Human deficiencies of vitamin B6. I. Studies on parameters of the assay of the glutamic oxaloacetic transaminase by the CAS principle.

Human deficiencies of vitamin B6 may be detected and accurately measured by a dual enzymic assay which is based on the CAS principle, i.e., the unsaturation and saturation of the coenzyme (pyridoxal 5'-phosphate) of a Coenzyme - Apoenzyme System. Glutamic oxaloacetic transaminase (GOT) is excellent for this assay. Two control groups (24 athletes and 25 students) showed deficiencies of GOT activity of 11+/-6% and 12+/-7%, respectively, which are negligible and which may be compared with analogous data from disease states. Contrary to a previous report, GOT activities must be determined within one day of preparation of the enzyme for meaningful assays. Frozen storage of erythrocyte hemolysates up to 21 days showed increasing apparent dissociation of pyridoxal 5'-phosphate from the coenzyme - apoenzyme system corresponding to increasing false deficiency levels. Dilution of the hemolysates and frozen storage augmented the apparent dissociation and false levels. This dual GOT assay is effective to detect and measure human deficiencies of vitamin B6.

Coenzyme Q deficiency in mice following infection with Friend leukemia virus.

The specific activities of the succinate dehydrogenase-coenzyme Q reductase were assayed on mitochondrial preparations of this enzyme from spleen, liver, blood and peritoneal macrophages of mice infected with Friend leukemia virus, and of control mice. Significant increases in the deficiency of coenzyme Q-enzyme activity were found in the spleen and blood of infected animals as the infection progressed. No significant deficiency was observed in the liver or the peritoneal macrophages. The development of the deficiency and splenomegaly during infection seemingly correlate with previous data which showed that administration of coenzyme Q resulted in a reversal of splenomegaly and a reduction in mortality. A limiting factor in the resistance of mice to Friend leukemia virus appears to be the availability of coenzyme Q.

A murine monoclonal antibody (928) recognizing a new epitope formed with a combination of HLA-DPA1*0201 and DPB1*0301 gene products.

A murine monoclonal antibody (mAb), 928, that recognizes a cell surface antigen (928 Ag) on a human Epstein-Barr virus-transformed fetal liver-derived lymphoid progenitor cell line (FL4.4) was generated. The 928 mAb reacted with only FL4.4; it did not react with any other 57 cell lines tested. Two color flowcytometry analysis of peripheral blood mononuclear cells (PBMC) revealed that the 928 mAb reacted with B cell and monocyte fractions from only two individuals out of 63 unrelated donors. Biochemical analyses showed that the 928 Ag composes of two molecules (33 and 34 Kd) and forms a SDS-resistant, noncovalently linked dimer conformation, the feature being similar to that of peptide-bound MHC class II molecules. Treatment of FL4.4 cells with the 928 mAb significantly facilitated homotypic cell aggregation. In addition, treatment of PBMC of the 928 Ag+ donor with recombinant IL-4 augmented the expression of the 928 Ag on CD64+ monocytes. Typing of HLA-DRB1, DPA1 and DPB1 alleles of the 928 Ag expressing and nonexpressing cells revealed that the 928 Ag is expressed only on PBMC of HLA-DPA1*0201 and DPB1*0301 positive donors. Finally, anti-DP antibody precleared 928 Ag from the cell lysate. These results demonstrate that the 928 mAb recognizes a polymorphic determinant of HLA-DPA1*0201-DPB1*0301 gene products. The possibility that amino acids in the groove of the peptide-binding site of HLA-DP molecules are critical for the 928 epitope is discussed.

Vitamin A and E requirements during total parenteral nutrition.

Studies were undertaken to determine rational dosages of vitamin A and E during long-term total parenteral nutrition (TPN). Four kinds of vitamin prescriptions containing different amounts of vitamin A and E were prepared from commercially available products and/or hospital pharmacy products. Patients were divided into four groups according to the vitamin prescription used. Plasma vitamin levels of different patient groups were determined by a modified fluorimetric method and were compared with those of a normal subject group. The stability of vitamin A and E in TPN solution after admixing was determining by measuring the remaining vitamin contents by high pressure liquid chromatography. From the results, it was concluded that 1) about 50% of vitamin A was decomposed by sunlight (about 2000 lux) 3 hr after admixing and an orange-colored vinyl cover could protect its photodecomposition; 2) vitamin E was stable at any condition tested; 3) 2500 IU of vitamin A and 15 IU of vitamin E could meet the daily requirements; 4) the plasma levels of vitamin A and E were correlative (p less than 0.01); and 5) concomitant administration of vitamin E was essential to keep the poorer level of vitamin A in plasma.

Testicular secretion of inhibin in the male golden hamster (Mesocricetus auratus).

To identify the cellular source of inhibin in the male golden hamster, we have used complementary approaches, immunohistochemistry and enzyme-linked immunosorbent assay (ELISA). Strong positive staining of the inhibin alpha subunit was observed in both the Sertoli and Leydig cells of the testes. No specific staining was observed for the inhibin betaA subunit, whereas specific staining for the inhibin betaB subunit was strongly positive in the Leydig cells. Inhibin pro-alphaC and inhibin B were detected in peripheral plasma, and testicular homogenate also contained large amounts of inhibin pro-alphaC and inhibin B. However, inhibin A was not detected either in peripheral plasma or in testicular homogenate. Plasma concentrations of inhibin pro-alphaC and inhibin B were significantly (P < .001) decreased 24 hours after orchidectomy. These results strongly suggest that the Leydig cells are the main source of dimeric inhibin B in the male golden hamster.

Measurements of magnetic relaxation times of normal tissue and renal cell carcinoma.

Spin-lattice (T1) and spin-spin (T2) magnetic relaxation times of 25 human renal cell carcinomas and their assorted normal tissues were measured with a Bruker NMR spectrometer operating at 20 MHz. The tissue samples were examined within four hours after surgery. The results (mean +/- SD) were as follows: renal cell carcinoma, T1 = 638 +/- 168 msec and T2 = 109 +/- 41 msec; normal renal tissue, T1 = 594 +/- 165 msec and T2 = 100 +/- 24 msec. These results indicate that there was no significant difference in T1 and T2 between normal renal tissue and carcinoma. Our results suggest that it is difficult to separate relaxation times of renal cancer from those of normal parenchyma and that the difference between T1 and T2 alone does not permit recognition of renal cell cancer. Paramagnetic contrast agents may be useful in MR imaging to differentiate renal cancer from normal parenchyma.
