Vascular and non-vascular ligands for L-selectin.

Ligands for L-selectin are expressed not only on vascular endothelial cells but also in the extravascular tissues. In this article, we summarize the current understanding of the "vascular" ligands for L-selectin. We also describe identification of "non-vascular" ligands for L-selectin and discuss their possible biological significance.

[A case of multiple sclerosis with initial symptoms of narcolepsy].

We reported a 13-year-old boy diagnosed as multiple sclerosis associated with narcolepsy. He had suffered from retrobulbar optic neuritis at the age of 11 years which was improved gradually by prednisolone. Four months later he was admitted because of excessive somnolence. The diagnosis of narcolepsy was made based on hypnagogic hallucination, sleep paralysis, changes of personality and the sleep onset sREM (SOREM). The elevation of anti-measles antibody and a positive oligoclonal band in CSF, low density areas in the bilateral internal capsule on CT scan, and high signal areas in the same region on T2-weighted MRI confirmed the diagnosis of multiple sclerosis. An abnormal arousal response occurred occasionally in sleep stage 2 and 4, which started with electrical silence followed by a period with irregular high voltage slow waves and repetitive focal spike train. Those clinical symptoms and abnormalities of biochemical and electrophysiological studies normalized after treatment with prednisolone. However, abnormalities on MRI showed no improvement even after long term administration of prednisolone (2.5 mg/day).

[A case of metastatic renal tumor originating from lung cancer difficult to distinguish from renal cell carcinoma].

A 66-year-old female underwent left lower lobectomy for her lung cancer (adenocarcinoma) in November 1992, followed by the resection of brain metastasis in October 1993. Later, a left renal tumor with paraaortic lymph node swelling was found by a follow-up abdominal CT. She was treated with left nephrectomy and the resection of the paraaortic lymph nodes in June 1997. The histopathology of the resected tumor and the lymph nodes revealed a metastatic renal tumor originating from the pulmonary adenocarcinoma. There have been 38 reported cases of metastatic renal tumor from lung cancer in the Japanese literature.

[Leiomyoma of the bladder: report of two cases].

We report 2 cases of leiomyoma of the urinary bladder. A 41-year-old female visited our hospital with the complaint of pollakisuria. A solid tumor of the urinary bladder was found by ultrasonography. A large shadow defect at the left-anterior wall was shown by drip infusion pyelography (DIP). Computed tomographic scan (CT) and magnetic resonance imaging (MRI) also revealed a large tumor. T1-weighted image revealed a homogeneous low intensity tumor and T2-weighted image disclosed heterogeneous low intensity tumor. Cystoscopy revealed a large submucosal tumor. Partial cystectomy was performed, and she has had neither recurrence nor metastasis for 36 months. A 32-year-old male was referred to our hospital with the complaint of macrohematuria. A solid tumor of the urinary bladder was found by ultrasonography. A shadow defect was not clearly detected by DIP. A large tumor was detected on the anterior wall by MRI. T1-weighted image showed a homogeneous low intensity tumor and T2-weighted image disclosed a high intensity tumor. Cystoscopy revealed a submucosal tumor on the anterior wall. Urine cytology did not suggest a malignancy. The biopsied specimens revealed only an inflammatory change in the mucosa. Partial cystectomy was carried out. He has had neither recurrence nor metastasis for 29 months. Histological diagnosis in both cases was leiomyoma of the urinary bladder.

Anemia-inducing factor expressed in gastric cancer is homologous with complement regulatory factor CD59?

Anemia-inducing factor (AIF) was isolated from gastric cancer tissue; however, the human placenta used as the volume of AIF for further analysis did not prove sufficient. This substance was named placental anemia-inducing factor (PAIF). PAIF directly reduces the number of erythrocytes in vitro and reduces the RBC count in rabbits to 80% when i.v. administration of 27 microg/kg of body weight is given. The aim of this study is to better define PAIF and to examine whether the identifical substance expresses on either the surface or in the cytoplasm of established gastric cancer cell lines. PAIF is a glycoprotein with about 20 KD, whose 17 amino acid residues of N terminus were sequenced after Edman treatment. The N-terminus of PAIF were determined as Lqcyncpnptadcktav. This is homologous with that of CD59, which is thought as a regulator of membrane attack complex of complement system. Expression of PAF or CD59 in four established gastric cancer cell lines were examined by indirect immunofluorescence method and by Northern blot hybridization. The cells (1 x 106) were seeded into plastic plates for three days and reacted overnight at 4 degrees C in 0.5 ml of PBS with anti-PAIF polyclonal antibody or with anti CD59 rat monoclonal antibody. Both PAIF and CD59 were stained positively on the surface and/or in the cyroplasm. The total RNAs were prepared from the four kinds of cell lines and normal human lymphocytes. CD59 mRNA was probed in all cell lines by BamH1-EcoR1 fragment of PSRa CD59. The signal levels of MKN-28, MKN-45 and KATO-III were stronger than that of MKN-74, whereas the signal of normal lymphocytes was the lowest. Although there is no decisive evidence that PAIF is exactly the same substance as CD59, and although the biological functions of these two substances are conflictive, and still to be further investigated, the 17 amino acid residues of N-terminus of PAIF expressed in gastric cancer cells were homologous with those of CD59. A derivative of CD59 may exist in gastric cancer.

