cytophotometric and flow cytometric measurements from malignant tumors and premalignant lesions. DNA, non-histoprotein and dry mass (by interference microscopy) have been assessed. Ploidy and the compartments of the cell cycle were evaluated. For most tumors (cervix, breast, ovary, melanoma, larynx, oro-facial, bladder) a bad prognosis was suggested by the following cytophotometric findings: aneuploidy, polyploidy, and/or a high proportion of S-G2/M nuclei. This has been statistically confirmed for breast cancer by case studies conducted over a period of five years. Among the changes associated with lung cancer were alterations in protein content. Cytophotometric findings can provide useful additional information concerning a patient's prognosis.

[Impulse cytophotometry and catamnesis in breast neoplasms].

CEll suspensions from 62 cancers of breast were investigated by flow cytophotometry after staining with pepsin-Ethidium bromide. In all patients a radical operation was made, followed by radiotherapy. The surviving patients were observed for 5 years. From the DNA-histograms 48% of the tumours were classified as diploid (or peridiploid), 40% as polyploid and 11% as aneuploid. Concerning ploidy a rank with worsening prognosis was found from diploid over polyploid to aneuploid cancers. In this sense bad prognosis means: dead or relapse during 5 years, histologically undifferentiated tumours, great primary tumour, metastasis in lymph nodes. In diploid tumours bad prognosis is indicated by a higher part of S- and G2-phase nuclei in the flow histograms. Most of the findings, but not all, are proved statistically. Now as ever, histological differentiation is still the most easily statement for prognosis.

[DNA content and sulfurous nuclear proteins of Ehrlich ascites tumor cells in relation to growth].

The proliferative behaviour of the Ehrlich ascites tumor in dependence on age after transplantation is characterized by pulse cytophotometrical DNA measurements as well as by determination of the sulfhydryl groups of the nuclear proteins. With increasing tumor age the DNA measurements show a decrease of the percentage of cells in S phase. During the first days of growth increases the percentage of cells in G1 phase. The percentage of cells in G2 and M phases increases strongly in the later days of growth. By the reaction of protein thiols with DDD and Fast Blue B cells with higher and lower concentration of SH groups of the nuclear proteins can be distinguished in two groups. By succeeding analysis of one and the same cell could be proved that the cells with higher SH concentration also possess a higher concentration of nuclear dry mass. On the basis of our own investigations and of data in the literature is suggested that the cells with higher nuclear SH concentration represent the visualized proliferating compartment of the tumor cell population.

[Flow cytophotometry and cell cycle analysis].

By flow cytophotometry it is possible to obtain a DNA frequency distribution of cell nuclei. The curve is the result of the superposition from the 3 compartments: G0 + G1, S and G2 + M. Basing on a model, published formerly we simulated curves with different parameters. For our model we made the following assumptions: The G1- and the G2-peak are log-normal Gaussian distributions with the same coefficient of variation. The superposition of the 3 compartments is made after logarithmic classification and standardisation to the normal distribution. A program was developed for iterative approximation of the simulated curve to the measured values from the flow histogram. 7 points are needed for approximation and the standard deviation in changed step by step. In this way the moiety of the 3 compartments can be obtained. By cytophotometry it is only possible to estimate the relation between the number of cells in the 3 phases. The relative length of a cycle phase influences the shape of the DNA histogram, e.g. a relative prolongation of the G2-phase produces an increase of the G2-(4c)peak. The relationship between cell cycle and DNA histogram is influenced additionally by the cell kinetic in a different way in steady state or in exponential growth. By new measuring and staining methods it is possible to differentiate between several compartments, e.g. between G0 and G1. The flow cytophotometry is a valuable method to gain information about the cell kinetic.

Cholesteryl esters in pig liver during atherogenic diets and foreign serum protein infusions.

