Formation of 8-hydroxydeoxyguanosine in calf thymus DNA treated in vitro with phenylhydroquinone, the major metabolite of O-phenylphenol.

The generation of 8-hydroxydeoxyguanosine (8OHdG) in calf thymus DNA treated with O-phenylphenol (OPP) or its major metabolites, phenylhydroquinone (PHQ) and phenylbenzoquinone (PBQ), was studied. The content of 8OHdG residues was increased in DNA treated with PHQ, and the generation of 8OHdG was highly dependent on PHQ concentration. PBQ had little effect on the formation of 8OHdG, and OPP had no effect. The formation of 8OHdG by PHQ was reduced by oxygen radical scavengers such as catalase, sodium benzoate and sodium azide. The PHQ-induced 8OHdG formation was accelerated by the addition of CuCl or CuCl2 to the reaction mixture, but was decreased by the addition of chelating agents such as EDTA, bathocuproinedisulfonic acid disodium salt (bathocuproine disulfonate) and O-phenanthroline. These results demonstrate that hydroxyl radicals generated in the process of oxidation of PHQ contribute to the formation of 8OHdG in DNA, and copper ions facilitate the oxidative DNA damage. Copper ions greatly accelerated the PHQ-induced DNA cleavage in vitro, although they had no effect on cleavage without PHQ. On the other hand, DNA cleavage occurred by the addition of FeCl2 in the absence and presence of PHQ. FeCl2 stimulates 8OHdG formation only slightly with or without PHQ. Furthermore, the stimulatory effect of FeCl2 on 8OHdG formation was observed even in the presence of EDTA. The formation of 8OHdG in bladder DNA is likely to be one of a series of events leading to bladder tumors seen in rats fed OPP-containing diet.

Formation of an oxidative DNA damage, 8-hydroxydeoxyguanosine, in mouse lung DNA after intratracheal instillation of diesel exhaust particles and effects of high dietary fat and beta-carotene on this process.

Diesel exhaust particles (DEP) cause tumors in the respiratory tracts of experimental animals. It was previously shown that DEP produced superoxide and hydroxyl radical. To examine whether oxygen radicals are involved in mouse lung tumorigenesis induced by DEP, formation of an oxidative DNA damage, 8-hydroxydeoxyguanosine (oh8dG), by DEP was investigated. Furthermore, the role of high dietary fat and beta-carotene on this process was studied. After intratracheal instillation of DEP, a significant increase of oh8dG in mouse lung DNA was observed. High dietary fat enhanced the formation of oh8dG in lung DNA. Intake of beta-carotene suppressed the formation of oh8dG in lung DNA, but the protective effect of beta-carotene against this process was not statistically significant. These results suggest that formation of oh8dG in lung DNA was induced by oxygen free radicals produced by DEP. Thus, it is possible that oh8dG is a promutagenic lesion in DEP-induced lung tumorigenesis in mice and high dietary fat enhances this process through the generation of oh8dG in mouse lung DNA.

Formation of a mutagen, glyoxal, from DNA treated with oxygen free radicals.

Oxygen free radicals cause extensive chemical changes in DNA, including base and sugar modifications and strand breaks. In the present study we found that a mutagen, glyoxal, is produced by exposure of DNA to an oxygen radical forming system (5 mM FeSO4-EDTA, 37 degrees C, 60 min). It was produced with 17 times higher efficiency than 8-hydroxydeoxyguanosine. Thus it is possible that the formation of glyoxal is one of the major types of damage in DNA exposed to oxygen free radicals.

Effect of Kil769, a novel K(+)-channel opener, on sensitivity to Ca2+ of contractile elements and inositol phosphate formation in porcine coronary artery.

To determine whether Kil769, a novel K(+)-channel opener, acts intracellularly in vasorelaxation, we compared the effects of Kil769 on force of contraction, intracellular Ca2+ concentration ([Ca2+]i) and inositol phosphate (IP1) formation with those of Ca(2+)-channel blockers in isolated porcine coronary artery. Kil769 (10 microM) and verapamil (1 microM), which produced submaximal relaxation, reduced the increase in [Ca2+]i and force of contraction induced by 25 mM KCl. Verapamil reduced [Ca2+]i and the force of contraction to a similar extent but Kil769 reduced force of contraction more strongly than it did [Ca2+]i. Kil769 also inhibited U46619 (9,11-dideoxy-9 alpha,11 alpha-methano-epoxy-PGF2 alpha)-induced IP1 formation and glibenclamide blocked its inhibitory effect. These results suggest that the opening of K+ channels induced by Kil769 reduces the Ca2+ sensitivity of contractile elements and inositol phospholipid hydrolysis which is related to the Ca2+ release from intracellular storage.

