[Assessment of tissue and intracellular pH following cold-induced vasogenic brain edema--in vitro and in vivo study].

Acid-base balance is closely related to the brain function and various methods have been applied to estimate the intracellular, extracellular or tissue pH. Umbelliferone, a fluorescent pH indicator, has been used to study either intracellular or tissue pH. In the present study, umbelliferone was used for histochemically estimating the tissue pH in the rat brain. The pH nomograms were made from the difference between fluorescent intensities at 450 nm on both excitations at 370 nm and 340 nm at different tissue water contents and different umbelliferone concentrations. Linear correlations were noted under each condition but 0.125% w/v umbelliferone provided the best estimation of tissue pH because of the least influence of water contents. Using this concentration of umbelliferone, a histochemical study was done to estimate the sequential changes in tissue pH following cold-induced vasogenic brain edema in rats. The results revealed a clear delineation of brain edema and the mild alkalosis in the edematous tissue, which probably reflects the pH of leaked plasma fluid. While the pH in the cortex around the lesion was neutral in the histochemical study, it was acidic in the in vivo study. This discrepancy is probably due to the former indicating the tissue pH while the latter the intracellular pH. Thus, the alkalosis in the edema fluid and the intracellular acidosis in the cortex around the lesion were noted in the cold-induced vasogenic edema.

Biotransformation of isoflurane: urinary and serum fluoride ion and organic fluorine.

The serum and urinary concentrations of fluorinated metabolites of isoflurane after inhalation of three different concentrations of isoflurane were studied in 18 ASA physical status 1 or 2 patients, scheduled for orthopedic or otolaryngeal surgery. Isoflurane was administered for 60 min during fentanyl-nitrous oxide-oxygen, and its end-tidal concentration was maintained at 0.3, 0.6, or 1.15% (groups I, II, and III). The organic fluorine was determined by combustion and fluoride ions were analyzed by ion chromatography. The amounts were expressed in terms of fluoride ion. The concentrations of serum fluoride ion and organic fluorine increased significantly 15 min after the onset of inhalation of isoflurane. The mean peak values of fluoride ions were 3.8 +/- 1.1, 3.9 +/- 1.4, and 4.2 +/- 0.9 mumole/1 (M +/- SD) in patients in groups I, II, and III, respectively. The half-lives of fluoride ion and of organic fluorine as metabolites of isoflurane, calculated from the amounts excreted in urine, were 36 h and 41 h, respectively. The cumulative amounts of fluoride ion and organic fluorine excreted up to the 6th postoperative day were 548 +/- 230 and 785 +/- 452 mumoles in group I, 594 +/- 138 and 1,378 +/- 807 mumoles in group II, and 1,032 +/- 496 and 728 +/- 265 mumoles (M +/- SD) in group III, respectively. The urinary excreted fluoride ion increased in proportion to the dose of isoflurane and approximately 1.3 mmol was excreted per 1 MAC X hour inhalation of isoflurane. The authors concluded that isoflurane might be biotransformed to a greater extent than reported previously, although the serum fluoride ion level was found to be low.

[Hypervascular chordoma: report of a case].

The authors report a rare case of sellar chordoma with a marked vascularity documented on cerebral angiogram. Its possible pathogenesis and some therapeutic problems were briefly discussed. Only a few cases of chordoma with such a positive tumor stain have been reported previously.

[The effect of Gd-DTPA on magnetic resonance imaging: a potential blood-brain barrier indicator].

Magnetic resonance image (MRI) has been applied on central nervous system to evaluate the anatomical and functional aspects. On the other hand, gadolinium-diethylenetriamine-pentaacetic acid (Gd-DTPA) is considered to be a valuable contrast agent on MRI. This paramagnetic compound shortens the magnetic relaxation times of surrounding hydrogen nuclei by altering local magnetic environments. A molecular weight of this compound was 590, therefore this does not pass through the normal blood-brain barrier (BBB), and it may possible to detect a breakdown of BBB in vivo. In order to investigate the effect of Gd-DTPA as an indicator of BBB disruption on MRI, we measured T1 value of hemisphere of rat in osmotic BBB disruption model and experimental global cerebral ischemic model with and without Gd-DTPA. The MRI system employed was JNM-SMR 270 (JEOL) and the superconducting magnet was operated at a field strength of 6.34 tesla. Gd-DTPA was injected intra-venously through femoral vein and its dose was 0.5 mmol/kg. Osmotic BBB disruption model was made by intra-arterial injection of 25% mannitol through an internal carotid artery. Calculated T1 value of the ipsilateral hemisphere decreased with Gd-DTPA to 43.3 +/- 10.2% (n = 4) of control value immediately after BBB disruption with mannitol administration but no change of T1 value was recognized with Gd-DTPA in rats which Gd-DTPA was injected 4 hours after BBB disruption. Global ischemic insult was made by 3 vessel occlusion permanently and temporarily (30 or 120 minutes).(ABSTRACT TRUNCATED AT 250 WORDS)

[Measurement of cerebral blood volume by reflections spectrophotometry: methodological improvement and quantitative evaluation].