Eight-month-old pigs were fed a diet supplemented with 15% lard (group II) or 15% lard plus 1.5% cholesterol (group III) for 4 months. Five of the animals fed the high fat and cholesterol diet received two i.v. infusions of foreign serum protein (group IV). One group of animals was treated with the same infusions without dietary fat or cholesterol supplement (group V). Liver lipids and serum cholesterol levels were determined. In group II the concentration and fatty acid composition of hepatic cholesteryl esters showed no marked differences from controls (group I). The same findings were present in 2 of 5 animals of group III; in the other 3 animals higher hepatic cholesteryl ester concentrations and predominantly an elevation of monoenoic acid fraction, in one case also a rise of trienoic acid fraction were obtained. These alterations were found in the liver of all animals of group IV. The reverse was observed in the animals which were given the same serum infusions but only the stock diet (group V): a reduction of hepatic cholesteryl ester concentration and no striking alteration in fatty acid pattern. In cryostat sections of all liver specimens no stainable lipid could be detected. There was no correlation between liver and serum fatty acid composition of cholesteryl esters. The total cholesterol levels in serum showed no increase in group V, a slight increase in group II, a moderate increase in group IV, and a marked, partly large increase in group III. These results are discussed in relation to the effect of cholesterol and fat feeding on cholesterol metabolism and essential fatty acid requirement.

[Biomorphosis of arteries].

The discrimination between normal aging and disease is very difficult, especially in the vascular system. The main factors, gene expression, physical forces, and environmental conditions are effective in normal and pathological circumstances. The changes in extracellular and cellular level are described. During aging, a fibrosis and a reduction of the cellular functions in the vascular wall are found. This deteriorates the blood streaming and favours pathological processes, e.g. atherosclerosis.

Flow-through fluorocytophotometry of different brain tumours.

The flow-through cytophotometric method was used to investigate the single-cell DNA content in 60 tumours of the CNS and allied structures (9 meningiomas, 5 ependymomas, 15 astrocytomas, 11 anaplastic astrocytomas, 8 glioblastomas, 7 medulloblastomas, 2 oligodendrogliomas, 1 anaplastic oligodendroglioma, and 2 neurinomas). The cytophotometric parameters were correlated with morphological and clinical data of the tumours. The results are summarised in Tables 1--7. With regard to the DNA content of their cell nuclei, the gliomas present a behaviour similar to tumours in other organs, but in the present study the cytophotometric signs of malignancy in gliomas are not so evident as in carcinomas or sarcomas. The information obtained by flow cytophotometric methods may be helpful in diagnosing the degree of malignancy in brain tumours.

Major venous thrombosis: a complication of transvenous pacemaker electrodes.

Two patients with transvenous permanent pacemakers developed thrombosis and occlusion of the superior vena cava around a broken pacing wire, with clot extending into the innominate, subclavian, and axillary veins. Both patients had symptoms and signs of the superior vena cava syndrome which failed to resolve despite anticoagulant and thrombolytic treatment. Obstruction to venous flow, however, was relieved in large measure by the development of collateral venous channels. One of the patients has shown mild symptoms for two years and the other for two months of follow-up. It is suggested that anticoagulant prophylaxis should be given to all patients with a broken wire which cannot easily be removed.

[Interference microscopic determinations of dry mass of cell nuclei of normal and dysplastic epithelium, carcinoma in situ and cervical carcinoma cultivated in vitro (author's transl)].

Dry mass of cell nuclei was studied in 34 explant cultures of normal epithelium (8), persisting dysplasias (7), Ca in situ (9) and cervical carcinomas (10) by interference microscopic determination. Dysplasias, Ca in situ and carcinomas can be distinguished from normal epithelium by a significantly higher dry mass value. Such a difference does not exist between dysplasia and Ca in situ. The percentage of dispersion of the dry mass is significantly higher in carcinomas than in all other examined tissues. The biological meaning of these results is discussed. The interference microscopic determination of dry mass of the cell nuclei is a suitable parameter for the characterization of properties of cervical carcinomas and their intraepithelial prestages cultivated in vitro.
