Pyridoxal 5'-phosphate binding site of pig heart alanine aminotransferase.

After borohydride reduction, carboxymethylation, and tryptic digestion of the holoenzyme of pig heart alanine aminotransferase, a single icosapeptide containing the N6-(phosphopyridoxyl)lysine residue was isolated by a combination of gel filtration and ion-exchange chromatogrpahy. Its primary structure was determined as Gln-Glu-Leu-Ala-Ser-Phe-His-Ser-Val-Ser-Lsy(Pxy)-Gly-Phe-Met-Gly-Glu-Cys-Gly-Phe-Arg.

Immunofluorescent and biochemical studies on tyrosine hydroxylase and dopamine-beta-hydroxylase of the bullfrog sciatic nerves.

Immunofluorescence specific for tyrosine hydroxylase (TH) or dopamine-beta-hydroxylase (DBH) was accentuated in both proximal and distal segments of the sciatic nerve after ligation. Estimations of the enzyme activities confirmed the above results. Mean axoplasmic flow rates of TH and DBH in bullfrog sciatic nerve were found to be 8 and 123 mm/day, respectively. They were decreased by colchicine or by cold temperatures (4 degrees C).

Neutral lipid and sphingolipid composition of the brain of a patient with membranous lipodystrophy.

Lipids were extracted from brains of a patient with membranous lipodystrophy (ML) and three normal patients and the neutral lipid and sphingolipid constituents were investigated. The storage of a large amount of free fatty acid was observed in the ML brain, but no cholesterol ester was found. Total lipid, cholesterol and cerebroside contents were slightly decreased in the white matter of the ML brain. The composition of free and sphingolipid fatty acids did not differ between ML and normal brains.

Uptake, distribution, and elimination of sodium linear alkylbenzene sulfonate and sodium alkyl sulfate in carp.

Uptake, distribution, and elimination of labeled surfactants, 35S-labeled sodium laurylbenzenesulfonate (35S-C12-LAS) and 35S-labeled sodium lauryl sulfate (35S-C12-AS), were investigated in carp exposed to a low concentration level of the respective solutions found in river water in Japan by whole-body autoradiography and liquid scintillation counting. Whole-body autoradiograms showed that the fish absorbed 35S-C12-LAS and 35S-C12-AS from the solutions principally through gills and that the surfactants absorbed were rapidly distributed throughout the body tissues and organs. High concentrations were found in gills, blood, hepatopancreas, kidney, and gall bladder. The elimination in the fish that were kept in clean fresh water after exposure were relatively rapid in most tissues and organs except for gall bladder. These results suggest that the surfactants absorbed in fish may have an adverse effect on many tissues and organs of fish. What influenced the distribution profile and concentration factors of the surfactants in tissues and organs of carp were the exposure time and the molecular structure of the surfactants.

Psychotropic agents. 3. 4-(4-Substituted piperidinyl)-1-(4-fluorophenyl)-1-butanones with potent neuroleptic activity.

A series of 1-(4-fluorophenyl)-4-(1-piperidinyl)-1-butanones substituted with benzimidazole, benzotriazole, or quinoxaline at the 4 position of the piperidine ring was synthesized and subjected to neuroleptic tests. Neuroleptic activities of several compounds were comparable to those of haloperidol. In particular, 4-[4-(2,3-dihydro-2-thioxo-1H-benzimidazol-1-yl)-1-piperidinyl]-1-(4-fluorophenyl)-1-butanone (10) was characterized by having a potent neuroleptic activity with less liability to the extrapyramidal side effect.

Assessment of field dependence in young children.

In a series of exploratory studies to develop instruments for measuring field dependence in young children, a total of 312 Japanese middle-class children, ages 5 to 6, served as subjects. Without changing the essential nature of the original test, the geometric embedded-figures test attained relatively high internal consistency (KR-20 in the .80's) and stability (R = .83 to .52, at intervals of 1.5 to 13 mo.). Correlation pattern between the Embedded Figures Test and Wechsler subscales similar to that of older subjects was influenced by the speed factor in embedded figures and Wechsler performance scales. Correlation between Kato's portable Rod-and-frame test and embedded figures was significant only in boys. In addition, the correlation pattern between the rod and frame and the preschool Wechsler was not consistent with Witkin's conceptualization. A newly developed darkroom Rod-and-Frame Test correlated with embedded figures for both sexes. Additional analyses indicated the need for more refined instructions to subjects regarding the concept of upright. Some children had more than one anchoring point for responding. Nyborg's scoring method and the relationship of Piagetian spatial concept to rod-and-frame performance are discussed.