[Anti-atheromatous effects of fenofibrate, a hypolipidemic drug. I: Anti-atheromatous effects are independent of its hypolipidemic effect in cholesterol-fed rabbits].

Anti-atheromatous effects of fenofibrate were studied in cholesterol-fed rabbits. Rabbits in the control group (HCD) and fenofibrate group (F-HCD) were fed the 0.5% cholesterol diet and the 0.5% cholesterol plus 0.11% fenofibrate diet (corresponding to ca. 30 mg/kg/day), respectively, for 2, 4 or 10 weeks. Fenofibrate did not change serum levels of total cholesterol, HDL-cholesterol and triglyceride during the feeding period. The percentage of plaque area (PPA) formation in the thoracic aorta was time-dependently increased. PPA values were reduced in the rabbits treated with fenofibrate for 2 and 4 weeks, but not in those treated for 10 weeks. Fenofibrate had no effect on the plaque thickness. The percentage of circulating free platelets in the HCD group was reduced at the 4th week of feeding, but that in the F-HCD group was not. The anti-platelet effect of fenofibrate might cause the anti-atheromatous effect. Fenofibric acid, an active metabolite of fenofibrate, had no inhibitory effect on LDL peroxidation in vitro. From these results, we conclude that fenofibrate manifests an anti-atheromatous effect independent of the hypolipidemic effect in cholesterol-fed rabbits and that it may inhibit an early event in the atherogenesis.

[Anti-atheromatous effects of fenofibrate, a hypolipidemic drug. II: Anti-atheromatous effects are independent of its hypolipidemic effect in hereditary hyperlipidemic rabbits].

Anti-atheromatous effects of fenofibrate were studied in KHC rabbits with a hereditary deletion in LDL receptors, a defect similar to that in Watanabe heritable hyperlipidemic rabbits. KHC rabbits (10-weeks-old) were given a dietary admixture of fenofibrate (ca. 30 or ca. 100 mg/kg/day) for 20 weeks. Fenofibrate did not change serum total cholesterol, high density lipoprotein and triglyceride levels. Fenofibrate decreased the percent of plaque area formation in the thoracic aorta, but not the degree of foam cell formation, fibrosis and edematous change, and thickness of the intima. Furthermore, fenofibrate markedly inhibited the medial damage with foam cell formation in the aorta at week 20. From these results, we conclude that fenofibrate manifested the anti-atheromatous effect in KHC rabbit and also the previous model of cholesterol-fed rabbits, particularly against the medial damage in KHC rabbits, independent of the hypolipidemic effect.

[Pain management in advanced pediatric cancer patients--a proposal of the two-step analgesic ladder].

Pain treatment for 17 pediatric cancer patients in our institution was evaluated and disirable cancer pain management for children was discussed. Most of the patients (aged 1-17 years) suffering severe pain for about one month were in the advanced stage of the malignant diseases (e.g. leukemia). The pain etiology was mostly tumor-associated while therapy-related pain accounted for 23.5%. These pains were treated with NSAIDs or pentazocine before the consultation with inadequate relief. Oral morphine sulfate or continuous intravenous morphine chloride was administered to 16 patients with successful pain relief and side effects such as nausea (52.9%) and drowsiness (41.2%). It took 5.5 days on average until adequate pain control methods were determined. It is known that most NSAIDs frequently used for pediatric pain possibly cause adverse effects such as platelet dysfunction or mucous membrane injury with a suppository, which could lead to a fatal disorder in clinically ill pediatric cancer patients. Moreover sufficient doses for the pain relief are not necessarily given to the pediatric patients because of a limit to the dosage of NSAIDs. The period of pediatric cancer pain in which the patient require a methodical treatment and receive benefit from pain relief is relatively short in the advanced stage, not to mention the early stage in which chemotherapy is efficacious against cancer disease itself. Therefore, to obtain effective pain control within a short time, the authors propose the pain management for advanced pediatric cancer patients by the two-step analgesic ladder prescribing weak or strong opioid analgesics first, adapted from the three-step ladder of the WHO Cancer Pain Relief, 1986.