The authors described methodological improvement and quantitative evaluation in measurement of cerebral blood volume (CBV) by reflectance spectrophotometry. The measurement of blood volume by this method has not been applied to the brain tissue because of the methodological difficulties. In order to obtain the accurate CBV values with this method, it is necessary to keep the proper contact between the sensor for the measurement and the brain tissue to be measured. For this purpose we developed a balance arm. A sensor for CBV measurement was fixed with the one side of the arm and a weight was installed to the other side. The pressure of the sensor to the brain can be adjusted by moving the weight, and the sensor was placed on the brain tissue with the proper pressure. Moreover, the sensor was attached with an acryl-made cap to make the pressure adjustment easier by enlarging the contact surface and to enable to observe the vasculature on the surface of the brain. With this improved method the reasonable changes of CBV could be obtained when the CO2 gas was inhalated and cerebral ischemia was produced in cats. The possibility of the quantitative evaluation was investigated using the homogenized brain tissue of cats. After the elimination of the blood from the brain by the perfusion with the saline, the brain was removed and homogenized. Then, the definite volume of blood, of which hemoglobin volume was already known, was added and measured by this method.(ABSTRACT TRUNCATED AT 250 WORDS)

Changes of vasoactive intestinal polypeptide-like immunoreactivity in cerebrovascular nerve fibers after cervical sympathectomy in the dog.

The presence of vasoactive intestinal polypeptide-like immunoreactivity (VIP-LI) was confirmed in many neuronal perikarya of the superior cervical ganglion (SCG), as well as in many perivascular nerve fibers of the major cerebral arteries in the dog. After bilateral extirpation of the SCG, VIP-LI was decreased markedly in the basilar artery and the posterior cerebral artery, and moderately in the middle and anterior cerebral arteries.

Increased membrane fluidity of erythrocytes in a case with malignant hyperthermia.

In a case with malignant hyperthermia (MH), membrane fluidity of erythrocytes was studied by electron spin resonance (ESR)-spin labeling technique. The measurement was made with samples obtained during and after pyrexic crisis until full recovery. The results indicated that membrane fluidity increased during pyrexic crisis and that these changes were serially restored. As the physicochemical characteristics of the erythrocyte membrane resemble those of cellular and cell organella membrane, it was concluded that membrane composition and configuration might have been altered pathophysiologically during the crisis of MH in our case.

Magnetic resonance imaging of human cerebral infarction: enhancement with Gd-DTPA.

Five patients (1 female and 4 males) with cerebral infarction of 4 h to 27 months duration were studied 9 times with magnetic resonance (MR) using Gd-DTPA. Spin-echo (SE) MR images (MRI) were obtained before and after the administration of Gd-DTPA, and correlative CT scans were performed on the same day. In 2 cases, 4 h and 27 months after the ictus, there was no enhancement with Gd-DTPA. There was faint enhancement in 2 cases with cerebral infarction of about 24 h duration and obvious enhancement in all cases in the subacute stage. Compared with enhanced CT, MR using Gd-DTPA demonstrated more obvious enhancement of infarcted areas. MR enhancement using Gd-DTPA showed a gradual increase and the accumulated Gd-DTPA in infarcted areas slowly diffused to the periphery. MR enhancement with Gd-DTPA is similar to that of enhanced CT, but may be more sensitive in the detection of blood brain barrier breakdown.

Reflectance spectrophotometric measurement of in vivo local oxygen consumption in the cerebral cortex.

A simple method was developed to measure in vivo local oxygen consumption quantitatively in the brain cortex. Reflectance spectra of tissue hemoglobin at the brain's surface were measured for assessment of both local tissue hemoglobin content and its oxygen saturation. Local oxygen consumption was calculated from the spectral changes of tissue hemoglobin during complete cessation of blood flow by compression of the cortical surface in the suprasylvian gyrus with the tip of an optic probe. This procedure was performed without any brain damage and only took approximately 5 s. The calculated local oxygen consumption during this short period of compression remained constant for a few seconds. Then, it decreased rapidly, although the local tissue hemoglobin was not completely deoxygenated. The value of local cerebral oxygen consumption obtained by this method was 3.02 +/- 0.61 mL O2/100 g brain/min; it was not influenced by the change in systemic blood pressure. The effect of pentobarbital on cerebral oxygen consumption was also studied. At the stage of burst and suppression on electrocorticogram, cerebral oxygen consumption decreased significantly (p less than 0.001) to 1.03 +/- 0.07 mL O2/100 g brain/min.

