Serum isoamylases in cystic fibrosis.

Salivary and pancreatic isoamylases were determined in the sera of 33 patients with cystic fibrosis (CF) and in 34 CF-parents. Pancreatic serum isoamylase activities were greatly decreased in patients with CF, and only two values from this group fell within the normal range. Salvary isoamylases, however, were markedly increased so that the total serum amylase activities were normal. We interpret the low pancreatic isoamylase levels in serum to reflect reduced exocrine masses in the pancreas of CF-children. In heterozygotes, the mean activity of the pancreatic isoamylase was significantly higher than in the reference group, while salivary isoamylases were within the normal range. Due to a genetic polymorphism pancreatic isoamylase may occur in two main fractions. This variant type of pancreatic isoamylase appeared more frequently in CF-heterozygotes than was expected from a reference group of blood donors.

Screening for cystic fibrosis by analysis of albumin in meconium.

A clinical study of the albumin content in meconium was performed on two categories of newborn infants: a screening series of 8,830 infants and a high-risk group for Cystic Fibrosis (CF) of 70 infants. A single radial immunodiffusion technique and test strips were used. Three CF infants were detected in the screening series (1:3,000) and 16 in the high-risk group. The diagnostic accuracy for CF was fairly good. The specificity was 99.8% for the immunodiffusion technique and 99.2% for test strips. A high concentration of albumin in meconium was found not only in CF but also in preterm babies and infants with gastrointestinal disturbances, such as atresias, malaena neonatorum and malabsorption syndromes. The sensitivity was 90% for the immunodiffusion technique and 78% for the test strip. False-negative results were probably due to proteolytic activity and might be avoided if the samples are stored at a low temperature before analysis. CF screening of all meconiums by the use of test strips followed by analysis of positive tests by the immunodiffusion technique is suggested.

Cystic fibrosis detection by means of a test-strip.

The effectiveness of meconium screening for albumin as an indication of cystic fibrosis is examined. BM-Test Meconium was applied to 69,000 investigations. In 60 positive tests, cystic fibrosis was confirmed later. No increased albumin content was observed in four cases of cystic fibrosis.

Electrolyte redistribution in cystic fibrosis fibroblasts studied by electron probe X-ray microanalysis.

The elemental composition of fibroblasts from cystic fibrosis (CF) patients and from healthy controls was compared by means of electron probe X-ray micro-analysis. Significantly lower sodium levels (p < .01) and higher calcium levels (p < .02) were found in CF fibroblasts, indicating a disturbance of ion regulation in these cells. Treatment with Staphylococcus aureus protein A and gammaglobulin (proteins that may influence the ciliotoxic CF factor) did not change the elemental composition of the fibroblasts.

Analysis of ejaculates from patients with cystic fibrosis.

Ejaculates from two men with cystic fibrosis (CF) were examined. Both had azoospermia. A considerable decrease in volume and fructose content was noted. The absolute amounts of calcium, magnesium, and zinc per ejaculate showed normal values in one of the patients but were two to three times increased the the other compared to mean values of a control group. Thus the concentrations of these cations were increased at least fivefold in both patients. The amount of Mg(2+)-and Ca(+)-dependent ATPase was comparable to that of controls, but values were higher than in men with oligospermia. Both the divalent cations and the Mg2+- and Ca2+-dependent ATPase curve profiles of split ejaculate fractions were atypical. Secretory granules and vesicles were plentiful in the seminal plasma of both patients while amorphous substance was practically absent. The present findings agree with a less affected function of the prostate gland and a dysfunction of the seminal vesicles in these patients.