A sulfatide receptor distinct from L-selectin is involved in lymphocyte activation.

Sulfatide induces leukocyte activation, which is thought to be mediated via L-selectin. Here we demonstrate that lymphocytes express a receptor for sulfatide distinct from L-selectin and that this receptor is involved in sulfatide-induced cell activation. While cell surface L-selectin expression was abolished by phorbol 12-myristate 13-acetate (PMA), lymphocytes retained the ability to bind sulfatide in liquid phase as well as in immobilized solid phase. The novel sulfatide receptor obtained from PMA-treated lymphocytes showed a molecular size of 65 kDa. Stimulation through this receptor triggered cytosolic free Ca2+ elevation and intercellular aggregation.

Protein expression, characterization, and regulation of CYP4F4 and CYP4F5 cloned from rat brain.

We previously reported the cDNA cloning of three new forms of P450, CYP4F4, CYP4F5, and CYP4F6, from a rat brain cDNA library. In the present study, we expressed CYP4F4 and CYP4F5 in Escherichia coli using the pCWOri expression vector with a modification of their N-terminal amino acid sequences and the incorporation of a C-terminal [His]4 tag to aid in purification. CYP4F5 recombinant protein was purified to a specific content of 7.7 nmol/mg protein from the membrane fraction of E. coli and showed omega-hydroxylation activity toward leukotriene B4 (LTB4), a chemical mediator of inflammation. On the other hand, the solubilized membrane fraction of CYP4F4-expressed recombinant protein catalyzed the omega-hydroxylation of prostaglandin A1, prostaglandin E1, and 6-trans-LTB4 as well as LTB4. The effects of the peroxisome proliferator, clofibrate, on mRNA expression of CYP4F4, 4F5, and 4F6 were studied by Northern blot analysis. The expression levels of the mRNA of these CYP4Fs were shown to be reduced by clofibrate in liver.

The Role of Axillary Lymph Node Dissection in Breast Cancer Management.

Although it is generally accepted that axillary dissection provides no survival advantage in patients with breast cancer, it is commonly regarded as a reliable method of assessing nodal status and treating regional disease. However, it is time to consider eliminating routine axillary dissection in patients who are clinically node-negative. A sentinel lymph node biopsy may assess axillary nodal status while obviating a full axillary dissection. At present, axillary dissection remains the standard approach for the surgical management of all patients with invasive carcinoma of the breast, regardless of tumor size or patient age, though it is unnecessary for patients with small intraductal carcinomas.

Role of Fas-Fas ligand interactions in the immunorejection of allogeneic mouse corneal transplants.

BACKGROUND: The expression of Fas ligand (FasL) in the eye has been proposed to be an important component of ocular immune privilege. Since the unusually favorable outcome of corneal transplantation is thought to result from the immune privilege of the eye, examination of the function of FasL on corneal allografts would be a test of that hypothesis. METHODS: To investigate the role of Fas-FasL interaction in corneal allografts, orthotopic corneal transplantation was performed using C57BL/6 (B6, FasL+) and B6-gld (FasL-) mice as cornea donors and BALB/c mice as recipients. The rejection rate of B6-gld grafts (FasL- group) was compared with that of normal B6 control corneas. RESULTS: The rejection rate at the final observation (8 weeks) in the FasL- group (89%) was significantly higher than in the FasL+ control group (47%). FasL expression was found on the corneal endothelium by staining with anti-FasL monoclonal antibodies. The TdT-mediated dUTP nick-end labeling assay revealed that apoptotic cells were attached to the endothelium in the control group but not in the FasL- groups. CONCLUSIONS: Apoptosis of infiltrating cells on the corneal endothelium resulting from Fas-FasL interaction plays an important role in the high success rate of corneal transplantation.

Rapid activation of MAP kinase by estrogen in the bone cell line.