Fluorescence of the tryptophyl residues of the constant fragment of the immunoglobulin light chain.

The constant (CL) domain of the type-lambda immunoglobulin light chain contains two tryptophyl residues. The fluorescence lifetimes of the CL fragment and its two modified forms, the reduced CL, and reduced and alkylated CL fragments, were measured at pH 7.5 and 25 degrees C. The fluorescence decay kinetics of these fragments were described in terms of two lifetimes. The static and dynamic quenching reactions by acrylamide of the tryptophyl residues of these fragments were measured and their dynamic properties were compared with the static properties reported previously (Goto, Y. & Hamaguchi, K. (1979) J. Biochem. 86, 1433-1441; Goto, Y. & Hamaguchi, K. (1986) Biochemistry 25, 2821-2828; Ashikari, Y., Arata, Y., & Hamaguchi, K. (1985) J. Biochem. 97, 517-528). It was found that although the static conformation of the reduced CL fragment is very similar to that of the intact CL fragment, the dynamic conformations of these two fragments differ considerably.

Antitumour activity of purified arabinogalactan-peptidoglycan complex of the cell wall skeleton of Rhodococcus lentifragmentus.

Antitumour activity of arabinogalactan peptidoglycan (AP) complex (peptidoglycan and arabinogalactan liberated by an acid or alkaline treatment from Rhodococcus lentifragmentus AN-115 cell wall skeleton) was examined in mice and compared with that of the cell wall skeleton. The growth of syngeneic fibrosarcoma Meth A cells after implantation in BALB/c mice was significantly suppressed by AP complex, and also regressed after intratumoral injection of AP complex on days 1, 4 and 7 after tumour implantation. Although the activity of peptidoglycan was less than that of AP complex, peptidoglycan also showed both tumour-suppressive and regressive activities. Arabinogalactan did not show antitumour activity. It is interesting that peptidoglycan has an important role in the effect against tumours.

[Host mediated anti-tumor effect of Nocardia rubra cell wall skeleton on syngeneic tumor in mice].

The mode of action of Nocardia rubra cell wall skeleton (N-CWS) on Meth A fibrosarcoma (Meth A) was studied in BALB/c mice. N-CWS suppressed or regressed the intradermal growth of syngeneic Meth A cells in normal BALB/c and athymic BALB/c mice. The intradermally and subcutaneously infiltrated cells harvested from injection sites of N-CWS in normal mice showed in vitro cytotoxic activity against Meth A cells. Pretreatment of normal BALB/c mice with immunosuppressing agents such as hydrocortisone, carrageenan, or silica particles significantly reduced the anti-tumor effect of N-CWS. The growth of Meth A cells, rechallenged into BALB/c mice in which Meth A cells had once been suppressed or regressed by N-CWS treatment, was also inhibited, but not in similarly treated athymic nude mice. This resistant mechanism was shown to be dependent out cellular components but not on humoral components by the Winn Assay. The present results suggest that N-CWS exerts its anti-tumor activity by mediation of the immune system of the host and that the main effector cells in the early stage of tumor rejection are macrophages; T cells may also be involved in the later stage.

Experimental induction of cerebral aneurysms in monkeys.

Saccular cerebral aneurysms were successfully induced in two monkeys treated with ligation of the common carotid artery, experimental hypertension, and beta-aminopropionitrile feeding. The cerebral aneurysms developed on the large arteries at the base of the brain, such as the anterior communicating artery and the internal carotid artery at the origin of the posterior communicating artery. Because of the similarity of the monkey to man as a species, the present results strongly suggest the significance of postnatal aggravating factors in the development of cerebral aneurysms in man.

A new antitumor antibiotic, FR-900482. III. Antitumor activity in transplantable experimental tumors.

FR-900482 (4-formyl-6,9-dihydroxy-14-oxa-1,11-diazatetracyclo[7.4.1.02,7, O10,12]tetradeca-2,4,6-triene-8-ylmethyl carbamate), a new antibiotic with antitumor activity was isolated from fermentation broth of Streptomyces sandaensis. Its antitumor activities were studied and compared with that of mitomycin C (MMC) in animals. FR-900482 in doses of 0.32 approximately 10 mg/kg (ip) prolonged the life of mice bearing ascitic P388, L1210, B16, MM46, Ehrlich or EL4 tumors and rats bearing ascitic AH130 or AMC60 tumors. FR-900482 in doses of 5.6 approximately 18 mg/kg (iv) inhibited human LX-1, MX-1, SC-6 and LC-6 tumors xenografted sc in nude mice. FR-900482 was more effective than or equally effective to MMC in all the tumors used. FR-900482 was ineffective against cyclophosphamide-resistant P388, but was effective against MMC- or vincristine-resistant P388. The results suggest that FR-900482 may have clinical potential.
