A method for the routine clinical examination of serum gliclazide by high-performance liquid chromatography (HPLC) on a column packed with a macroporous anion-exchange resin, Diaion CDR-10, was developed. The elution was performed with acetonitrile-methyl alcohol-1.2 M ammonium perchlorate (4:3:7, v/v/v) at a flow-rate of 0.4 ml/min. The retention time of gliclazide was 15 min. It seems that the retention mechanism of gliclazide under the HPLC conditions described is not only ion-exchange mode but reversed-phase mode between the anion-exchange resin and the mobile phase. The detection limit of gliclazide was 0.2 microgram/ml in plasma. The coefficient of variation for the within-day assay was 5.0% (0.2 microgram/ml, n = 8). The decay curve of serum gliclazide in diabetic patients was determined.
In order to examine the effect of cemented tungsten carbide dust on the animal lung, saline suspensions were intratracheally administered into the lungs of rats in a single dosage. About one-fifth of the animals died during the first three days. The acute response of the lungs was hemorrhagic edema with intense alveolar congestion. The animals killed at six months all presented pulmonary lesions of patchy fibrosis in the vicinity of the deposited dusts, occasionally associated with focal traction emphysema and bronchobronchiolar ectasia. At twelve months, two-third of the animals had neither fibrosis nor dust deposition, although the remaining animals showed pulmonary lesions similar to those seen in the six-months responders. Fibrosis of the lungs seemed to consist of collapsed alveoli with condensation of the preexistent reticulin fibers, but without noticeable collagenization. It is supposed that both the early toxic and the late fibrogenic effects of the carbide dust are attributable to the cytotoxic action of cobalt present in the dust particles. It is possible that recovery of the pulmonary lesions results from removal of the dusts from the lesions.

A low molecular inhibitor (LMW-INH) of the alternative pathway activation was isolated form healthy human urine. Its molecular weight was slightly higher than 1000. LMW-INH inhibited C3 convertase formation in fluid phase, on sheep erythrocytes and on zymosan particles. In contrast with beta 1H globulin LMW-INH showed no effect on the C3b binding site for B, and it inhibited the activation of CVF.B complex by D only when LMW-INH was simultaneously present with D. These results indicate that the reaction mechanism of LMW-INH is different from that of beta 1H globulin.

Low serum CH50 (s-CH50) levels were found in 16 out of 574 patients with primary renal disease. However, five of these 16 patients had normal plasma CH50 (p-CH50). The most characteristic finding in the complement profile in these sera was a marked reduction in C4 and C2 hemolytic activities with a minimal reduction in C3 and C5 activities. Sera separated at 37 degrees C showed almost normal CH50 levels, nearly equal to p-CH50, while sera prepared at room temperature and at 4 degrees C showed a remarkable decrease in their CH50 levels. Further, when the serum from one patient showing this difference was added to pooled normal human serum, the C4 activity of the mixture decreased markedly at 4 degrees C but not at 37 degrees C. It is concluded that the difference is caused by cold activation of serum complement through the classical pathway in vitro.

Two cases of papillary type of C cell carcinoma of the thyroid were reported. They showed papillary arrangement with fibrovascular stalk in properly fixed tissues. Histochemically argyrophil reaction was positive in the cytoplasm and amyloid deposited in the stroma. Ultrastructurally secretory granules were found in their cytoplasm. The papillary type is not an artifact but one of the histologic variations of this carcinoma.

Sera of normal individuals or patients with lung cancer were assayed for Forssman-like antibody by a quantitative and specific method using ethylenediaminetetraacetate-containing buffer to inactivate complement in the test serum. It was shown that although Forssman-like antibody levels were distributed widely, (a) the levels of young (20 to 45 years of age) normal subjects of Blood Groups A and AB were lower than those of Blood Groups O and B, (b) the levels of old (60 to 80 years of age) normal subjects were lower than those of young normal subjects of Blood Groups O and B, and (c) the levels of old lung cancer patients were lower when compared to age-matched normal individuals of their blood group.
The effect of brain trauma on the formation of metastatic growth was examined by iv injection of 6 strains of rat ascites hepatoma. Three of the tumors (AH7974F, AH66F, and AH13) were in the single-cell state and three (AH272, AH7974, and AH601) contained cell aggregates. None of the tumors showed any tumor growth in the intact brain after injection into the tail vein. However, after traumatization of the brain tissue it was possible to observe tumor formation in the lesion, mainly by injection of the single-cell type tumors. The brain tissue was most susceptive to the tumor cells 10 min and 7 days after trauma. On the other hand, after the injection into the carotid artery of traumatized animals, the tumor formation in the brain was found in both tumor groups.
A long-term cell culture of human glioblastoma was investigated microscopically, virologically, and biochemically. Reverse transcriptase activity was detected in cultured human glioblastoma cells. 3H uridine was incorporated into particles of buoyant density at 1.07 g/ml (Ficoll) which is equal to that of Oncorna virus particles, but 3H thymidine was not incorporated at all. Furthermore, reverse transcriptase activity was also demonstrated with the particles, suggesting that the cultured human glioblastoma cells were producing type C Oncorna virus. Ultrastructural observations of cell culture of glioblastoma showed type C virus particles in cisternae and culture medium. Budding of the virus was also seen on the surface of the cell. The mean diameter of the particles was approximately 100 nm. Ca. 1.1 nm of spikes protruded from the envelope. Both types of virions were observed, i.e. the doughnut-shaped type form and the solid circular form.
The studies of serum from a case with C9 (the ninth component of complement) deficiency are described. A 29-year-old woman in good health was found to have low serum complement levels (CH50). C9 of her serum was undetectable by the hemolytic assay and by the immunochemical analysis but all other components were normal. It was demonstrated that low CH50 of her serum was due to the hemolysis of the sensitized sheep erythrocytes (EA) by the complement components from C1 to C8.