We examined the effect of estrogen on mitogen-activated protein kinase (MAPK) in osteoblastic cells. Rat ROS 17/2.8 cells were exposed to 17beta-estradiol (E2) and MAPK activity in the cells was measured by an in vitro phosphorylation assay. E2 treatment caused a rapid and transient MAPK activation within 5 min. Insulin-like growth factor-I, which acts via their membrane receptors, caused a similar effect, but it required 10 min to reach the maximum level. Western blot analyses with anti-MAPK and anti-phosphotyrosine antibodies demonstrated that the E2 activation of MAPK was accompanied by phosphorylation of the enzyme. The concentration range (10 nM-1 pM) of E2 needed for this MAPK activation was less than that (1 microM-0.1 nM) needed for the transcriptional activation via the nuclear estrogen receptor (ER). These data provide the first evidence of MAPK activation by E2 through phosphorylation, which may be mediated through a putative plasma membrane receptor in the cultured bone cells.

Anterior chamber inoculation of splenocytes without Fas/Fas-ligand interaction primes for a delayed-type hypersensitivity response rather than inducing anterior chamber-associated immune deviation.

The inoculation of antigens into the anterior chamber (AC) of the eye induces an antigen-specific immune response that inhibits delayed-type hypersensitivity (DTH). This regulatory response is known as anterior chamber-associated immune deviation (ACAID). The ACAID response appears to be complex, as it can be elicited by a wide variety of soluble and cell-associated antigens, including foreign, self, tumor, and alloantigens. To evaluate the contribution of Fas/Fas ligand (FasL) interaction to the induction of ACAID to alloantigens, gld and lpr mutant mice were used in conjunction with normal C3H, MRL, and BALB/c mice. ACAID was induced by inoculation of non-irradiated splenocytes from donor mice into the AC of various recipients. After 1 week, recipients were primed intradermally with donor splenocytes. One week later DTH was measured by ear swelling. C3Hgld mutants lacking functional FasL did not develop ACAID after the AC inoculation of BALB/c splenocytes. Conversely, the AC inoculation sensitized these mutants. MRL/pr mutants, which lack Fas, developed ACAID following inoculation of BALB/c cells. AC inoculation of lpr splenocytes did not induce ACAID, but sensitized C3H recipients. Treatment of the AC inoculum with an anti-Fas antibody blocked ACAID induction in a transient manner, as the recipients developed ACAID later. These results show that interaction of the Fas and FasL is required to induce ACAID to allogeneic cells. In the absence of Fas expression on donor splenocytes, or FasL expression by the recipient, AC inoculation primes for a DTH response rather than inducing ACAID.

Primary malignant fibrous histiocytoma of the descending colon.

We herein present the case of a 50-year-old woman with malignant fibrous histiocytoma arising from the descending colon and localizing in the colonic wall. Malignant fibrous histiocytoma of the large bowel is a very rare tumor. A total of 18 cases, including our case, have been reported in the world medical literature so far and we also reviewed these cases. In our case, combined adjuvant chemotherapy was administered after a complete resection had been performed. No clinical signs of local recurrence or distant metastasis were found at 7 years after the operation.

Fucoidin, a potent inhibitor of L-selectin function, reduces contact hypersensitivity reaction in mice.

In order to investigate the role of L-selectin (CD62L) in delayed-type hypersensitivity (DTH) reaction, effect of fucoidin, a potent inhibitor of CD62L function, was examined in a model of mouse contact hypersensitivity reaction. Intravenous injection of fucoidin to sensitized mice just before hapten challenge resulted in a significant and dose-dependent reduction of the ear swelling in contact hypersensitivity. The ear swelling caused by the hapten challenge was also inhibited when fucoidin was administered at the sensitization phase. Histological analyses of the ear sections revealed that the fucoidin-induced suppression of contact hypersensitivity reflected a marked inhibition of the ear edema and the leukocyte infiltration. The activity of fucoidin was specific in that its related saccharides exerted little effect on the reaction. These results suggest that CD62L may play an important role in both afferent and efferent phases of cutaneous DTH reaction. Since DTH response is one of the most significant features of several chronic inflammatory diseases, our data also show that blocking of CD62L function may be beneficial for the treatment of these diseases in humans.

Isolation and amino acid sequence of flavostatin, a novel disintegrin from the venom of Trimeresurus flavoviridis.

Flavostatin, a novel disintegrin purified from the venom of Trimeresurus flavoviridis, consists of 68 amino acids, including an Arg-Gly-Asp sequence and 12 Cys residues at positions highly conserved among disintegrins. The N-terminal sequence of flavostatin was identical to those of triflavin and flavoridin, previously reported disintegrins from the Trimeresurus flavoviridis venom. Differences among the C-terminal sequences of these disintegrins are considered to affect their biological potencies. Isolated flavostatin inhibited ADP collage, and thrombin receptor agonist peptide-induced platelet aggregation in human platelet-rich plasma with an IC50 range of 59 to 98 nM. Contrary to expectations, these values were similar to those for triflavin.
