Diagnosis of CNS lymphoma using immunofluorescent phenotyping of CSF mononuclear cells.

We describe the use of a panel of monoclonal antibodies, directed against leukocyte surface antigens to characterize CSF mononuclear cells with regard to malignancy when cytopathology was inconclusive. Cytocentrifuged preparations from three patients in which traditional modalities had not yielded a diagnosis were studied, utilizing a panel of antibodies for B and T cell antigens. All three patients were found to have B cell lymphoma of the CNS. Rapid institution of the appropriate therapy resulted in marked improvement of CNS symptoms in each case. Our results indicate that in patients with CNS disease and CSF pleocytosis of undefined nature, this technique may provide rapid and precise diagnostic information.

Genetic and environmental determinants of 17 serum biochemical traits in Brazilian twins.

The genetic and environmental effects on the levels of 17 serum biochemical quantitative traits (calcium, phosphorus, glucose, urea nitrogen, uric acid, total protein, albumin, total bilirubin, alkaline phosphatase, lactate dehydrogenase (LDH), glutamic-oxaloacetic transaminase (SGOT), total lipid, cholesterol, triglyceride, alpha-lipoprotein, pre-beta-lipoprotein and beta-lipoprotein) were estimated in 105 pairs of healthy twins of both sexes (57 MZ and 48 DZ) by path analysis. The genotype effect h2 was significant for all traits (P less than 0.001) and its value extended from 0.52 (alpha-lipoprotein) to 0.81 (alkaline phosphatase), whereas environmental effect b2 was significant (P less than 0.05) in only 10 traits of the 17 analyzed, with the maximum value of 0.13 (cholesterol). Correlations between genotypes of paired traits were estimated and, of 136 values, 47 were significant at the 5% levels, thus indicating partial and common genetic mechanisms.

Familial determinants of blood pressure in northeastern Brazil.

Genetic heritability in this triracial population is 0.41 for systolic pressure in children, 0.14 for systolic pressure in adults, and 0.34 for diastolic pressure in both generations. Cultural inheritance is much smaller, and there is no evidence of maternal effects or major loci.

The effects of inbreeding and of some genetic polymorphisms on blood pressures, pulse rate and hematocrit in Northeastern Brazil.

The possible role of genetic mechanisms, as revealed by inbreeding depression and pleiotropic effects of the ABO, Es D and CA II loci, on blood pressures, pulse rate and hematocrit, was studied in a sample of 7,642 migrant Brazilian individuals of rural origin. It was not possible to confirm previous claims of the effects of ABO blood groups system and inbreeding on diastolic blood pressure. On the other hand, a significant inbreeding depression on pulse rate of about 1.23 bmp/10% F, among adult individuals, was revealed. The observed significant effects of several markers on hemodynamic variables, due to its number, were attributed to chance.

Studies of globin chain synthesis and globin mRNA content in a patient homozygous for hemoglobin Lepore.

Globin chain synthesis and globin mRNA content were studied in blood cells of a patient homozygous for Hb Lepore. Peripheral blood cells incubated with tritiated leucine synthesized approximately 1.5 to 3% as many Lepore globin chains as alpha chains. Globin mRNA in peripheral blood cell RNA was assayed by molecular hybridization assays using human alpha and beta cDNA, and the results indicated the presence of approximately 1% to 2% as much beta-like mRNA (presumably deltabeta Lepore mRNA) as alpha mRNA. The amount of Lepore deltabeta chain mRNA in peripheral blood cells is therefore proportional to the amount of Lepore globin chain synthesis in the same cells. An incidental observation was the finding that peripheral blood cell RNA of this patient, at a time when she was being heavily transfused, contained substantially higher levels of beta-like mRNA (relative to alpha mRNA) than in subsequent studies. Cell-free translation of this mRNA however revealed that it contained authentic beta chain mRNA which must have been derived in some way from the transfused blood cells.

Linkage data on MN and the Hb beta locus.

A sample of 28 informative families was studied for linkage between Hb beta and MN. Values of the neuterized recombination fraction from these and other families from the literature excluded a recombination fraction of less than .30 between these loci. Our results support different recombination values for males and females (theta equals .34 abd .50, respectively). A simple approach to estimate the sample size required as well as a study of the relationship between sibship size and sample size under conditions of loose linkage are also presented.

Studies on a circulating anticoagulant in systemic lupus erythematosus: evidence for inhibition of the function of activated plasma thromboplastin antecedent (factor XIa).

Plasma proteins which interfere with blood coagulation have often been described in patients with systemic lupus erythematosus (SLE). The most frequent type interferes with the conversion of prothrombin to thrombin and thus prolongs the prothrombin time. Infrequently, SLE patients exhibit anticoagulants which appear to block the earlier stages of coagulation such as those involving factor VIII or the formation of activated factor XI (factor XIa). The anticoagulant reported here was studied by means of a sequential clotting system utilizing crude coagulation factors and was noted to interfere with the action of activated plasma thromboplastin antecedent (PTA) during the activation of factor IX. This anticoagulant was found in gamma-globulin-rich ethanol fractions of plasma. After gel filtration, it was found principally in fractions containing IgM globulins but also, to a lesser extent, in IgG-rich fractions. In this respect, it is similar to anticoagulants reported in certain other cases of SLE. Attempts to confirm the immunoglobulin nature of the anticoagulant by immunoabsorption were, however, inconclusive