[A case of spinal astrocytoma presenting spinal transverse sign due to hematomyelia].

An unusual case of cervical astrocytoma due to hematomyelia was reported. A 68-year-old man was admitted to our institute because of sudden onset of paraplegia and urinary and bowel disturbance. CT and plain X ray on admission showed no abnormal findings, but emergency MRI revealed a high signal intensity area in Th 8-9 level on T1 weighted image. Preoperative diagnosis was idiopathic hematomyelia and emergency laminectomy was performed. After the dural incision from Th 8-10, the intramedullary hematoma was totally removed, but at the cephalic portion within the hematoma cavity an abnormal tumorous tissue was detected. It was noticed with the bleeding sauce of hematomyelia, and it, too, was totally removed. The microscopic finding concerning the surgical specimen was fibrillary astrocytoma (grade 2), and its vascular component was relatively rich.

Cytogenical analysis of brain tumors by FISH (fluorescence in situ hybridization) and FCM (flow cytometry).

In the present study, an analysis was made of chromosomal aberrations in brain tumors using the fluorescence in situ hybridization (FISH) technique. At the same time DNA histograms were obtained by flow cytometry (FCM) to make a comparative study of histological malignancy and prognosis. The subjects included 30 gliomas (7 of astrocytoma grade II, 15 of anaplastic astrocytoma (grade III), 8 of glioblastoma (grade IV)) and 26 meningiomas. In the study of FISH, DNA probes for chromosomes No. 7, 9, 10, and 17, were used to cause a reaction with chromosome No. 22 added for meningiomas. In the study with FCM, DNA index from DNA histogram was calculated using lymphocytes as the internal standard. In gliomas as a whole, chromosomes No. 7 and 17 showed high values, whereas, chromosomes No. 9 and 10 low values. Analysis by the grades of glioma showed that compared with gliomas of other grades, grade IV gliomas were higher for chromosome No. 17 and lower for chromosome No. 10. In meningiomas, while many cases showed a low value for chromosome No. 22, most cases of recurrent and atypical meningiomas showed a high value for chromosome No. 17. In gliomas, DNA index showed a correlation with the grade, and a positive correlation particularly with chromosome No. 17 in FISH. Recurrent and atypical meningiomas had a high DNA index.

2-Hydroxyadenine (isoguanine) as oxidative DNA damage: its formation and mutation inducibility.

In our experiments we found that hydroxylation occurs at the C-2 position of adenine by oxygen radical treatment (Fe(2+)-EDTA) of dA, dATP, and single- and double-stranded DNA. This oxidatively damaged base, 2-hydroxyadenine (also known as isoguanine), was produced more efficiently in monomers than in polynucleotides. 2-Hydroxydeoxyadenosine triphosphate was incorporated opposite T and C in a DNA template by DNA polymerase alpha and only opposite T by the Klenow fragment. The Klenow fragment, DNA polymerases alpha and beta incorporated dTMP and other nucleotides opposite 2-OH-Ade in DNA templates in vitro in a sequence-dependent manner. These results suggest that the formation of 2-OH-Ade in DNA will induce mutations in cells.

DNA cleavage induced by glycation of Cu,Zn-superoxide dismutase.

Human Cu,Zn-superoxide dismutase (Cu,Zn-SOD) undergoes site-specific and random fragmentation by non-enzymic glycosylation (glycation). Released Cu2+ from the glycated Cu,Zn-SOD probably facilitates a Fenton reaction to convert H2O2 into hydroxy radical, which then participates in the non-specific fragmentation [Ookawara et al. (1992) J. Biol. Chem. 267, 18505-18510]. In the present study, we investigated the effects of glycated Cu,Zn-SOD on cloned DNA fragments and nuclear DNA and analysed the formation of 8-hydroxydeoxyguanosine (8-OH-dG). Incubation of cloned DNA fragments with Cu,Zn-SOD and reducing sugars resulted in cleavage of the DNA. The extent of the cleavage corresponded to the reducing capacity of the sugar. Metal-chelating reagents, EDTA and bathocuproine, and an H2O2 scavenger, catalase, inhibited the DNA cleavage. Hydroxy radical scavengers and aminoguanidine, an inhibitor of glycation, also inhibited the reaction. Moreover, the glycation of Cu,Zn-SOD caused the substantial formation of 8-OH-dG in DNA. When isolated nuclei were incubated with CuCl2 plus H2O2, nuclear DNA cleavage was observed. Incubation of isolated nuclei with Cu,Zn-SOD that had been pre-incubated with glucose also resulted in nuclear DNA cleavage. These results suggest that hydroxy radical is produced through a Fenton reaction by Cu2+ and H2O2 released from the glycated Cu,Zn-SOD, and participates in nuclear DNA cleavage. This mechanism may partly explain the deterioration of organs under diabetic conditions.

