Rat pleural mesothelial cells in culture.

A culture system has been developed for long-term maintenance of rat pleural mesothelial cells. Mesothelial cells were isolated from the parietal pleura of rats and cultured in NCTC 109 medium supplemented with 10% fetal bovine serum. The cell explants attached to the dish and formed a confluent monolayer of polygonal cells within 10 to 15 days. Subcultures were made in the same medium. The mean population doubling time was approximately 30 hr. The ultrastructure of the mesothelial cells in culture was studied by light and electron microscopy and was compared with that of cells obtained from sub-mesothelial components.

Reactivity of individual functional groups of histones in calf thymus chromatin.

Using the technique of competitive labelling (Kaplan, H., Stevenson, K.J. and Hartley, B.S. (1971) Biochem. J. 124, 289-299) with 1-fluoro-2,4-dinitro-[3,5-3H]benzene ([3H]Dnp-F), the chemical reactivity of the alanyl amino-terminus of H3, the prolyl amino-terminus of H2B and the sulphydryl groups of H3 within calf thymus chromatin were determined. Over pH values from 6 to 10, none of the groups had pH-reactivity profiles which fit titration curves, indicating that they are involved in specific interactions with other structural elements of chromatin. The alanyl amino-terminus is completely nonreactive over the entire pH range studied. The prolyl amino-terminus is reactive but its reactivity is considerably lower than expected for a secondary amino group. An unusual feature of this group is that its pH-reactivity profile has sharp discontinuities at pH values of 7.8 and 9. The average reactivity of the two sulphydryl groups of H3, unlike the two amino-termini, appears to approximate that expected of a free sulphydryl group.

Lower eyelid reconstruction by V-Y advancement cheek flap.

Another method of one-stage total reconstruction of the lower eyelid is reported. The usually discarded cheek triangle of the Mustardé technique is advanced superiorly, covering a chondromucosal composite graft used for lining and support.

How pleural mesothelial cells react in vitro with chrysotile fibres.

A culture system of rat pleural mesothelial cells has been developed for short-term experiments and long-term maintenance. In short-term experiments, endocytosis of chrysotile fibres (100 microgram/ml) has been demonstrated by electron microscopy of the pleura in situ. Structural modifications, mainly cytoplasmic vacuolization, were observed when the cells were cultured with sublethal doses of chrysotile (0.5, 1, 2 and 10 micrograms/ml). Untreated cells had a mean population doubling time of 37 hours. When the cells were treated with 2 or 10 micrograms/ml, the mean population doubling time was slightly increased.

Antihypertensive efficacy of propranolol given twice daily.

The therapeutic efficacy of propranolol in four and two daily doses was compared in 63 treated hypertensive patients in a multicentre trial. After 3 months of a stable diastolic blood pressure while receiving propranolol four times a day the patients were switched to a twice-a-day regimen, the drug being given at 8 am and 8 pm, with the same total daily dose, for 3 more months. Blood pressures and heart rates were measured at 8 am, 12 noon, 4 pm and 8 pm at 4-week intervals. There were no significant changes in mean blood pressure after the change to the twice-a-day regimen, although some patients reported new side effects. Compliance appeared to be unaffected.

Chlordiazepoxide and oxazepam disposition in cirrhosis.

When disease impairs clearance of drugs, multiple-dose therapy may result in cumulation. The disposition of chlordiazepoxide (CDX), 50 mg infused intravenously over 10 min, was studied in 14 normal subjects and in 11 patients with biopsy-proven cirrhosis. In the normal subjects, mean (+/- SE) kinetic parameters were: t 1/2 beta, 10.0 (+/- 0.9) hr; Vd, 0.38 (+/- 0.04) l/kg; clearance, 0.54 (+/- 0.13) ml/min/kg. Clearance of total drug correlated inversely with serum albumin concentration in normal subjects (r = -0.63). Values in cirrhotic patients were: t 1/2 beta, 34.9 (+/- 8.7) hr; Vd, 0.34 (+/- 0.024) 1/kg; and clearance, 0.185 (+/- 0.34) ml/min/kg. Desmethylchlordiazepoxide (DMCDX), the major metabolite of CDX, appeared in blood of cirrhotic patients less rapidly than in normal subjects. Severity of liver disease did not indicate the impairment of CDX clearance. In 5 of the same cirrhotic patients, mean t 1/2 beta for oxazepam (7.1 +/- 1.0 hr) was 27% longer than in control subjects (5.6 +/- 0.7 hr); the difference is not significant. On kinetic grounds oxazepam may be preferable to chlordiazepoxide in cirrhotic patients since its elimination kinetics are not greatly altered in cirrhosis.

Effects of acetone, methyl ethyl ketone and methyl isobutyl ketone on a match-to-sample task in the baboon.

Acetone, methyl ethyl ketone (MEK) and methyl isobutyl ketone (MIBK) were evaluated for effects on a delayed match-to-sample discrimination task in the juvenile baboon. The animals were exposed to 1/2 the threshold limit value (TLV) of each gas for 24 hr per day during a 7-day period. They were also exposed to a combination of MEK and MIBK at the same exposure concentrations. Each exposure condition affected accuracy of performance minimally but resulted in increased and decreased extra responses during the delay intervals. Response times were slowed under acetone, MEK or MIBK. In contrast to the effects of the individual gases, exposure to a combination of the same doses of MEK and MIBK produced a consistent increase in extra responses during delay and a concomitant decrease in response times. Changes in tissue uptake and metabolism are suggested as possible mechanisms to explain this observation.
