Cerebral amyloid angiopathy associated with massive intracerebral hemorrhage.

Characteristic computed tomographic, clinical and pathologic features are discussed in two patients with cerebral amyloid angiopathy (CAA) associated with massive intracerebral hemorrhage. Amyloid angiopathy should be considered in any elderly patient in whom intracerebral hemorrhage occurs at an atypical site. Cortical involvement, multiplicity of hemorrhages, bilaterality, and repeated episodes should further arouse diagnostic suspicion. Clinical management is confined to prevention of systemic hypertension.

[Autoradiographic determinations of carbon-14-labeled monomers and residual monomers in the tissues of Wistar rats after implantation of cold-polymerizing polymethyl methacrylate (bone cement) (author's transl)].

Self-setting acrylics, named bone cement, are taken for the reconstruction of bone defects, for the fixation of endothesis and fractures. A disadvantage of these rapid-cure acrylic compounds is the toxicity of its monomer. Infraredspectrographic measurements showed an absolute content of 12,3 per cent monomer in an acrylic-resin-compound-implant. In order to perseive the incorporation of the diffused monomer after implantation of bone cement in rats, the monomer were marked with 14C (radioactive carbon). The autoradiographic examination revealed the excretion of the marked monomer in the urine and an accumulation in kidney, liver and lungs. The results were demonstrated.

The cellular prion protein binds copper in vivo.

The normal cellular form of prion protein (PrPC) is a precursor to the pathogenic protease-resistant forms (PrPSc) believed to cause scrapie, bovine spongiform encephalopathy (BSE) and Creutzfeldt-Jakob disease. Its amino terminus contains the octapeptide PHGGGWGQ, which is repeated four times and is among the best-preserved regions of mammalian PrPC. Here we show that the amino-terminal domain of PrPC exhibits five to six sites that bind copper (Cu(II)) presented as a glycine chelate. At neutral pH, binding occurs with positive cooperativity, with binding affinity compatible with estimates for extracellular, labile copper. Two lines of independently derived PrPC gene-ablated (Prnp0/0) mice exhibit severe reductions in the copper content of membrane-enriched brain extracts and similar reductions in synaptosomal and endosome-enriched subcellular fractions. Prnp0/0 mice also have altered cellular phenotypes, including a reduction in the activity of copper/zinc superoxide dismutase and altered electrophysiological responses in the presence of excess copper. These findings indicate that PrPC can exist in a Cu-metalloprotein form in vivo.

Immunhistological evaluation of Creutzfeldt-Jakob disease with reference to the type PrPres deposition.

German patients suspected of having Creutzfeldt-Jakob disease (CJD) and related diseases were studied pathologically. The immunohistochemical findings after hydrolytic autoclaving pretreatment sensitively detected the synaptic-type deposition of the protease-resistant isoform of the prion protein (PrPres which thus served to establish the consensus diagnosis of CJD.