Visualization of neural control of intracellular Ca2+ concentration in single vascular smooth muscle cells in situ.

The intermittent rise in intracellular Ca2+ concentration ([Ca2+]i oscillation) has been observed in many types of isolated cells, yet it has not been demonstrated whether it plays an essential role during nerve stimulation in situ. We used confocal microscopy to study Ca2+ transients in individual smooth muscle cells in situ within the wall of small arteries stimulated with perivascular sympathetic nerves or noradrenaline. We show here that the sympathetic adrenergic regulation of arterial smooth muscle cells involves the oscillation of [Ca2+]i that propagates within the cell in the form of a wave. Ca2+ release from intracellular stores plays a key role in the oscillation because it is blocked after the store depletion by ryanodine treatment. Ca2+ influx through the plasma membrane sustains the oscillation by replenishing the Ca2+ stores. These results demonstrate the involvement of [Ca2+]i oscillations in the neural regulation of effector cells within the integrated system.

Evidence for the accumulation of oxidative stress during cellular ageing of human diploid fibroblasts.

We have examined the level of 8-hydroxydeoxyguanosine (8-OH-dG) in the DNA of human diploid fibroblast TIG-1 cells at various stages of cellular ageing. Cellular DNA was obtained from cell lysates by simple precipitation with ethanol and analyzed 8-OH-dG levels by an electrochemical detection system. The data reveals that this method is valid for the detection of 8-OH-dG at levels of 1 x 10(-6) 8-OH-dG/dG without altering the recovery of other deoxynucleosides. Using this method, we demonstrate a significant increase in 8-OH-dG content during cellular ageing up to a 50 population doubling level (PDL) of TIG-1 cells, with levels of 0.5 x 10(-6) and 1.6 x 10(-6) 8-OH-dG/dG in young (22 PDL) and old (50 PDL) cells, respectively. The level decreases with further ageing and is 1.3 x 10(-6) 8-OH-dG/dG in senescent cells. We also show that the repair activity for the 8-OH-dG lesion declines significantly during cellular ageing, a phenomenon that might be associated with the increase in 8-OH-dG levels.

Quantal properties of H-type glutamatergic synaptic input to the striatal medium spiny neurons.

The striatal medium spiny neuron receives two distinct glutamatergic synaptic inputs, S-type and H-type inputs, which have very different amplitudes. We report here that peak amplitudes of unitary H-type excitatory postsynaptic currents (EPSCs) could be reduced by decreasing external Ca2+, unlike S-type EPSCs. This suggests that H-type EPSCs are multiquantal, and supports the idea that the cellular origin of H-type EPSCs is distinct from that of the S-type EPSCs.

Changes of aggressive behavior and brain serotonin turnover after very low-dose X-irradiation of mice.

Social isolation has been widely described to induce compulsive aggressive behavior and produce a large decrease of brain serotonin turnover in male mice. The aggressiveness by isolation in mice has been often used as a means for a better understanding of disturbed behavior in human beings. We found that male ICR white Swiss mice exhibiting isolation-induced aggression became gradually calm and showed remarkably quiet behavior 7 to 10 days after whole body irradiation of very low-dose X-rays (5-15 cGy). Higher doses (25-35 cGy), however, could not induce such effects. We also obtained the data on brain biochemistry giving a further support for the above low-dose effects on the mouse behavior. Brain serotonin turnover which has been known to be related to aggressive behavior in 5 or 15 cGy irradiated mice was faster than in aggressive control animals.
