Polysaccharide produced by the genus Pullularia. II. Trans-alpha-glucosidation by acetone cells of Pullularia.

Acetone cells of Pullularia sp. were incubated with maltose, and the saccharides produced were fractionated on a charcoal column. The fractions were subjected to paper-chromatographic analysis and enzymatic assay. By these methods, the saccharides have been shown to include glucose, maltose, maltotriose, panose, and the lower members of glucose polymers containing the 1,4- and 1,6-alpha-glucosidic linkages, but neither isomaltose nor dextrantriose. The use of glucose oxidase results in the formation of the higher members of glucose polymer.

Thermodynamic databases for proteins and protein-nucleic acid interactions.

Thermodynamic data regarding proteins and their interactions are important for understanding the mechanisms of protein folding, protein stability, and molecular recognition. Although there are several structural databases available for proteins and their complexes with other molecules, databases for experimental thermodynamic data on protein stability and interactions are rather scarce. Thus, we have developed two electronically accessible thermodynamic databases. ProTherm, Thermodynamic Database for Proteins and Mutants, contains numerical data of several thermodynamic parameters of protein stability, experimental methods and conditions, along with structural, functional, and literature information. ProNIT, Thermodynamic Database for Protein-Nucleic Acid Interactions, contains thermodynamic data for protein-nucleic acid binding, experimental conditions, structural information of proteins, nucleic acids and the complex, and literature information. These data have been incorporated into 3DinSight, an integrated database for structure, function, and properties of biomolecules. A WWW interface allows users to search for data based on various conditions, with different display and sorting options, and to visualize molecular structures and their interactions. These thermodynamic databases, together with structural databases, help researchers gain insight into the relationship among structure, function, and thermodynamics of proteins and their interactions, and will become useful resources for studying proteins in the postgenomic era.

Biochemical characterization and biologic actions of two toxins (D-1 and D-2) from Clostridium difficile.

Two toxins were isolated from a toxigenic strain of Clostridium difficile. The toxins were purified by gel filtration and ion-exchange column chromatography to homogeneity as judged from polyacrylamide gel electrophoresis and were designated D-1 and D-2. Toxin D-1 was lethal for mice, increased vascular permeability, and induced fluid accumulation in ligated rabbit ileal loops, and toxin D-2 displayed cytotoxicity in HeLa cells with a minimum of 1 pg of toxin. The molecular weights of toxins D-1 and D-2, as estimated by gel filtration, ranged from 550,000 to 600,000 and from 450,000 to 500,000, respectively. These toxins were heat labile and inactivated by pronase and trypsin. Amino acid analyses of both toxins showed them to be of relatively similar composition. Antisera prepared against purified toxin D-1 neutralized all of the biologic activities of toxin D-1, whereas it did not affect any of the biologic activities of toxin D-2. A sensitive latex-agglutination immunoassay was developed for screening for C. difficile toxin D-1 in patients with pseudomembranous colitis.

Pachymeningitis in microscopic polyangiitis (MPA): a case report and a review of central nervous system involvement in MPA.

A case of microscopic polyangiitis (MPA) with pachymeningitis is described. The patient had renal, skin, gallbladder and peripheral nervous system involvement, simultaneously with pachymeningitis. Necrotizing glomerulonephritis with crescent formation, and necrotizing small vessel vasculitis in the kidney and skin were confirmed by biopsy. A highly elevated titer of antineutrophil cytoplasmic antibody for myeloperoxidase (MPO-ANCA) was observed. All of the clinical and laboratory abnormalities improved with high-dose pulse and conventional steroid therapy. The literature on central nervous system involvement in MPA and perinuclear-ANCA (p-ANCA)-related vasculitis is reviewed. This case serves to emphasize that pachymeningitis can occur as one of the features of MPA.
